Botanical extracts are a plentiful resource of molecules with different biological activities, such as insecticides and antimicrobial pesticides. In this context, the aim of this work was to evaluate the efficacy of botanical extracts from the Brazilian savannah against Diabrotica speciosa and bacterial strains isolated from the gut of this insect under aseptic conditions. The bacterial isolates were identified by genomic and proteomic approaches, and bioassayed against eighteen botanical extracts in vitro. The best results of bacterial inhibitions were obtained for the extracts of Casearia sylvestris and Psidium laruotteanum. Fractions of C. sylvestris and P. laruotteanum, quantitatively evaluated by chromatographic analyses, showed a relationship between the bactericidal activity and phytochemical profile. In vivo assays showed that P. laruotteanum was also effective for the control of D. speciosa. Those results show that selected natural products can have both antimicrobial and insecticidal activities.  相似文献

The development of an effective immune response can help decrease mortality from malaria and its clinical symptoms. However, this mechanism is complex and has significant inter-individual variation, most likely owing to the genetic contribution of the human host. Therefore, this study aimed to investigate the influence of polymorphisms in genes involved in the costimulation of B-lymphocytes in the naturally acquired humoral immune response against proteins of the asexual stage of Plasmodium vivax. A total of 319 individuals living in an area of malaria transmission in the Brazilian Amazon were genotyped for four SNPs in the genes CD40, CD40L, BLYS and CD86. In addition, IgG antibodies against P. vivax apical membrane antigen 1 (PvAMA–1), Duffy binding protein (PvDBP) and merozoite surface protein 1 (PvMSP–119) were detected by ELISA. The SNP BLYS –871C>T was associated with the frequency of IgG responders to PvAMA–1 and PvMSP–119. The SNP CD40 –1C>T was associated with the IgG response against PvDBP, whereas IgG antibody titers against PvMSP–119 were influenced by the polymorphism CD86 +1057G>A. These data may help to elucidate the immunological aspects of vivax malaria and consequently assist in the design of malaria vaccines.  相似文献

The possible role of sialic acids in host cells–fungi interaction and their association with glycoproteins were evaluated using a clinical isolate of the dimorphic fungus Mucor polymorphosporus. Lectin-binding assays with spores and yeast cells denoted the presence of surface sialoglycoconjugates containing 2,3- and 2,6-linked sialylglycosyl groups. Western blotting with peroxidase-labeled Limulus polyphemus agglutinin revealed the occurrence of different sialoglycoprotein types in both cell lysates and cell wall protein extracts of mycelia, spores, and yeasts of M. polymorphosporus. Sialic acids contributed to the surface negative charge of spores and yeast forms as evaluated by adherence to a cationic substrate. Sialidase-treated spores were less resistant to phagocytosis by human neutrophils and monocytes from healthy individuals than control (untreated) fungal suspensions. The results suggest that sialic acids are terminal units of various glycoproteins of M. polymorphosporus, contributing to negative charge of yeasts and spore cells and protecting infectious propagules from destruction by host cells.  相似文献

The in vitro effects of amphotericin B deoxycholate suspension (fungizone) on Paracoccidioides brasiliensis growth, cell viability and transformation were investigated. We also analyzed the protein synthesis patterns of both cellular forms, yeast and mycelium in the presence of AmB. This drug, at 30 μg/ml, highly inhibited yeast growth, which could be recovered depending on treatment time, where the most effective reversion was observed after 6 hr of incubation. The yeast cell viability, that had been partially affected by the drug, could also be efficiently recovered after AmB was removed. The effect of AmB on the cellular dimorphism process showed a strong reduction in the mycelium to yeast transformation (80% inhibition compared to the control without the drug). On the other hand, the transformation from yeast to mycelium in the presence of AmB was 50% affected, relative to the control. In contrast to the growth and cell viability experiments, the reversion effects on dimorphism were partial when the drug was removed, even with only 6 hr treatment. The two-dimensional gels of 35S-labeled proteins revealed a strong reduction in the three species of 80, 71 and 56 kDa in yeast and mycelium when treated with AmB.  相似文献

Blooms of toxic cyanobacteria are very common in Brazilian waterbodies, as a consequence of eutrophication processes. Our investigations were focused on the detection of neurotoxins during a cyanobacterial bloom in Tapacurá reservoir, which serves as a water supply for Recife city in northeastern Brazil. We also investigated the possible presence of neurotoxins in strains of Anabaena spiroides isolated from this environment. Samples were collected from March to May 2002 at the water surface and close to the dam. Limnological parameters (conductivity, pH, inorganic nutrients) and cyanobacterial abundance were measured. The samples were assayed for toxicity by mouse bioassay and acetylcholinesterase-inhibiting activity by a colorimetric method; saxitoxins (paralytic shellfish poisons) were quantified by a HPLC-FLD postcolumn derivatization method. The dominant cyanobacteria during the bloom were found to be A. spiroides, Pseudanabaena sp., Cylindrospermopsis raciborskii and Microcystis aeruginosa. The mouse bioassays showed the presence of neurotoxins during both A. spiroides and C. raciborskii dominance, whereas anticholinesterase activity was only observed during periods of A. spiroides dominance. The A. spiroides strains isolated during the study also exhibited an acetylcholinesterase inhibitor. HPLC-FLD chromatograms of bloom material extracts revealed the presence of saxitoxin, neosaxitoxin and dc-saxitoxin, probably produced by C. raciborskii.  相似文献

The aim of this study was to investigate the long-lasting effects of prolonged administration of ethanol doses on oxidative processes of aged rats. We determined the activity of superox-ide dismutase (SOD), catalase (CAT), and gluta-thione peroxidase (GPx) in erythrocytes of rats 15, 20, and 24 months old treated with an average daily dose of 1.5 g/Kg of ethanol or saline administered intraperitoneally for 13 weeks and after a 2 month period of withdrawal from treatment. The activity of all three enzymes decreased significantly with aging in the controls, while no age-related changes were found among treated rats. These findings are important since they are the first to show a long-lasting toxic effect of low ethanol doses observed in association with the aging process.  相似文献

Two dung beetle subspecies of Canthon rutilans Castelnau (Coleoptera: Scarabaeidae, Scarabaeinae), C. rutilans rutilans and C. rutilans cyanescens, inhabit different environmental temperature conditions in southern Brazil. We developed a laboratory breeding experiment with 60 pairs of individuals of the two subspecies at five temperature conditions (from 15 to 35 °C) to compare the influence of temperature on the behaviour of adults and the development of larvae. The behavioural patterns of the adults in both subspecies differ according to temperature. The size of food balls was smaller and lighter in C. rutilans cyanescens. Although temperature did not influence the feeding behaviour (measured as the number of food balls made), reproductive behaviour (measured as the number of brood balls made by pair) was significantly lower at 15 °C for both subspecies. Besides that, brood balls from C. rutilans rutilans were bigger and heavier at lower temperatures. The number of offspring and the time of emergence depend on temperature too. However, the weight of the offspring and the longevity of adults depend on the subspecies and temperature treatment. These results demonstrate that the subspecies have different thermal adaptations: C. rutilans rutilans has reproductive behaviour adapted to living under colder and broader conditions than C. rutilans cyanescens.  相似文献

COVID-19 appeared in December 2019, needing efforts of science. Besides, a range of light therapies (photodynamic therapy, ultraviolet [UV], laser) has shown scientific alternatives to conventional decontamination therapies. Investigating the efficacy of light-based therapies for environment decontamination against SARS-CoV2, a PRISMA systematic review of Phototherapies against SARS-CoV or MERS-CoV species discussing changes in viral RT-PCR was done. After searching MEDLINE/PubMed, EMBASE, and Literatura Latino-Americana e do Caribe em Ciências da Saúde we have found studies about cell cultures irradiation (18), blood components irradiation (10), N95 masks decontamination (03), inanimate surface decontamination (03), aerosols decontamination (03), hospital rooms irradiation (01) with PDT, LED, and UV therapy. The best quality results showed an effective low time and dose UV irradiation for environments and inanimate surfaces without human persons as long as the devices have safety elements dependent on the surfaces, viral charge, humidity, radiant exposure. To interpersonal contamination in humans, PDT or LED therapy seems very promising and are encouraged.  相似文献
