A procedure was developed for the detection of 2′,3′-cyclic nucleotide 3′-phosphohydrolase in myelin. This assay was sufficiently sensitive to detect the low levels of 2′,3′-cyclic nucleotide 3′-phosphohydrolase in human erythrocytes. The 2′,3′-cyclic nucleotide 3′-phosphohydrolase of human erythrocytes was determined to be exclusively associated with the inner (cytosolic) side of the membrane. Leaky ghostsand resealed ghosts were assayed for 2′,3′-cyclic nucleotide 3′-phosphohydrolase, (Ca2+/Mg2+-ATPase, and acetylcholinesterase activity, and the 2′,3′-cyclic nucleotide 3′-phosphohydrolase profile is the same as that of the (Ca2+/Mg2+)-ATPase, an established inner membrane maker. 相似文献

In natural waters, the uptake of transition metals such as copper (Cu) by aquatic biota depends on the activity of the free
cupric ion ({Cu2+}) rather than on total Cu concentration. Thus, an important ecological function of dissolved organic matter (DOM) in aquatic
ecosystems is Cu–DOM complexation, which greatly decreases the {Cu2+}. However, Cu bioavailability is greatly modified by source and environmental history of DOM because DOM affinity for Cu
varies by orders of magnitude among DOM sources; moreover, DOM is photochemically unstable. During 72-h irradiation experiments
at intensities approximating sunlight with DOM from a palustrine wetland and a third-order river, we investigated photooxidative
effects on DOM complexation of Cu as well as spectral and chemical changes in DOM that might explain altered Cu complexation.
Irradiation decreased Cu complexation by riverine DOM, but unexpectedly increased Cu complexation by wetland DOM, resulting
in 150% greater {Cu2+} in riverine DOM at the same dissolved organic carbon concentrations. The specific ultraviolet absorption (SUVa) and humic substances tracked photochemical changes in the conditional stability constants of Cu–DOM complexes, suggesting
that the aromaticity of DOM influences its affinity for Cu. Carbonyl concentration in 13C nuclear magnetic resonance spectra (13C-NMR) covaried directly with Cu binding-site densities in DOM. However, no aspect of Cu–DOM complexation consistently covaried
with fluorophores (i.e., the fluorescence index) or low molecular weight organic acids. Our results suggest that global increases
in UV radiation will affect Cu–DOM complexation and subsequent Cu toxicity depending on light regime as well as DOM source.

Bcor (BCL6 corepressor) is a widely expressed gene that is mutated in patients with X-linked Oculofaciocardiodental (OFCD) syndrome. BCOR regulates gene expression in association with a complex of proteins capable of epigenetic modification of chromatin. These include Polycomb group (PcG) proteins, Skp-Cullin-F-box (SCF) ubiquitin ligase components and a Jumonji C (Jmjc) domain containing histone demethylase. To model OFCD in mice and dissect the role of Bcor in development we have characterized two loss of function Bcor alleles. We find that Bcor loss of function results in a strong parent-of-origin effect, most likely indicating a requirement for Bcor in extraembryonic development. Using Bcor loss of function embryonic stem (ES) cells and in vitro differentiation assays, we demonstrate that Bcor plays a role in the regulation of gene expression very early in the differentiation of ES cells into ectoderm, mesoderm and downstream hematopoietic lineages. Normal expression of affected genes (Oct3/4, Nanog, Fgf5, Bmp4, Brachyury and Flk1) is restored upon re-expression of Bcor. Consistent with these ES cell results, chimeric animals generated with the same loss of function Bcor alleles show a low contribution to B and T cells and erythrocytes and have kinked and shortened tails, consistent with reduced Brachyury expression. Together these results suggest that Bcor plays a role in differentiation of multiple tissue lineages during early embryonic development. 相似文献

West Indian spiny lobsters, Panulirus argus, synthesized a hemolymph bactericidin after being injected with killed suspensions of gram-negative bacillus EMB-1 isolated from the normal gut of this lobster. To study differences between the primary response and secondary response, animals were given a primary antigen injection of EMB-1 followed by a second injection of the same antigen 22 to 51 days later. As a rule, secondary bactericidal responses were enhanced over the primary in a manner reminiscent of specific anamnesis in mammalian immunoglobulin synthesis. Immunological memory was also suggested when tertiary responses were compared to secondary and by the persistence of residual titers for many days or weeks without additional antigenic stimulation. 相似文献

The efficiency of transformation of pneumococcus and a strain of viridans streptococcus (strain D) to streptomycin resistance is influenced by the species in which the mutation to resistance occurred, as well as by the species in which the mutated gene has been replicated. Pneumococcus and streptococcus strain D transform in higher frequency with DNA that has been replicated in bacteria of the same species than with DNA from the heterologous species. However, the difference between the frequencies of interspecific and intraspecific transformation is much greater with pneumococcus as receptor than with streptococcus. In addition pneumococcus transforms in higher frequency with wholly homologous (pneumococcal) DNA than with DNA from pneumococci that have replicated the streptococcal Smr gene. Pneumococcus is transformed in lower frequency by wholly heterologous (streptococcal) DNA than by DNA from streptococci that have replicated the pneumococcal Smr gene. Streptococcus behaves similarly in that wholly homologous (streptococcal) DNA transforms it more efficiently than when the transforming fragment contains a pneumococcal moiety. Streptococcus is transformed in the same or lower frequency by wholly heterologous (pneumococcal) DNA than by DNA from pneumococci that have replicated the streptococcal Smr gene. When erythromycin resistance was used as genetic marker instead of streptomycin resistance, similar results were found. 相似文献

Animal toxins are small proteins built on the basis of a few disulfide bonded frameworks. Because of their high variability in sequence and biologic function, these proteins are now used as templates for protein engineering. Here we report the extensive characterization of the structure and dynamics of two toxin folds, the "three-finger" fold and the short alpha/beta scorpion fold found in snake and scorpion venoms, respectively. These two folds have a very different architecture; the short alpha/beta scorpion fold is highly compact, whereas the "three-finger" fold is a beta structure presenting large flexible loops. First, the crystal structure of the snake toxin alpha was solved at 1.8-A resolution. Then, long molecular dynamics simulations (10 ns) in water boxes of the snake toxin alpha and the scorpion charybdotoxin were performed, starting either from the crystal or the solution structure. For both proteins, the crystal structure is stabilized by more hydrogen bonds than the solution structure, and the trajectory starting from the X-ray structure is more stable than the trajectory started from the NMR structure. The trajectories started from the X-ray structure are in agreement with the experimental NMR and X-ray data about the protein dynamics. Both proteins exhibit fast motions with an amplitude correlated to their secondary structure. In contrast, slower motions are essentially only observed in toxin alpha. The regions submitted to rare motions during the simulations are those that exhibit millisecond time-scale motions. Lastly, the structural variations within each fold family are described. The localization and the amplitude of these variations suggest that the regions presenting large-scale motions should be those tolerant to large insertions or deletions. 相似文献

A strong foundation of basic and applied research documents that the estuarine fish Fundulus heteroclitus and related species are unique laboratory and field models for understanding how individuals and populations interact with their environment. In this paper we summarize an extensive body of work examining the adaptive responses of Fundulus species to environmental conditions, and describe how this research has contributed importantly to our understanding of physiology, gene regulation, toxicology, and ecological and evolutionary genetics of teleosts and other vertebrates. These explorations have reached a critical juncture at which advancement is hindered by the lack of genomic resources for these species. We suggest that a more complete genomics toolbox for F. heteroclitus and related species will permit researchers to exploit the power of this model organism to rapidly advance our understanding of fundamental biological and pathological mechanisms among vertebrates, as well as ecological strategies and evolutionary processes common to all living organisms. 相似文献
