Candidate phyla (CP) are broad phylogenetic clusters of organisms that lack cultured representatives. Included in this fraction is the candidate Parcubacteria superphylum. Specific characteristics that have been ascribed to the Parcubacteria include reduced genome size, limited metabolic potential and exclusive reliance on fermentation for energy acquisition. The study of new environmental niches, such as the marine versus terrestrial subsurface, often expands the understanding of the genetic potential of taxonomic groups. For this reason, we analyzed 12 Parcubacteria single amplified genomes (SAGs) from sediment samples collected within the Challenger Deep of the Mariana Trench, obtained during the Deepsea Challenge (DSC) Expedition. Many of these SAGs are closely related to environmental sequences obtained from deep‐sea environments based on 16S rRNA gene similarity and BLAST matches to predicted proteins. DSC SAGs encode features not previously identified in Parcubacteria obtained from other habitats. These include adaptation to oxidative stress, polysaccharide modification and genes associated with respiratory nitrate reduction. The DSC SAGs are also distinguished by relative greater abundance of genes for nucleotide and amino acid biosynthesis, repair of alkylated DNA and the synthesis of mechanosensitive ion channels. These results present an expanded view of the Parcubacteria, among members residing in an ultra‐deep hadal environment.  相似文献

Purpose

Bioethanol is not currently produced in Chile. However, mixtures of bioethanol-gasoline at 2 and 5 % have been authorized. The production and use of the bioethanol-gasoline blend “E5” has been assessed using life cycle assessment (LCA) with the aim to compare the environmental profiles of bioethanol produced from Eucalyptus globulus with gasoline in Chile and to determine the potential of this biofuel-replacing gasoline in the transport sector.

Methods

The standard framework of LCA described by ISO was selected to assess the ecological burdens derived from the biofuel production using the SimaPro v7.8 software. The system boundaries included eucalyptus cultivation, bioethanol production, E5 blend production, and final use of E5. The inventory data for Eucalyptus cultivation were previously collected through surveys with forest managers. Inventory data for bioethanol production were obtained by process simulation models using Aspen Plus v7.1, and for non-simulated or modeled information, secondary information (scientific articles and reports) was used. Conventional gasoline, produced and used in Chile, was used as base scenario for comparison with E5 scenario.

Results and discussion

The environmental results showed reduction of the environmental impacts in most of the assessed categories when E5 blend is assessed and compared with gasoline. Reduction was evident for climate change, photochemical oxidation formation, terrestrial acidification, marine eutrophication, terrestrial ecotoxicity, marine ecotoxicity, depletion of water, and fossil resources. However, there was an increase in other impact categories, such as ozone layer depletion, human toxicity, terrestrial ecotoxicity, and marine eutrophication. The hotspots for E5 blend were the blending production and the combustion in the engine, whereas in the production process, the electricity production was the major contributor to most of the impact categories. When increasing the bioethanol content from E5 to E10 blend, the environmental impact increases in most of the evaluated categories except in the CC, WD, and FD categories. However, compared with other studies related to wood-based E10, the values for the environmental impacts obtained were lower than the reported.

Conclusions

The use of E5 blend can help to reduce the environmental impact in 8 of the 12 categories analyzed. Environmental impacts obtained are lower compared with other studies reported for E10 blend production from wood resources.

The effects of castration of both resident and intruder rats on territorial aggressive behavior were studied. The results suggest that residence in a home cage is more important than gonadal status in determining the outcome of an aggressive encounter. Resident rats were more likely to be dominant especially if they were intact. Intact residents directed less aggressive behavior toward castrated intruders than toward intact intruders. Intruder rats generally showed low levels of aggressive behavior and were only dominant when the resident had been castrated. Thus, the aggressive behavior of a male rat depends upon both his gonadal status and that of his opponent.  相似文献

Summary Water-soluble Folch-Lees proteolipid apoprotein from bovine CNS white matter induces a voltage-dependent conductance in black lipid membranes. Na+ is required for the induced conductance change but the established conductance has very low ionic selectivity. The induced conductance fluctuates with a minimum amplitude of 10–11–10–10 mho. The magnitude of the conductivity change is dependent on protein concentration and on the composition of lipid bilayers. At a fixed voltage the induced conductance of a phosphatidylcholine-cholesterol membrane is proportional to the sixth power of the protein concentration and the first power of Na+ concentration. The interactions between the apoprotein and the lipids are both electrostatic and hydrophobic, but the interaction leading to the conductance increase appears to be mainly hydrophobic. Both the increase in conductance and the current fluctuations remain after extensive washing of the chambers to remove the protein. Furthermore, pronase or glutaraldehyde added to either the cis or trans side of the membrane does not affect the apoprotein-established conductance. However, if the bilayer is formed in the presence of both the apoprotein and pronase or if the apoprotein is treated with pronase prior to its addition to the chamber, no conductance change is observed. The association of the apoprotein with the membrane thus appears to render the protein inaccessible to proteolytic digestion, suggesting that the apoprotein is at least partially imbedded in the membrane interior.  相似文献

Reactive oxygen species (ROS) production by an NADPH oxidase (NOX) encoded by AtrbohC/RHD2 is required for root hair growth in Arabidopsis thaliana. ROP (RHO of plants) GTPases are also required for normal root hair growth and have been proposed to regulate ROS production in plants. Therefore, the role of ROP GTPase in NOX-dependent ROS formation by root hairs was investigated. Plants overexpressing wild-type ROP2 (ROP2 OX), constitutively active (CA-rop2), or dominant negative (DN-rop2) rop2 mutant proteins were used. Superoxide formation by root hairs was detected by superoxide dismutase-sensitive nitroblue tetrazolium reduction, and ROS production in the root hair differentiation zone was detected by dihydrofluorescein diacetate oxidation. Both probes showed that ROS production was increased in ROP2 OX and CA-rop2 plants, and decreased in DN-rop2 plants, relative to wild-type plants. When CA-rop2 was expressed in the NOX loss-of-function rhd2-1 mutant, ROS formation and root hair growth were impaired, suggesting that RHD2 is required for this ROP2-dependent ROS formation.  相似文献

The high intraluminal concentrations of HCO(3)(-) in the human pancreatic ducts have suggested the existence of a membrane protein supplying the Cl(-)/HCO(3)(-) exchanger. Membrane-bound carbonic anhydrase IV (CA IV) is one of the potential candidates for this protein. The difficulties in isolating human pancreatic ducts have led the authors to study the molecular mechanisms of HCO(3)(-) secretion in cancerous cell lines. In this work, we have characterized the CA IV expressed in Capan-1 cells. A 35-kDa CA IV was detected in cell homogenates and purified plasma membranes. Treatment of purified plasma membranes with phosphatidylinositol-phospholipase-C indicated that this CA IV was not anchored by a glycosylphosphatidylinositol (GPI). In contrast, its detection on purified plasma membranes by an antibody specifically directed against the carboxyl terminus of human immature GPI-anchored CA IV indicated that it was anchored by a C-terminal hydrophobic segment. Immunoelectron microscopy and double-labeling immunofluorescence revealed that this CA IV was present on apical plasma membranes, and in the rough endoplasmic reticulum, the endoplasmic reticulum-Golgi intermediate compartment, the Golgi complex, and secretory granules, suggesting its transport via the classical biosynthesis/secretory pathway. The expression in Capan-1 cells of a 35-kDa CA IV anchored in the apical plasma membrane through a hydrophobic segment, as is the case in the healthy human pancreas, should make the study of its role in pancreatic HCO(3)(-) secretion easier.  相似文献

Aerial dispersal of the phytoseiid Metaseiulus occidentalis (Nesbitt) was evaluated as a component in managing pesticide-resistant populations established in California almond orchards. Peak dispersal occurred in late July and early August during 1982 and 1983. Most predators (and spider mites) left the orchards on the prevailing winds from the northwest. Within the orchard, the prevailing winds had less influence, and dispersal was usually random. Both spider mites and predators dispersed randomly with regard to height from the almond trees, but data obtained during one 24-h interval suggest they do not disperse randomly throughout the day. Most aerial movements occurred between 16–22 h when relative humidity and wind speeds increased and temperatures decreased. Spider mites and predators were trapped on panels located 200 m from the orchard. A survey of carbaryl resistance levels in M. occidentalis collected from almond orchards surrounding the release sites indicates that carbaryl-resistant M. occidentalis dispersed at least 800 m between 1981–83. However, growers wishing to use the resistant strains should release them in their orchards as natural dispersal appears to be too slow. Migration of native M. occidentalis into the release sites appeared to be sufficiently rare that dilution of carbaryl-resistant populations was minimal during a 2–4 year period.
Résumé La dispersion aérienne du phytoseïdae, M. occidentalis (Nesbitt), a été estimée comme élément de la lutte contre les populations résistantes aux insecticides établies dans les vergers de Californie. La dispersion maximale s'est produite fin juillet et début a oût en 1982 et 1983. La plupart des prédateurs (et des acariens) quittent les vergers avec les vents dominants du nordouest. Dans le verger, les vents dominants sont moins importants et la dispersion est généralement au hasard. Tant les acariens que les prédateurs se dispersaient au hasard par rapport à la taille des amandiers, mais les relevés sur 24 heures laissent supposer qu'il n'y a pas une distribution aléatoire pendant la journée. La plupart des mouvements aériens se produisirent entre 16 et 22 heures quand HR et vitesse du vent augmentaient et température diminuait. Les acariens et prédateurs ont été piégés sur des panneaux à 200 m du verger.

The yeast communities associated with the stingless bees Tetragonisca angustula, Melipona quadrifasciata and Frieseomelitta varia were studied. The bees T. angustula and F. varia showed a strong association with the yeast Starmerella meliponinorum. M. quadrifasciata more frequently carried a species related to Candida apicola, but also vectored low numbers of S. meliponinorum. Some of the yeasts isolated from adult bees were typical of species known to occur in flowers. Other yeast species found in adult bees were more typical of those found in the phylloplane. S. meliponinorum and the species in the C. apicola complex, also part of the Starmerella clade, may have a mutualistic relationship with the bees studied. Many yeasts in that group are often found in bees or substrates visited by bees, suggesting that a mutually beneficial interaction exists between them.  相似文献

Acinetobacter calcoaceticus RAG-1 and MR-481, two standard strains used in microbial adhesion to hydrocarbons (MATH), were characterized by contact angles, pH-dependent zeta potentials, elemental surface composition by X-ray photoelectron spectroscopy (XPS), and molecular composition by infrared spectroscopy (IR). Negatively stained (methylamine tungstate) and ruthenium red-stained cells were studied by transmission electron microscopy to reveal the absence or presence of surface appendages. Despite the fact thatA. calcoaceticus RAG-1 is known to be extremely hydrophobic in MATH, whereas MR-481 is a completely non-hydrophobic mutant, neither XPS nor IR indicated a significant difference in chemical composition of the cell surfaces. Contact angles with polar liquids, water and formamide, were considerably higher on RAG-1 than on MR-481, in accordance with their relative hydrophobicities as measured by MATH. However, no significant differences in contact angles were observed between the two strains with apolar liquids like diiodomethane,-bromonaphthalene, and hexadecane. Fibrous extensions on RAG-1, observed after ruthenium red staining, were absent on the non-hydrophobic mutant MR-481. Tentatively, these extensions could be held responsible for the hydrophobicity ofA. calcoaceticus RAG-1.  相似文献
