The objective of this study was to compare six samples of Mexican wild common bean (Phaseolus vulgaris L.) against three landraces and three improved cultivars with respect to physical and chemical attributes, and the culinary quality potential of their grain. A completely randomized experimental design was used to characterize the twelve genotypes. Data were analyzed by analysis of variance and pair-wise comparison of the treatment means by the Tukey test. In addition, correlation and principal-component analysis (PCA) were carried out using twelve characteristics of raw and four of cooked wild and domesticated grains. The results show a larger variability of the physical and chemical characteristics in wild than in domesticated beans. The PCA confirmed that grain gigantism was the main physical characteristic resulting of domestication, whereas the protein and tryptophan contents tended to be higher in wild than domesticated genotypes. Some wild samples from Chihuahua and Durango, Mexico, showed to be a genetic resource to improve food quality, because of their richness in minerals, protein, lysine, tryptophan, and dietary fibers.  相似文献

A total of 33 crude and cloned Trypanosoma rangeli stocks found as natural infections in human from Panama and other endemic areas of Central and South America were evaluated as producers of sialidase (SA) activity through the MU-NANA fluorescence test. Negative results were observed in 6 of the isolates: Panama (4), Honduras (1), and Brazil (1). In addition, an immunoblotting analysis confirm the presence of the SA antigen in these stocks without enzymatic activity. These findings must be considered in the interpretation of the biological significance of T. rangeli SA and in the proper characterization and identification of this parasite.  相似文献

In vitro physiology and carbon metabolism can be affected by the sink–source relationship. The effect of different sucrose concentrations (10, 30, and 50 g L−1), light intensities (80 and 150 μmol m−2 s−1), and CO2 levels (375 and 1,200 μmol mol−1) were tested during plantain micropropagation in temporary immersion bioreactors. Activities of pyruvate kinase, phosphoenol pyruvate carboxylase, and the photosynthesis rate were recorded. From the morphological and practical point of view, the best results were obtained when plants were cultured with 30 g L−1 sucrose, 80 μmol m−2 s−1 light intensity, and 1,200 μmol mol−1 CO2 concentration. This treatment improved leaf and root development, reduced respiration during in vitro culture, and increased starch level at the end of the hardening phase. In addition to that, the number of competent plants was increased from 80.0% to 91.0% at the end of the in vitro phase and the survival percentage from 95.71% to 99.80% during ex vitro hardening.  相似文献

Differences in the distribution of diverse cell surface coat markers were found between Naegleria fowleri and Naegleria lovaniensis. The presence of carbohydrate-containing components in the cell coat of the two species was detected by selective staining with ruthenium red and alcian blue. Using both markers, N. fowleri presented a thicker deposit than N. lovaniensis. The existence of exposed mannose or glucose residues was revealed by discriminatory agglutination with the plant lectin Concanavalin A. These sugar residues were also visualized at the cell surface of these parasites either by transmission electron microscopy or by fluorescein-tagged Concanavalin A. Using this lectin cap formation was induced only in N. fowleri. The anionic sites on the cell surface detected by means of cationized ferritin were more apparent in N. fowleri. Biotinylation assays confirmed that even though the two amoebae species have some analogous plasma membrane proteins, there is a clear difference in their composition.  相似文献

In herbivorous insects, the interaction between adult preference and progeny performance on specific host plants is modified by maternal feeding experience and host plant quality. Ultimately, changes in the strength of this interaction can affect insect population dynamics. In this study, we hypothesized that adult host plant preference influences progeny performance through a maternal feeding experience × host plant interaction, that is, the effect of adult feeding experience on progeny performance will depend on the host plant. Second, that decoupling of the preference–performance relationship due to host switching results in different population vital rates changing population dynamics. An increase in development time and a decrease in body size of individuals in the alternate host should decrease population growth. We tested these hypotheses using two lines of the tortoise beetle Chelymorpha varians Blanchard fed with two hosts (Convolvulus arvensis and Calystegia sepium). Maternal feeding experience treatments were crossed with host plant species, and the offspring’s developing time and adult size were measured. The host plant influence on the beetle’s population vital rates was tested using stage-structured matrix population models and life table response experiments. Host plant preference affected offspring body size through a host plant effect that contributed to adaptive life history responses only in the better quality host. C. varians’ population growth was positive when fed with either host; comparatively, however, C. sepium had a negative effect on growth by reducing all transition probabilities of the life cycle stages of the beetle. Here, we show that individuals of C. varians prefer and perform differently on distinct hosts and that these patterns influence population vital rates in different ways. When beetles prefer the host plant where their progeny performs best, life history responses and life stage transitions lead to higher population growth; otherwise, growth rate decreases.  相似文献

During malaria infection, high levels of proinflammatory molecules (e.g., cytokines, chemokines) correlate with disease severity. Even if their role as activators of the host immune response has been studied, the direct contribution of hemozoin (HZ), a parasite metabolite, to such a strong induction is not fully understood. Previous in vitro studies demonstrated that both Plasmodium falciparum HZ and synthetic HZ (sHZ), beta-hematin, induce macrophage/monocyte chemokine and proinflammatory cytokine secretion. In the present study, we investigated the proinflammatory properties of sHZ in vivo. To this end, increasing doses of sHZ were injected either i.v. or into an air pouch generated on the dorsum of BALB/c mice over a 24-h period. Our results showed that sHZ is a strong modulator of leukocyte recruitment and more specifically of neutrophil and monocyte populations. In addition, evaluation of chemokine and cytokine mRNA and protein expression revealed that sHZ induces the expression of chemokines, macrophage-inflammatory protein (MIP)-1alpha/CCL3, MIP-1beta/CCL4, MIP-2/CXCL2, and monocyte chemoattractant protein-1/CCL2; chemokine receptors, CCR1, CCR2, CCR5, CXCR2, and CXCR4; cytokines, IL-1beta and IL-6; and myeloid-related proteins, S100A8, S100A9, and S100A8/A9, in the air pouch exudates. Of interest, chemokine and cytokine mRNA up-regulation were also detected in the liver of i.v. sHZ-injected mice. In conclusion, our study demonstrates that sHZ is a potent proinflammatory agent in vivo, which could contribute to the immunopathology related to malaria.  相似文献

Analytical data are presented for the polysaccharide of six gum specimens from Acacia tortuosa, a tropical American Gummiferae species. These data show that the gums were basically similar but differed in the sugar composition reported for most Acacia gum exudates studied so far. The presence of traces of xylose (<1%) and the absence of rhamnose in the gum specimens studied was very unusual.  相似文献
