The efficacy of oxamyl in controlling Heterodera schachtii on cabbage was determined by applying various contbinations of soil drenches at 6.7 kg (a.i.)/ha and foliar sprays at 0.04 kg (a.i.)/100 liters of water to cabbage seedlings. Pretransplant drenches provided some control of H. schachtii over a 13-week period. A single foliar spray of oxamyl 1 week before transplanting apparently prevented penetration of H. schachtii larvae; post-transplant sprays were relatively ineffective. A pretransplant or transplant drench combined with a foliar application 2 weeks after transplanting provided the most effective control. The effectiveness of drenches plus post-transplant sprays is probably due to the spray augmenting the action of the drench in inhibiting the development of larvae after penelration. 相似文献

- Most of the world's large rivers are dammed for the purposes of water storage, flood control, and power production. Damming rivers fundamentally alters water temperature and flows in tailwater ecosystems, which in turn affects the presence and abundance of downstream biota.
- We collaborated with more than 200 citizen scientists to collect 2,194 light trap samples across 2 years and more than 2,000 river km. Samples contained 16,222 net‐spinning caddisfly (Hydropsyche) individuals across six species. We used these data to model the distribution of Hydropsyche throughout the Colorado River Basin in the western U.S.A. to identify the roles of water temperature, flows, and species‐specific morphology in determining aquatic species distributions throughout a large arid watershed that has been heavily altered by damming.
- We predicted that water temperatures would determine Hydropsyche presence and abundance to a greater extent than diel variation in river stage associated with hydropower production. Among many species, adult female Hydropsychids are morphologically adapted to swim to deep‐water oviposition sites. We predicted that the presence of this ability would negate the otherwise deleterious effects of high stage change on caddisfly egg mortality.
- We found that distributions of the two most widespread species, Hydropsyche occidentalis and Hydropsyche oslari (92% of total Hydropsyche captured), were both predicted by water temperatures. However, we also found that the abundance of H. oslari decreased by as much as 10‐fold as diel stage change increased, despite the presence of female morphological adaptations for deep‐water oviposition. We found sexual dimorphism and evidence for deep‐water swimming adaptations in 5/6 species.
- Our results show that net‐spinning caddisflies have species‐specific responses to environmental variation and suggest that environmental flows designed to reduce diel stage change and destabilise water temperatures may improve habitat quality for these ubiquitous and important aquatic insects.

Most available techniques for the quantitation of enzymatic degradation of peptide hormones are time-consuming and require expensive equipment and/or novel reagents. Our aim here was to develop a rapid and sensitive assay for the measurement of degradation of cholecystokinin octapeptide (CCK-8) as well as other short, hydrophobic peptides. The proposed technique is based on our novel observation that intact CCK-8, but not its degradation product(s), binds to Lloyd reagent, a form of aluminum silicate. When radiolabeled CCK-8 was exposed to rat liver cytosol containing endogenous CCK-degrading activity, there was a time-dependent decrease in the binding of radiolabel to aluminum silicate [from 86 to 8% over 60 min at 37 degrees C]. The decrease in binding closely paralleled the extent of CCK-8 degradation over time as assessed by high-performance liquid chromatography and immunoprecipitation with specific polyclonal antibodies to CCK-8. While aluminum silicate did not efficiently bind to C-terminal and N-terminal CCK tetrapeptides, magnesium silicate bound to both tetrapeptides (> 82%), but not to their radiolabeled degradation products. Both aluminum and magnesium silicate also extensively bound (> 82%) to other peptide hormones including Met-enkephalin, somatostatin, and secretin, but did not bind their degradation products. We present a modified histochemical method for staining osteoclasts and adjacent mononuclear cells which takes advantage of the recently described substrate specificity for ATP of osteoclastic acid phosphatase. Staining of osteoclasts using ATP as substrate exhibits by light microscopy the same tartrate resistance as conventional acidic phosphatases, without the bone surface staining seen with other substrates. This feature, coupled with specific staining of fewer vicinal mononuclear cells, makes this method potentially useful for studying osteoclast ontogeny and function. 相似文献

Growth hormone secreting cells of the rat anterior pituitary are heavily laden with granules of growth hormone and can be partially purified on the basis of their resulting high density. Two methods of preparative cell electrophoresis were investigated as methods of enhancing the purification of growth hormone producing cells: density gradient electrophoresis and continuous flows electrophoresis. Both methods provided a two- to four-fold enrichment in growth hormone production per cell relative to that achieved by previous methods. Measurements of electrophoretic mobilities by two analytical methods, microscopic electrophoresis and laser-tracking electrophoresis, revealed very little distinction between unpurified anterior pituitary cell suspensions and somatotroph-enriched cell suspensions. Predictions calculated on the basis of analytical electrophoretic data are consistent with the hypothesis that sedimentation plays a significant role in both types of preparative electrophoresis and the electrophoretic mobility of the growth hormone secreting subpopulation of cells remains unknown. 相似文献

Factors shaping community patterns of microorganisms are controversially discussed. Physical and chemical factors certainly limit the survival of individual taxa and maintenance of diversity. In recent years, a contribution of geographic distance and dispersal barriers to distribution patterns of protists and bacteria has been demonstrated. Organismic interactions such as competition, predation and mutualism further modify community structure and maintenance of distinct taxa. Here, we address the relative importance of these different factors in shaping protists and bacterial communities on a European scale using high-throughput sequencing data obtained from lentic freshwater ecosystems. We show that community patterns of protists are similar to those of bacteria. Our results indicate that cross-domain organismic factors are important variables with a higher influence on protists as compared with bacteria. Abiotic physical and chemical factors also contributed significantly to community patterns. The contribution of these latter factors was higher for bacteria, which may reflect a stronger biogeochemical coupling. Azole-resistant environmental Aspergillus fumigatus presents a threat to public health but the extent of this threat in Southeast Asia is poorly described. We conducted environmental surveillance in the Mekong Delta region of Vietnam, collecting air and ground samples across key land-use types, and determined antifungal susceptibilities of Aspergillus section Fumigati (ASF) isolates and azole concentrations in soils. Of 119 ASF isolates, 55% were resistant (or non-wild type) to itraconazole, 65% to posaconazole and 50% to voriconazole. Azole resistance was more frequent in A. fumigatus sensu stricto isolates (95%) than other ASF species (32%). Resistant isolates and agricultural azole residues were overrepresented in samples from cultivated land. cyp51A gene sequence analysis showed 38/56 resistant A. fumigatus sensu stricto isolates carried known resistance mutations, with TR34/L98H most frequent (34/38). 相似文献
