The extra-thecal epithelia of cryofixed undecalcified, freeze-substituted polyps of the scleractinian corals Galaxea fascicularis and Tubastrea faulkneri and axial and basal polyps of Acropora formosa have been examined, in anhydrously prepared thick slices, by confocal laser scanning light microscopy. The avoidance of chemical fixation and decalcification makes it possible to determine whether previously seen structures are real or artefactual products of swelling, shrinkage and distortion. All of the epithelia of all the corals examined are characterised by well defined intercellular spaces. Mucocytes are present in all cell layers in Galaxea and Tubastrea but are not present in any cell layers in the axial polyp of Acropora although they are abundant in the oral ectoderm of the basal polyps in this coral. Zooxanthellae are absent in Tubastrea, the epithelia of the exert septa of Galaxea and the axial polyp of Acropora. The calicoblastic ectoderm is generally composed of thin squamous cells with large intercellular spaces. At rapidly calcifying regions such as the tips of the exert septa of Galaxea, the calicoblastic cells are elongated with extensive arborisation of the basal regions of the cells. They are separated by large intercellular spaces and contain numerous fluorescent granules. The apical regions of these cells appear to be closely applied to the surface of the skeleton. There is no evidence of a space between the apical region of the calicoblastic cells and the skeleton.  相似文献

1.  We examined the retinas of 2 species of stomatopods in the superfamily Squilloidea, Cloridopsis dubia and Squilla empusa, and 2 species of the super-family Lysiosquilloidea, Coronis scolopendra and Lysiosquilla sulcata, using microspectrophotometry in the visible region of the spectrum.
2.  Retinas of all species included numerous photostable pigments, such as green reflecting pigment, hemocyanin, colored oil droplets, and vesicles. Both lysiosquilloid species also had intrarhabdomal filters within specialized photoreceptors of the midband.
3.  Squilloid species contained a single visual pigment throughout all photoreceptors, with peak absorption at medium wavelengths (near 515nm). Retinas of lysiosquilloids contained a diversity of visual pigments, with estimated max values ranging from 397 to 551 nm.
4.  Spectral sensitivity functions were estimated for the lysiosquilloid species based on estimates of visual pigment nax, photoreceptor dimensions, and specific absorbances of the visual pigments and intrarhabdomal filters. Ommatidia of midband Rows 1 to 4 contained pairs of narrowly tuned spectral receptors, appropriate for spectral discrimination, while ommatidia of midband Rows 5 and 6, and all peripheral ommatidia, had broad spectral sensitivity functions.
5.  Lysiosquilloid stomatopods have retinas that closely resemble those of gonodactyloids both structurally and in their visual pigment diversity. In contrast, squilloids have retinas that are much simpler. These differences appear to be related to the habitats and activity cycles of species belonging to the 3 major superfamilies of stomatopod crustaceans.

Summary Limnothrissa miodon is a clupeid from Lake Tanganyika which has been introduced to Lakes Kivu and Kariba and which invaded Lake Cahora Bassa. These lakes differ considerably from one another but the biology of Limnothrissa is similar in many respects in all of them. Similarities include its feeding and breeding biology, whilst its populations fluctuate, on both an annual and a seasonal basis, in relation to environmental changes. The major differences between the lakes concern the size to which Limnothrissa grows. Their average length in the two natural lakes, Lakes Tanganyika and Kivu, is about twice the average length in the two artificial lakes, Lakes Kariba and Cahora Bassa. This in turn affects their age of maturity and they breed during their first year in the reservoirs but during their second in the natural lakes. Their growth rates and performance are comparable up to about 6 months of age and the difference in their size seems to be due to high mortality in the artificial lakes where few Limnothrissa survive for more than a year. Possible explanations for their small size include the genetic composition of the original introduction and the unpredictability of the environment. Neither of these explanations is supported by evidence at present. Deficiencies in the available food and the effects of intense predation are probably the most likely causes. In both natural lakes, Limnothrissa are predatory or cannibalistic from 100 mm in length and 12 months in age, and fish in their diet may enable them to survive into their second year. The fish in Lakes Kariba and Cahora Bassa, in contrast, are neither predatory nor cannibalistic to any degree. There are no predatory fish species in Lake Kivu but there is a diverse community of pelagic piscivores in Lake Tanganyika which tends to contradict the predation hypothesis. However, the position of Limnothrissa in the reservoirs probably resembles that occupied in Lake Tanganyika by Stolothrissa which is also a small, annual species. The significance of this phenomenon is that Limnothrissa can maintain a high biomass and productivity in the face of intense predation. This trait may be of importance to fisheries management because it means that their yield can be greatly increased.  相似文献

Anthropometric and sociodemographic variables were taken from 4320 children in a baseline survey carried out in March-April 1988 in the district of Mbarara, south-west Uganda. After 12 months a follow-up survey assessed the mortality of the children during the preceding year. Lack of ownership of cattle, recent arrival in the village, using candles for lighting, being of birth order higher than 5 and having a father with less than 8 years of schooling were significantly associated with child mortality. The addition of mid-upper arm circumference significantly improved the logistic model of socioeconomic variables and mortality and did not diminish the predictive power of socioeconomic variables in relation to increased mortality. This suggests that nutritional status and specific socioeconomic factors are both, independently, important predictors of child mortality.  相似文献

Aeromonas salmonicida is a pathogenic aquatic bacterium and the causal agent of furunculosis in salmon. In the course of this study, it was found that when grown in vitro on tryptic soy agar, A. salmonicida strain 80204-1 produced a capsular polysaccharide with the identical structure to that of the lipopolysaccharide O-chain polysaccharide. A combination of 1D and 2D NMR methods, including a series of 1D analogues of 3D experiments, together with capillary electrophoresis-electrospray MS (CE-ES-MS), compositional and methylation analyses and specific modifications was used to determine the structure of these polysaccharides. Both polymers were shown to be composed of linear trisaccharide repeating units consisting of 2-acetamido-2-deoxy-D-galacturonic acid (GalNAcA), 3-[(N-acetyl-L-alanyl)amido]-3,6-dideoxy-D-glucose[3-[(N-acetyl-L-alanyl)amido]-3-deoxy-D-quinovose, Qui3NAlaNAc] and 2-acetamido-2,6-dideoxy-D-glucose (2-acetamido-2-deoxy-D-quinovose, QuiNAc) and having the following structure: [-->3)-alpha-D-GalpNAcA-(1-->3)-beta-D-QuipNAc-(1-->4)-beta-D-Quip3NAlaNAc-(1-]n, where GalNAcA is partly presented as an amide and AlaNAc represents N-acetyl-L-alanyl group. CE-ES-MS analysis of CPS and O-chain polysaccharide confirmed that 40% of GalNAcA was present in the amide form. Direct CE-ES-MS/MS analysis of in vivo cultured cells confirmed the formation of a novel polysaccharide, a structure also formed in vitro, which was previously undetectable in bacterial cells grown within implants in fish, and in which GalNAcA was fully amidated.  相似文献

The physiological organisation of plants is considered in relation to the carbon economy of plant parts. Although assimilate is partitioned according to the relative strength of sinks, in many species there is also a very close relationship between partitioning and shoot phyllotaxy, giving rise to sectorial patterns of allocation whereby only certain sinks are supported by any source leaf. Essentially these sinks are in the same orthostichy as the source leaf. This constraint of the vascular architecture on assimilate distribution to developing sinks such as leaves, flowers and fruits is not always absolute, as following the loss of their principal source leaves these sinks can in many cases be supplied with assimilate by other leaves via new inter-orthostichy pathways. The supply of assimilate to major sinks such as developing fruits becomes more and more localised with time so that a fruit in an axillary position becomes largely supported by its subtending leaf; the reproductive node—a metamer-can thus be regarded as a relatively autonomous unit of the plant (an IPU). Similary, once established after a developmental phase of assimilate import, tiller ramets and branches in unitary plants tend to become physiologically autonomous modules. However, the functional autonomy of tillers is reversed following defoliation or shading as they are then sustained by the import of assimilate, subject to its availability, from unaffected tillers. Consequently the plant becomes physiologically integrated by the flow of assimilate from one part to another. The mainly autonomous ramets of many stoloniferous and rhizomatous species display a similar pattern of physiological integration in response to source manipulation, but in some species the ramets appear to maintain their independent functioning as a normal feature of the carbon allocation within the clone. In other clonal species, as the clone develops and becomes more structurally complex, vascular constraints start to restrict the movement of resources, and the clone becomes composed of a number of semi-autonomous IPUs. In unitary plants branches appear to remain very physiologically isolated in terms of their carbon economy once they become established, irrespective of a range of source-sink manipulations.These different patterns of physiological integration and organisation are discussed in relation to different strategies of assimilate utilisation and conservation.  相似文献

Specific interactions of chromatin with the nuclear envelope (NE) in early embryos of Drosophila melanogaster have been mapped and analyzed. Using fluorescence in situ hybridization, the three-dimensional positions of 42 DNA probes, primarily to chromosome 2L, have been mapped in nuclei of intact Drosophila embryos, revealing five euchromatic and two heterochromatic regions associated with the NE. These results predict that there are approximately 15 NE contacts per chromosome arm, which delimit large chromatin loops of approximately 1-2 Mb. These NE association sites do not strictly correlate with scaffold-attachment regions, heterochromatin, or binding sites of known chromatin proteins. Pairs of neighboring probes surrounding one NE association site were used to delimit the NE association site more precisely, suggesting that peripheral localization of a large stretch of chromatin is likely to result from NE association at a single discrete site. These NE interactions are not established until after telophase, by which time the nuclear envelope has reassembled around the chromosomes, and they are thus unlikely to be involved in binding of NE vesicles to chromosomes following mitosis. Analysis of positions of these probes also reveals that the interphase nucleus is strongly polarized in a Rabl configuration which, together with specific targeting to the NE or to the nuclear interior, results in each locus occupying a highly determined position within the nucleus.  相似文献

A phenol sulfotransferase from rat liver (EC 2.8.2.9), expressed inEscherichia colifrom a single cDNA, was purified as two separable but catalytically active proteins. The proteins appeared to be identical to each other and to the natural liver sulfotransferase by comparison of their amino acid constitution, amino-terminal end group, and interaction with a polyclonal antibody raised against the liver enzyme. Each of the recombinant forms, α and β, catalyzed the sulfuryl group transfer from 4-nitrophenylsulfate to an acceptor phenol, a reaction in which 3′-phospho-adenosine 5′-phosphate (PAP) is a necessary intermediate. Only form β, however, catalyzes the physiological transfer of a sulfuryl group from 3′-phosphoadenosine 5′-phosphosulfate (PAPS) to the free phenol. Evidence is presented that sulfotransferase α, but not β, has 1 mol of PAP tightly bound per enzyme dimer. The ability to utilize PAPS as a sulfate donor could be altered: form α could be treated and purified as form β to acquire the ability to use PAPS, whereas form β was treated by extended incubation with PAP, lost its ability to use PAPS, and was purified as form α.  相似文献
