The activity of the main enzymes related to the sucrose metabolism, photosynthesis, and sucrose concentration were studied in sugarcane (Saccharum spp hybrid) plantlets. Acclimatization was developed in two steps. (1) Light intensity of 1,000 μmol m−2 s−1 and 90% relative humidity during the first 21 d; followed by 2,000 μmol m−2 s−1 and approximately 80% of relative humidity. All measurements were carried out at the end of rooting phase concomitant with day 0 of acclimatization and at 7-d intervals thereafter (0, 7, 14, 21, 28, 35, 42 d). As the in vitro plantlets were transferred to the acclimatization phase, photosynthesis increased significantly during the first 7 d. After this period, the increase was constant with only a small but nonsignificant decline after being transferred to the uncontrolled external conditions. The activity of the sucrose synthase began to show a decrease, starting from day 7, and was related to the changes that began to happen in these plants from its adaptation to new ex vitro conditions. Due to the increase of fresh weight favored by the high light intensity and lower relative humidity, an increase of the sucrose phosphate synthase activity was observed. The maximum activity of the acid and neutral invertases was reached at 14 and 21 d, respectively, after 21 d of acclimatization period. There was a marked tendency for the activity of both enzymes to decrease. The sucrose content was decreased only in the first 7 d. The metabolism of sugarcane plantlets seemed to be susceptible to the environmental changes during the acclimatization phase but did not contribute to inhibitory factors for normal development.  相似文献

Cu(2+) ions and GSH molecules interact swiftly to form the complex Cu(I)-glutathione. We investigated the potential capacity of such complex to reduce molecular oxygen. The addition of SOD to a solution containing Cu(I)-glutathione led to a sustained decline of the basal oxygen level. Such effect was partially reverted by the addition of catalase. The complex was able to induce the reduction of cytochrome c and the oxidation of dyhydroethidium into 2-hydroxyethidium. Both effects were totally blocked by SOD. The ability of the complex to generate superoxide radicals was confirmed by EPR spin-trapping. Cu(I)-glutathione induces no oxidation of fluorescein, a hydroxyl radical-sensitive probe. We conclude that in solutions containing the complex, oxygen is continually reduced into superoxide, and that-in absence of interceptors-the latter radicals are quantitatively re-oxidized into molecular oxygen. We suggest that, by functioning as a continuous source of superoxide, the complex could potentially affect a broad range of susceptible biological targets.  相似文献

Some ultrastructural changes can be observed in diseased Saccharum officinarum L. (cv. Cuba 120-78) plants with visual symptoms of yellow leaf syndrome (YLS), used to discriminate between healthy and diseased plants. Abaxial epidermis of diseased leaves shows a large amount of adhered superficial bodies, which partially occluded some stomata. Bundle sheath cells surrounding the bottom of phloem of diseased leaves are separated from the conducting tissues by a large layer of an amorphous matrix similar to wax. Debris of the end wall can be observed in large xylem vessels. Sometimes, spherical bodies similar to phytoplasma can be observed in the intercellular spaces of bundle sheath cells. These particles have never been observed in healthy plants. YLS was also associated to an increase of the concentration of reducing sugars, glucose index, and glycoproteins recovered in juices whereas the amount of sucrose decreases. Sugarcane juices obtained from both healthy and YLS-affected Cuba 120-78 cultivars of sugarcane contained putrescine (PUT), cadaverine (CAD), spermidine and spermine (SPM) as free and macromolecules-conjugated compounds. Only CAD and SPM appeared as acid-soluble conjugates to small molecules whereas PUT and CAD are the major polyamines (PAs) conjugated to macromolecules, mainly to high molecular mass glycoproteins. The disease was associated to an increase in total PA fraction. Arginase and ornithine decarboxylase activities, responsible for the synthesis of PUT, were higher in YLS juices than in those obtained from healthy plants. CAD and SPM presumably conjugated mostly to chlorogenic, syringic and ferulic acids in juices from YLS plants.  相似文献

Dengue fever (DF) and dengue hemorrhagic fever/dengue shock syndrome (DHF/DSS) are considered the most important arthropod-borne viral diseases in terms of morbidity and mortality. The emergency and severity of dengue (Den) infections increase the necessity of an early, quick and effective dengue laboratory diagnostic. Viral isolation is considered a gold standard for diagnosis of dengue infection using monoclonal antibodies (mAbs) as a tool for determining serotype specificity. Alternatives have been used to improve sensitivity and time to dengue diagnosis. Based on the early expression of dengue C protein in the life cycle, we focused our study on the application of an anti-dengue 2 virus capsid protein mAb in dengue diagnosis. The kinetic expression of dengue-2 capsid in mosquito cells and its immuno-localization in experimentally infected suckling albin Swiss (OF-1) mice brain tissues was established. The results demonstrate the possible utility of this mAb in early dengue diagnosis versus traditional isolation. In addition, a preliminary study of an enzyme immunoassay method using 8H8 mAb for specific detection of dengue C protein antigen was performed, making possible recombinant C protein quantification. In lizards, one of the most important behavioral mechanisms to cope with spatial and temporal variations in thermal resources observed is activity time. The longer a lizard can maintain activity, the more time it has to forage and reach larger adult body size. We studied the behavioral adjustments to different climatic regimens on daily and seasonal scales in three natural populations of the lizard Microlophus atacamensis along a latitudinal temperature and rainfall gradient. We also used Niche Mapper to determinate the amount of thermally suitable time for activity for this species. Abundance and daily activity patterns varied greatly over the year for the three populations. In summer and spring, the daily activity times were greater, and were reduced in fall and winter seasons. In summer, when stressful heat loads should prohibit activity over a midday gap, lizards did not show bimodal patterns of activity. Instead, they move to the cooler intertidal habitat. Abundance and thermal quality in the southernmost coolest site was lower, and the potential annual activity time decreases with latitude. Contrary to expectations, lizards from this locality showed the largest body sizes possibly due to diet and/or time to sexual maturation. Our results indicate that the intertidal habitat is a key factor that influences daily and seasonal activity of M. atacamensis lizards. While this habitat is not climatically optimal for lizards, it allows them to behaviorally extend their activity window and gain access to food in the intertidal areas.  相似文献

Proteolysis of the Glu441-Ala442 bond in the glycosaminoglycan (GAG) β domain of the versican-V1 variant by a disintegrin-like and metalloproteinase domain with thrombospondin type 1 motif (ADAMTS) proteases is required for proper embryo morphogenesis. However, the processing mechanism and the possibility of additional ADAMTS-cleaved processing sites are unknown. We demonstrate here that if Glu441 is mutated, ADAMTS5 cleaves inefficiently at a proximate upstream site but normally does not cleave elsewhere within the GAGβ domain. Chondroitin sulfate (CS) modification of versican is a prerequisite for cleavage at the Glu441-Ala442 site, as demonstrated by reduced processing of CS-deficient or chondroitinase ABC-treated versican-V1. Site-directed mutagenesis identified the N-terminal CS attachment sites Ser507 and Ser525 as essential for processing of the Glu441-Ala442 bond by ADAMTS5. A construct including only these two GAG chains, but not downstream GAG attachment sites, was cleaved efficiently. Therefore, CS chain attachment to Ser507 and Ser525 is necessary and sufficient for versican proteolysis by ADAMTS5. Mutagenesis of Glu441 and an antibody to a peptide spanning Thr432-Gly445 (i.e. containing the scissile bond) reduced versican-V1 processing. ADAMTS5 lacking the C-terminal ancillary domain did not cleave versican, and an ADAMTS5 ancillary domain construct bound versican-V1 via the CS chains. We conclude that docking of ADAMTS5 with two N-terminal GAG chains of versican-V1 via its ancillary domain is required for versican processing at Glu441-Ala442. V1 proteolysis by ADAMTS1 demonstrated a similar requirement for the N-terminal GAG chains and Glu441. Therefore, versican cleavage can be inhibited substantially by mutation of Glu441, Ser507, and Ser525 or by an antibody to the region of the scissile bond.  相似文献
