Intein-based biosynthetic incorporation of unlabeled protein tags into isotopically labeled proteins for NMR studies 总被引:6,自引:0,他引:6
Segmental isotopic labeling of proteins using protein ligation is a recently established in vitro method for incorporating isotopes into one domain or region of a protein to reduce the complexity of NMR spectra, thereby facilitating the NMR analysis of larger proteins. Here we demonstrate that segmental isotopic labeling of proteins can be conveniently achieved in Escherichia coli using intein-based protein ligation. Our method is based on a dual expression system that allows sequential expression of two precursor fragments in media enriched with different isotopes. Using this in vivo approach, unlabeled protein tags can be incorporated into isotopically labeled target proteins to improve protein stability and solubility for study by solution NMR spectroscopy. 相似文献

A biomarker of aluminium accumulation in the human body can play a valuable role in determining health effects of chronic aluminium exposure, complementing other human and environmental monitoring data. In vivo neutron activation provides such a non-invasive biomarker. To date, the best in vivo neutron activation system used thermalised neutrons from a nuclear reactor at Brookhaven National Laboratory, which suffered only slightly from interference from other elements, primarily phosphorus, and from the disadvantage of restricted accessibility. At McMaster, we use a nuclear reaction on an accelerator to select neutron energy, which eliminates the interferences. Spectral decomposition analysis improved sensitivity. A new 4pi detection system also enhanced sensitivity. Together these improvements yield a minimum detection limit of 0.24 mgAl in a hand, slightly better than at Brookhaven and equivalent to "normal" levels. Further improvements should result from a new irradiation cavity and from using a higher proton current on the accelerator to shorten irradiation times. The heterotrimeric eukaryotic initiation factor 2 (eIF2) plays a critical role in the mechanics and regulation of protein synthesis. Unlike yeast and archaeal eIF2, the purified baculovirus-expressed recombinant human eIF2 subunits used in these studies reveal that the alpha- and beta-subunits interact with each other. Consistent with this observation, the beta-subunit specifically interacts with the purified eIF2B in ELISA studies and this interaction is enhanced when wt eIF2alpha in the recombinant trimeric complex is phosphorylated or replaced by a mutant phosphomimetic eIF2alpha (S51D). These findings together with other observations raise the possibility that the beta-subunit plays a key role in the regulation and function of mammalian eIF2 complex. PERK, an eIF2alpha kinase, is found to interact with wt and mutants of eIF2alpha in which the serine 51 or 48 residue is replaced by alanine or aspartic acid thereby suggesting that the phosphorylation site in the substrate is not important for interaction. Fluorescence spectroscopic and fluorescence resonance energy transfer analyses reveal that the energy transfer occurs from PERK to eIF2alpha. The dissociation constant of alpha-subunit-PERK complex (Kd alpha-subunit) is 0.74 microM and the interaction is stoichiometric. 相似文献

Abstract: Aggregation pheromone of the Australian population of New Guinea sugarcane weevil, Rhabdoscelus obscurus (Boisduval), in conjunction with other semiochemicals, was used to develop an efficient trapping method for the weevil population in Guam. In a field experiment at Yigo, plastic bucket traps baited with the lure of the Australian R. obscurus population in combination with ethyl acetate and cut sugarcane captured significantly more weevils than traps baited with pheromone + ethyl acetate, pheromone + sugarcane or individual lure components alone. Traps baited with various semiochemical-based lures and treated with insecticide captured significantly greater numbers of weevils than those not treated with insecticide. Traps baited with cut sugarcane caught significantly more weevils than those without sugarcane. Semiochemical-based trapping in weevil management has potential either in mass trapping or as part of an integrated pest management (IPM) programme. Based on the present findings, a future line of work for the control of this weevil is proposed. 相似文献

Here we report, for the first time, the results of detailed GC and GC/MS analyses of the essential oil of a rare taxon in Serbia, Hypericum elegans Stephan ex Willd . One hundred and sixty two constituents identified accounted for 98.6% of the oil. The major components of the oil were undecane (31.9%), α‐pinene (16.7%), nonane (6.1%), bicyclogermacrene (5.8%), 2‐methyloctane (3.7%), and germacrene D (3.6%). Non‐terpenoids as chemotaxonomic markers constituted the main fraction of H. elegans oil, whereby n‐alkanes were the most abundant contributors of this fraction. Based on these results and previously published ones, we performed an intrasectional multivariate statistical comparison of corresponding essential‐oil chemical compositions. Principal component analysis (PCA) and agglomerative hierarchical clustering (AHC) of the data on the volatile profiles of section Hypericum taxa revealed that H. elegans either represents an oil chemotype of its own (AHC) or could be considered related to H. perforatum (PCA). 相似文献

Insect juvenile hormone (JH) has been related to modulation of vitellogenin (Vg) synthesis, a protein produced by fat body cells, secreted in haemolymph and sequestered by developing oocytes. A stimulatory JH action has been described for the majority of species studied thus far. In some insects, however, Vg synthesis has been inhibited or unaffected by JH. The aim of this study was to re-examine the action of JH on Vg synthesis in Apis mellifera workers, since contrasting effects of this hormone were described. Newly emerged worker bees were treated with different doses of pyriproxyfen (PPN), a potent JH analogue. Vg and total protein were quantified in haemolymph samples of newly emerged up to 6-day-old worker bees. Protein synthesis activity of fat body cultured in vitro and ultrastructure of fat body cells were also examined. High doses (1.25, 2.5, 5 and 10 &mgr;g) of PPN inhibited the onset and accumulation of Vg in the haemolymph of young worker bees in a dose-dependent fashion. This inhibition was not a result of fat body cell degeneration or death, as illustrated by fat body cells ultrastructure analysis, but by impairing Vg synthesis, as demonstrated by in vitro culture of fat body cells. Low doses (0.001, 0.01 and 0.1 &mgr;g) neither affected the normal synthesis and secretion of Vg into the haemolymph nor caused an early onset of Vg in treated bees (which could be interpreted as a JH-activating effect), as shown by Vg quantification at 24-h intervals. The results suggest that a low JH titre in honey bee workers permits the onset and accumulation of Vg in haemolymph, whereas high JH levels turn off Vg synthesis. 相似文献

Subscapular skinfold, elbow breadth and upper arm indicators of nutritional status were studied in the population of Dalmatia in Croatia. Age- and sex-specific percentiles were obtained from 4373 subjects, 18 to 74 years of age, and compared to the U.S. NHANES I and II reference data. There were significant differences between these data sets in all studied variables. The results complement those reported previously for BMI and triceps skinfold and indicate that high prevalence of overweight in Dalmatians largely reflects their muscularity and skeletal robustness rather than excess body fatness. The findings suggest that the U.S. upper percentiles of BMI and skinfolds are inadequate for the assessment of excess body fatness in Dalmatian population. The obtained population-specific percentile distributions should be used provisionally as the reference data for group comparisons in the Dalmatian region. 相似文献

The paper presents the results on hereditary and environmental dental findings in identification of human remains exhumed from mass graves in the Republic of Croatia. The total of 17,880 teeth from all the categories (incisors, canines, premolars and molars) was examined. Hereditary findings of the teeth such as shape, size, position, as well as age were used in all of the cases confirming and completing the identification. In only 15% of the cases they were the starting points for the identification that would be later confirmed with another 3-5 traditional identification procedures. Disturbances in tooth eruption were recorded in 22% of the cases, impaction of teeth in 10%, and retarded eruption of teeth in 12%. Disturbances of tooth position were recorded in 65% of the cases. Tooth rotation in 26% and diastema mediana in maxilla or mandible in 39%. Disorders of tooth number in the form of unilateral and bilateral missing of lateral maxillary incisors were recorded only in 2% of the monitored cases. Abnormalities of the tooth shape were found in 11% of the cases. The majority of them were found on the tooth crowns 6%, and less on the tooth roots 5%. Environmental dental findings that were the most significant for the identifications were prosthetic appliances in 30% of cases. Prostheses were helpful in the identification of 3% of the cases, while crowns and bridges were helpful in 27% of the cases. Ante mortem teeth extractions were helpful in 25% of the cases. Teeth restorations were recorded in 20% of the identified cases, amalgams in 19% and aesthetic filings in 1%. Dental caries was helpful in only 10% of the cases, superficial caries in 3% and caries of dentin in 7% of cases. 相似文献

Lom-AKH-I is a member of the adipokinetic hormone/red pigment concentrating hormone (AKH/RPCH) family of peptides found in flying insects. A molecular dynamics simulation at room temperature (293 K) in water has been performed to survey the folding path of the Lom-AKH-I peptide in water and to establish the secondary structure of Lom-AKH-I.
