The reproductive and damage potential of Ditylenchus destructor on peanut, Arachis hypogaea cv. Sellie, was determined in greenhouse tests. Final nematode population densities (Pf) in roots, hulls, and seeds increased (P = 0.01) as a function of increasing initial population (Pi). Final population densities were higher in hulls than in seeds and roots. Final densities in hulls and seeds were positively (P = 0.01) correlated. Fresh root and hull weight and number of pods and seeds per plant were not affected by D. destructor. Second generation germination and pod and seed disease severity increased (P = 0.01), whereas fresh seed weight decreased (P = 0.01) as a function of increasing Pi, and Pf in seeds and Pf in hulls. At Pi 250 and higher, 10-25% of seeds germinated into second generation seedlings before harvest. At Pi 250 and higher, fresh weight of harvested seed was suppressed 20-50%. At Pi 50 or Pf greater than 20 per seed, pod disease severity was 3-7 (on a scale of 1 to 10) and 15-80% of seeds were blemished or unsound.  相似文献

Populations of the staghorn coral, Acropora cervicornis, collapsed throughout the Caribbean region from the late 1970s through the 1990s. We tested the hypothesis that this recent, multidecadal interruption in coral growth was a novel event in the late Holocene. Eight cores, extracted from a lagoonal reef in Discovery Bay, Jamaica dated to 440–1260 CalBP and consisted almost entirely of A. cervicornis rubble. The A. cervicornis in the cores showed significantly less internal bioerosion than A. cervicornis from modern death assemblages in Discovery Bay, indicating generally shorter post‐mortem exposure at the sediment–water interface in the past. A. cervicornis grew continuously and was buried rapidly during the millennium preceding the 1980s, with the exception of a possible hiatus in growth and burial at some point 300–600 years ago. In the 1980s, a combination of perturbations, which included overfishing and (possibly) other forms of human interference, caused an unprecedented disruption in the growth and burial of staghorn coral populations in Discovery Bay.  相似文献

Escherichia coli O157:H7 is an important food-borne pathogen. Often E. coli O157:H7 is difficult to detect, because it is present sporadically at very low levels together with very high levels of competitor organisms which can be difficult to distinguish phenotypically. Cultural methods are time-consuming and give variable results in the detection of E. coli O157:H7. This study examined the performance of BAX for Screening/E. coli O157:H7, a new rapid method for the detection of E. coli O157:H7, against traditional and improved cultural methods and an immunodiffusion assay. All cultural methods demonstrated inadequacy in detecting the presence of E. coli O157:H7 in inoculated samples. The limitations of these cultural methods further complicate evaluation of screening methodologies. The BAX for Screening/E. coli O157:H7 assay outperformed the other methods, with a detection rate of 96.5%, compared to 39% for the best cultural method and 71.5% for the immunodiffusion method. The BAX for Screening/E. coli O157:H7 assay proved to be a rapid, highly sensitive test for the detection of low levels of E. coli O157:H7 in ground beef.  相似文献

Four strains of Butyrivibrio fibrisolvens, transformed with a gene encoding fluoroacetate dehalogenase, maintained a combined population of 106 to 107 cells ml−1 in the rumens of test sheep. Five inoculated sheep showed markedly reduced toxicological symptoms after fluoroacetate poisoning when behavioral, physiological, and histological effects were compared with those of five uninoculated control sheep.  相似文献

Huntington disease is an autosomal dominant neurodegenerative disorder caused by the pathological expansion of a polyglutamine tract. In this study we directly assess the influence of protein size on the formation and subcellular localization of huntingtin aggregates. We have created numerous deletion constructs expressing successively smaller fragments of huntingtin and show that these smaller proteins containing 128 glutamines form both intranuclear and perinuclear aggregates. In contrast, larger NH2-terminal fragments of huntingtin proteins with 128 glutamines form exclusively perinuclear aggregates. These aggregates can form in the absence of endogenous huntingtin. Furthermore, expression of mutant huntingtin results in increased susceptibility to apoptotic stress that is greater with decreasing protein length and increasing polyglutamine size. As both intranuclear and perinuclear aggregates are clearly associated with increased cellular toxicity, this supports an important role for toxic polyglutamine-containing fragments forming aggregates and playing a key role in the pathogenesis of Huntington disease.  相似文献

We have investigated two characteristics of the glutamate system in the developing rabbit retina. 1) Glutamate immunoreactivity was observed at birth within developing processes of four cell types; two of which, photoreceptors and ganglion cells, are known to be glutamatergic in the adult. Two other cell types, type A horizontal cells and amacrine cells, are immunoreactive to both glutamate and GABA at birth, suggesting that endogenous pools of glutamate in GABAergic neurons serve as precursor for GABA synthesis. Thus it appears that endogenous glutamate pools are present within neurons prior to synaptogenesis as part of the early expression of either the glutamate or GABA transmitter phenotype. 2) Analysis of3H-glutamate metabolism during retinal development showed that rapid conversion of glutamate to glutamine does not occur until the second postnatal week, coincident with the expression of Muller (glial) cell activity. In the absence of glial metabolism in the neonate, extracellular concentrations of glutamate remain relatively high and are likely to have major effects on neuronal maturation.Special issue dedicated to Dr. Frederick E. Although serum hormones varied seasonally in all adult animals, only dominant male and female wolves urine-marked. Serum testosterone and urine-marking rates, which increased during the fall/winter breeding season, were positively correlated in both male and female dominant wolves. Estradiol, which increased in conjunction with proestrus and estrus, was not correlated with female urinemarking. These findings suggest that hormonal influence on urine-marking in the wolf is modulated by social factors and contrast with those for both domestic dogs and coyotes, two other members of the genus Canis.  相似文献

The denitrifying behavior of selected soil bacteria was compared in a culture solution and in soil that was sterilized by autoclaving. The essential characteristics concerning nitrate reduction and the formation of nitrogenous gases did not change significantly for most bacteria in the two environments. Bacteria whose denitrification product was nitrous oxide evolved the same gas both in soil and in a liquid system, whereas other bacteria formed only nitrogen gas. The validity of laboratory observations in relation to field studies in the domain of denitrification is discussed and evaluated.  相似文献
