X-ray phase-contrast computed tomography is an emerging imaging technology with powerful capabilities for three-dimensional (3D) visualization of weakly absorbing objects such as biological soft tissues. This technique is an extension of existing X-ray applications because conventional attenuation-contrast images are simultaneously acquired. The complementary information provided by both the contrast modalities suggests that enhanced material characterization is possible when performing combined data analysis. In this study, we describe how protein, lipid, and water concentrations in each 3D voxel can be quantified by vector decomposition. Experimental results of dairy products, porcine fat and rind, and different human soft tissue types are presented. The results demonstrate the potential of phase-contrast imaging as a new analysis tool. The 3D representations of protein, lipid, and water contents open up new opportunities in the fields of biology, medicine, and food science.  相似文献

For in vivo demonstration of acid phosphatase activity in the rhizosphere of soil-grown plants filter papers were treated with a mixture of 1-naphthyl phosphate as substrate and the diazonium salt Fast Red TR as an indicator. After enzymatic hydrolysis, 1-naphthol forms a red complex with Fast Red TR. This method was applied to 8-day old maize plants and 3-year old Norway spruce plants growing in rhizoboxes in soil under non-sterile conditions. The treated filter paper is placed at the surface of roots and soil and acid phosphatase activity is visualized as a red-coloured root print on the filter paper. The method can be used as a qualitative analysis of acid phosphatase in the rhizosphere. It also allows a rough estimate of phosphatase activity in different root zones.  相似文献

In vitro nicotianamine synthase activity was measured in tobaccounder Fe-deficient or Fe-sufficient conditions. Its activitywas not induced by Fe-deficiency, in contrast to barley roots,implying that the molecular biological regulation of nicotianaminesynthase in response to Fe-deficiency may be different betweentobacco and barley. Linalool is a monoterpene compound reported to be a major component of essential oils in, various aromatic species. Several Linalool-producing species are used in traditional medical systems, includingAeolanthus suaveolens G. Dom (Labiatae) used as anticonvulsant in the Brazilian Amazon. Psychopharmacological in vivo evaluation of Linalool showed that this compound have dose-dependent marked sedative effects at the Central Nervous System, including hypnotic, anticonvulsant and hypothermic properties. The present study reports an inhibitory effect of Linalool on Glutamate binding in rat cortex. It is suggested that this neurochemical effect might be underlining Linalool psychopharmacological effects. These findings provide a rational basis for many of the traditional medical use of Linalool producing plant species.  相似文献

Abstract: A simple and sensitive detection system, using polymerase chain reaction (PCR) and a soil microcosm, was developed to detect a bacterial catabolic gene in the rhizosphere. The inoculated population of Alcaligenes eutrophus JMP134, a phenol and 2,4-dichlorophenoxy acetic acid utilizer, was readily detected by this technique, which permitted taking of samples from specific locations of root (including rhizosphere) and soil. The number of JMP134 viable cells (102–103 cells), typically picked up by the nitrocellulose filter strip method, yielded sufficient amount of the target DNA to be detected by PCR. Primers encoding metapyrocatechase I (MPC I; catechol 2,3-dioxygenase) enabled the discrimination of at least five viable cells of JMP134 among the indigenous microorganisms inhabiting bush bean roots. This simplified PCR detection procedure facilitated monitoring of the specific degradative gene in the rhizosphere in only 5 h.  相似文献

Roots of grasses in response to iron deficiency markedly increase the release of chelating substances (`phytosiderophores') which are highly effective in solubilization of sparingly soluble inorganic FeIII compounds by formation of FeIIIphytosiderophores. In barley (Hordeum vulgare L.), the rate of iron uptake from FeIIIphytosiderophores is 100 to 1000 times faster than the rate from synthetic Fe chelates (e.g. Fe ethylenediaminetetraacetate) or microbial Fe siderophores (e.g. ferrichrome). Reduction of FeIII is not involved in the preferential iron uptake from FeIIIphytosiderophores by barley. This is indicated by experiments with varied pH, addition of bicarbonate or of a strong chelator for FeII (e.g. batho-phenanthrolinedisulfonate). The results indicate the existence of a specific uptake system for FeIIIphytosiderophores in roots of barley and all other graminaceous species. In contrast to grasses, cucumber plants (Cucumis sativus L.) take up iron from FeIIIphytosiderophores at rates similar to those from synthetic Fe chelates. Furthermore, under Fe deficiency in cucumber, increased rates of uptake of FeIIIphytosiderophores are based on the same mechanism as for synthetic Fe chelates, namely enhanced FeIII reduction and chelate splitting. Two strategies are evident from the experiments for the acquisition of iron by plants under iron deficiency. Strategy I (in most nongraminaceous species) is characterized by an inducible plasma membrane-bound reductase and enhancement of H+ release. Strategy II (in grasses) is characterized by enhanced release of phytosiderophores and by a highly specific uptake system for FeIIIphytosiderophores. Strategy II seems to have several ecological advantages over Strategy I such as solubilization of sparingly soluble inorganic FeIII compounds in the rhizosphere, and less inhibition by high pH. The principal differences in the two strategies have to be taken into account in screening methods for resistance to `lime chlorosis'.  相似文献

This paper focuses on the phylogenetic relationships of eight North American caenophidian snake species (Carphophis amoena, Contia tenuis, Diadophis punctatus, Farancia abacura, Farancia erytrogramma, Heterodon nasicus, Heterodon platyrhinos, and Heterodon simus) whose phylogenetic relationships remain controversial. Past studies have referred to these "relict" North American snakes either as colubrid, or as Neotropical dipsadids and/or xenodontids. Based on mitochondrial DNA ribosomal gene sequences and a likelihood-based Bayesian analysis, our study suggests that these North American snakes are not monophyletic and are nested within a group (Dipsadoidea) that contains the Dipsadidae, Xenodontidae, and Natricidae. In addition, we use the relationships proposed here to highlight putative examples of parallel evolution of hemipenial morphology among snake clades.  相似文献

We have developed stereocontrolled transesterification for the synthesis of chiral phosphite mester backbone under basic conditions. Substrate phosphite triesters with phenoxy derivative substituents as leaving groups were separated into individual stereoisomers and reacted with hydroxyl-containing compounds to form desired backbone.

The iron (Fe) efficient maize cultivar WF9 and the Fe inefficient maize mutant ys1 were grown in nutrient solutions with varied Fe supply. Changes in pH, reducing capacity of the roots and the release of Fe(III) reducing compounds were monitored over a period of 11 days. In both cultivars under Fe deficiency, there was no increased release of protons or reducing compounds and no increased reducing capacity of the roots. Indeed, the Fe(III) reduction of the roots of both cultivars tended to be higher in Fe sufficient plants. In contrast to some recent reports, these results demonstrate that the Fe efficient maize cultivar WF9 does not respond to Fe deficiency by strategy I mechanisms. This suggests that differences in Fe efficiency between the two cultivars are probably due to use of strategy II mechanisms for Fe acquisition.  相似文献

The dependence of acropetal Ca2+ transport on polar basipetal indoleacetic acid (IAA) transport was investigated in excised tomato fruits ( Lycopersicon esculentum L. Mill.) using an in vitro fruit system. Auxin transport inhibitors like triiodobenzoic acid (TIBA), chlorofluorenolmethyl ester (CME) and naphthylphthalamic acid (NPA) were used in order to investigate the effect of restricted polar basipetal auxin transport on the acropetal transport of 45Ca2+, 86Rb+ and 98Sr2+ into the same fruits. TIBA and CME inhibited basipetal transport of IAA. particularly in 10- to 12-day-old tomato fruits, and simultaneously restricted the acropetal transport of 45Ca2+. The auxin transport inhibitors failed to significantly reduce the upward transport of 86Rb+ and the transport of 96Sr2+ was less inhibited than that of 45Ca2+. TIBA and CME did not significantly affect the acropetal transport of labelled water into the fruit, nor the cation-exchange capacity or K+ and Mg2+ concentrations in the tomato fruit. These results support the view that a part of the Ca2+-specific acropetal transport into tomato fruits is associated with the polar basipetal IAA transport. This Ca2+ transport is independent of the transpiration stream into the fruit and the cation exchange capacity of the fruit tissue.  相似文献
