Iron deficiency responses were investigated in roots of soybean, a Strategy I plant species. Soybean responds to iron deficiency by decreasing growth, both at the root and shoot level. Chlorotic symptoms in younger leaves were evident after a few days of iron deficiency, with chlorophyll content being dramatically decreased. Moreover, several important differences were found as compared with other species belonging to the same Strategy I. The main differences are (i) a lower capacity to acidify the hydroponic culture medium, that was also reflected by a lower H(+)-ATPase activity as determined in a plasma membrane-enriched fraction isolated from the roots; (ii) a drastically reduced activity of the phosphoenolpyruvate carboxylase enzyme; (iii) a decrease in both cytosolic and vacuolar pHs; (iv) an increase in the vacuolar phosphate concentration, and (v) an increased exudation of organic carbon, particularly citrate, phenolics, and amino acids. Apparently, in soybean roots, some of the responses to iron deficiency, such as the acidification of the rhizosphere and other related processes, do not occur or occur only at a lower degree. These results suggest that the biochemical mechanisms induced by this nutritional disorder are differently regulated in this plant. A possible role of inorganic phosphate in the balance of intracellular pHs is also discussed.  相似文献

Multivariate analysis was used for interpreting data from a pot experiment using samples of three Spanish soils. Samples of soil fertilized with compost were compared with untreated control samples. We also compared the effect of adding the compost to soil with a controlled moisture content of 50% of its water holding capacity (WHC), and to a near-saturated soil (95% WHC). Hierarchical cluster analysis (HCA) and principal component analysis (PCA) were used; they perfectly differentiated sample groups both as a function of the treatment applied and by sampling date. The compost samples were characterized by higher pH, electrical conductivity (EC), organic matter (OM) content and cation exchange capacity (CEC), together with nutrient concentrations than the control pots. The pots with a soil–compost mixture at 95% WHC presented lower values of EC, CEC, inorganic N, K, Na and B than the mixtures at 50% WHC. Multivariate methods may therefore be useful for the analysis and interpretation of a large number of data in soil research.  相似文献

2',7'-Dichlorodihydrofluorescein (DCFH2) is one of the most widely used probes for detecting intracellular oxidative stress, but requires a catalyst to be oxidized by hydrogen peroxide or superoxide and reacts nonspecifically with oxidizing radicals. Thiyl radicals are produced when many radicals are "repaired" by thiols, but are oxidizing agents and thus potentially capable of oxidizing DCFH2. The aim of this study was to investigate the reactivity of thiol-derived radicals toward DCFH2 and its oxidized, fluorescent form 2',7'-dichlorofluorescein (DCF). Thiyl radicals derived from oxidation of glutathione (GSH) or cysteine (CysSH) oxidized DCFH2 with rate constants at pH 7.4 of approximately 4 or approximately 2x10(7) M(-1) s(-1), respectively. Both the rates of oxidation and the yields of DCF were pH-dependent. Glutathione-derived radicals interacted with DCF, resulting in the formation of DCFH* absorbing at 390 nm and loss of fluorescence; in contrast, cysteine-derived radicals did not cause any depletion of DCF fluorescence. We postulate that the observed apparent difference in reactivity between GS* and CysS* toward DCF is related to the formation of carbon-centered, reducing radicals from base-catalyzed isomerization of GS*. DCF formation from interaction of DCFH2 with GS* was inhibited by oxygen in a concentration-dependent manner over the physiological range. These data indicate that in applying DCFH2 to measure oxidizing radicals in biological systems, we have to consider not only the initial competition between thiols and DCFH2 for the oxidizing radicals, but also subsequent reactions of thiol-derived radicals, together with variables--including pH and oxygen concentration--which control thiyl radical chemistry.  相似文献

Volume changes of cardiac tissue under hyperosmotic stress in Rana catesbeiana were characterized by the identification of the osmolytes involved and the possible regulatory processes activated by both abrupt and gradual changes in media osmolality (from 220 to 280mosmol/kg H(2)O). Slices of R. catesbeiana cardiac tissue were subjected to hyperosmotic shock, and total tissue Na(+), K(+), Cl(-) and ninhydrin-positive substances were measured. Volume changes were also induced in the presence of transport inhibitors to identify osmolyte pathways. The results show a maximum volume loss to 90.86+/-0.73% of the original volume (measured as 9% decrease in wet weight) during abrupt hyperosmotic shock. However, during a gradual osmotic challenge the volume was never significantly different from that of the control. During both types of hyperosmotic shock, we observed an increase in Na(+) but no significant change in Cl(-) contents. Additionally, we found no change in ninhydrin-positive substances during any osmotic challenge. Pharmacological analyses suggest the involvement of the Na(+)/H(+) exchanger, and perhaps the HCO(3)(-)/Cl(-) exchanger. There is indirect evidence for decrease in Na(+)/K(+)-ATPase activity. The Na(+) fluxes seem to result from Mg(2+) signaling, as saline rich in Mg(2+) enhances the regulatory volume increase, followed by a higher intracellular Na(+) content. The volume maintenance mechanisms activated during the gradual osmotic change are similar to that activated by abrupt osmotic shock.  相似文献

Galectins are a large family of structurally related beta-galactoside-binding proteins that play a pivotal role in the control of cell differentiation, proliferation, activation and apoptosis of many different cell types including immune cells. By crosslinking specific glycoconjugates, different members of the galectin family behave as pro-inflammatory or anti-inflammatory "cytokine-like" mediators, acting at different levels of innate and adaptive immune responses. Here we will review recent advances on the role of galectins in key events of the immune and inflammatory response, such as tolerance induction, cell cycle progression, cell adhesion, chemotaxis, antigen presentation and apoptosis. In particular we will examine the influence of individual members of the galectin family in the physiology of different immune cell types involved in innate and adaptive immune responses. Moreover, we will discuss the importance of these sugar-binding proteins as therapeutic targets in Th1- and Th2-mediated immune disorders, an exciting area for future research.  相似文献

Vasodilator effects of peptides derived from egg white proteins   总被引:1,自引:0,他引:1  
The aim of this work was to investigate the effect of several peptides, identified before and after simulated gastrointestinal digestion of an egg white hydrolysate, on the vascular function, in rat aorta. The sequences IVF, RADHPFL and YAEERYPIL (0.1 mM) induced vasodilatation in intact aortic rings, with the maximum percentage of dilation corresponding to RADHPFL (40.5 ± 7.0%). Two of the end products of the gastrointestinal digestion, RADHP and YPI, also showed vasodilator activity with degrees of relaxation higher than 50%. However, all these peptides failed to induce relaxation in endothelium-denuded aortic rings. The relaxation induced by RADHP was concentration-dependent and it was partially blocked by the nitric oxide synthase inhibitor l-NAME (100 μM) and by the B1 bradykinin receptor antagonist Des-HOE 140 (30 nM), thus showing that it was mediated by NO production through the activation of B1 bradykinin receptors. These findings suggest that these peptides could reduce the vascular resistance and could be used as functional food ingredients in the prevention and treatment of hypertension.  相似文献

Despite the extensive literature on the effect on soil properties of afforestation of former arable land, we still lack full understanding of whether the changes proceed in the same direction and at the same rate, and of how long is required to achieve a state of soil equilibrium typical of a natural forest ecosystem. Therefore, as part of a study comparing post-arable sandy soils (Dystric Arenosols) afforested with Scots pine (Pinus silvestris L.) with arable soils and soils of continuous coniferous forests, the range and direction of changes in pH, organic carbon (Corg), total nitrogen (Ntot), ammonium (N-NH4) and nitrates (N-NO3) in soil solution, total (Ptot) and available (Pav) phosphorus were determined. The studies were carried out in south-east Poland (51°30′-51°37′N, 22°20′-22°35′E). Ten paired sites of afforested soils (five with 14- to 17-year-old stands and five with 32- to 36-year-old stands) with adjacent cultivated fields, and five sites of continuous forest with present stands of ca. 130–150 years old were selected. Soil samples were taken from the whole thickness of master horizons and, in the case of the A horizon of the afforested soils, from three layers: 0–5 (A0–5), 5–10 (A5–10) and 10–20 cm (A10–20). The cultivated soils in the Ap horizon showed higher pH (by ca. 1.0 unit), lower Corg and C:N, similar Ntot, lower N-NH4, higher N-NO3, higher Ptot and Pav contents compared with the Ah horizon of continuous forest soils. The results indicated decreased soil pH in the former plough layer of the afforested soils, with the greatest decrease observed in the 0–5 cm layer. In these soils, the Corg content was considerably higher in the A0–5 layer, but lower in the two deeper layers and in the whole A horizon (0–20 cm) compared with the Ap horizon of the arable soils. The results indicate that the Corg content, after an initial phase of decline, again achieved a level characteristic of arable soils. The Ntot content in all layers of the A horizon of the afforested soils was lower than in the Ap horizon of the arable soils, and showed a reduction with stand age, especially in deeper layers. The C:N ratios in the mineral topsoil increased with stand age. N-NH4 content increased and N-NO3 decreased after afforestation. The Ptot and Pav contents in all layers and in the whole A horizon of the afforested soils, on stands of both ages, was lower than in the Ap of the cultivated soils. From the results, it could be concluded that, after more than 30 years of tree growth, the soils of the A horizon were still more similar to arable than to continuous forest soils with respect to Corg, Ptot and Pav. With respect to pH, N-NH4 and N-NO3, especially in the 0–5 cm layer, they were more similar to continuous forest soils than to cultivated soils, but with respect to Ntot and C:N ratio they were somewhere in between.  相似文献
