Summary Chronic myelogenous leukemia (CML) patients in chronic phase display compromised lymphokine-activated killer (LAK) cell induction, which is partly restored after therapy with interferon . However, the relative resistance of the leukemic cells from these patients to autologous or allogeneic LAK lysis is not affected by this treatment. In an attempt to render CML cells more susceptible to lysis or cytostasis, they were precultured in serum-free medium with or without recombinant growth factors. In eight patients studied, interleukin-3 (IL-3) significantly enhanced the spontaneous short-term (6-day) proliferation of CML cells, with retention of ability to form colonies in methylcellulose. Culture in either medium alone or IL-3 led to a significant enrichment of CD14+ and CD33+ cells but to a reduction in CD34+ cells. In contrast, culture of the same cells in IL-2 (to generate autologous LAK activity) resulted in a loss of CD14+ and CD33+ as well as CD34+ cells but in a significant increase in CD3+ and CD56+ cells. Despite similarities in their phenotypes, IL-3 cultured cells but not those cultured in medium alone acquired susceptibility to lysis by the IL-2-cultured autologous LAK cells. First, the literature of the last two decades on nutritional effects of host plants on spider mites is briefly reviewed. Second, experiments are described that subjected micro-propagated apple trees to four different levels of each macronutrient N, P and K. Spider mites (Tetranychus urticae Koch) feeding on leaf disks of these plants were checked for their developmental time, egg production and longevity. Plant analysis revealed that the concentration of N, P and K corresponded to the respective treatments. The content of phenolic compounds in the leaves increased with N and P deficiency. In the N experiment, spider-mite preimaginal developmental rate and oviposition rate were both positively correlated with leaf N. Often, fecundity was positively correlated with N and carbohydrate content of the leaves, and negatively with the phenolic content. Longevity of the two-spotted spider mite was not significantly affected by any treatment. The K experiments yielded only minor differences in plant contents as well as in spider-mite biology.From these mite data, file-tables were constructed and statistically analyzed by the Jackknife technique. The life-table analysis showed a gradual decline in the intrinsic rate of natural increase (r m) with N and P deficiency. With all experiments pooled,r m was clearly correlated to leaf N and particularly to the content of phenolic compounds in the leaves. Nitrogen shortage had the most distinct influence on mite population growth: in a range of 1.5–3.0% leaf N,r m increased by a factor of 4, the number of multiplications per generation (R 0) by 11, and the doubling time of the population was prolonged 4-fold on severely N deficient leaves.  相似文献

Summary Protoplast formation in Curvularia inaequalis was achieved using non-commercial and commercial snail gut enzymes or Trichoderma harzianum enzymes. The cells were grown for enzyme treatment on cellophane sheets or in liquid cultures for varying periods of time. The production of T. harzianum enzymes is discussed. The highest protoplast yields were 2.6x107 protoplasts/ml enzyme solution. Protoplasts were shown to have zero to four nuclei. Protoplast regeneration was succesfully carried out in semisolid agar.  相似文献

Early developmental stages of theAnemonia germ are characterized by asynchronously dividing nuclei and an extreme delay of blastomere differentiation. The nuclei migrate to the periphery, whereas nutritive substances remain in the interior. Following this stage, the appearance of cell boundaries results in the formation of the blastoderm and the simultaneous division of the yolk into many fragments. Most of them are exclusively filled with reserve material; only very few contain zooxanthellae or nuclei. On the embryo's surface, conically shaped bundles of long microvilli are obvious. They appear to be less regularly arranged than the spines of oocytes before insemination. Pigment granules that have originated from fusing Golgi vesicles are crowded peripherally in the blastoderm cells. In the nucleoplasm single annulate lamellae that seem to be cut off from the nuclear envelope can frequently be found. There is no further cellular differentiation until gastrulation is completed. Though yolk-containing cellular fragments occupy nearly the whole blastocoel, entoderm formation occurs by invagination. Ultrastructural observations provide evidence of the existence of interstices between entoderm cells that allow all nutritive substances to pass gradually into the gastric cavity. In the region of the blastoporus there are cellular processes enveloping reserve material. Presumably, these observations indicate a so-called filtration of nutritive yolk (Korschelt & Heider, 1909) that might represent an additional mode for the transfer of yolk-containing cellular fragments from blastocoel into gastrocoel.  相似文献

"Viili," a fermented milk product, has a firm but viscous consistency. It is produced with traditional mesophilic mixed-strain starters, which have various stabilities in dairy practice. Thirteen morphologically different types of phages were found in 90 viili samples studied by electron microscopy. Ten of the phage types had isometric heads with long, noncontractile tails, two had elongated heads with long, noncontractile tails, and one had a unique, very long elongated head with a short tail. Further morphological differences were found in the tail size and in the presence or absence of a collar, a baseplate, and a tail fiber. To find hosts for the industrially significant phages, we examined the sensitivities of 500 bacterial isolates from starters of the viili. Seven of the phages attacked Streptococcus cremoris strains, three attacked S. lactis subsp. diacetylactis strains, and four attacked Leuconostoc cremoris strains. Some phages differed only in their host specificity. Hosts were not found for 4 of the 13 morphological types of phages.  相似文献

The seasonal variation in water chemistry was followed during 1980 and 1981 in four mountain brooks in the southern part of Swedish Lapland. In the area investigated the soil is calcareous and the brook water is very well buffered during the major part of the year, with alkalinity varying between 0.4 and 1.0 milliequivalents per liter and with pH values about 7.5. These years the snow had a pH of approximately 5.2, which was considerably higher than has been reported from adjacent areas in the lower, coniferous region. During snowmelt the water discharge increased drastically, and although the net transport of bicarbonate increased, alkalinity showed low values due to dilution with meltwater. pH decreased, but not further than to 6.3–6.5, far from values reported in 1979 (pH less than 5), apparently due to the comparatively clean snow. A slight deficit in alkalinity, as compared to the nonmarine calcium and magnesium content, points to an acidification impact on the area. During maximum runoff some chemical variables, like aluminium, iron, nitrogen and phosphorus, behaved reversely to what might be expected during dilution and reached maxima in concentrations. It is concluded that the extreme runoff characteristics of high mountain areas make brook water more sensitive to acid precipitation than might be expected when regarding only the calcareous properties of soil and bedrock.  相似文献
