Soil conditions may change the response of plants to inoculation with arbuscular mycorrhizal fungi (AMF). This, in turn, may influence the development of mycorrhizal associations. The effect of AM species isolated from different ecosystems was evaluated on Pueraria phaseoloides (kudzu). Experiments were conducted under controlled conditions in Leonard jars, on a substrate made of peat, rice husk, and vermiculite (1-1-1) enriched with rock phosphate plus a modified Hoagland nutrient solution. In the first experiment the initial pH of the substrate (8.0) was reduced by approximately one unit by inoculation with Kuklospora colombiana, Glomus clarum, and Glomus manihotis. The result was the same for Acaulospora longula species when 4-morpholine ethanesulphonic acid (MES) buffer was used. The final pH of the substrate was correlated with the above-ground plant biomass production suggesting that the most effective fungus regulated the pH of the substrate. This positive interaction leads to a higher concentration of P and K in plants. When the pH of the substrate was buffered with MES, in the second experiment, the efficiency of K. colombiana was reduced, even though plants inoculated with this fungus yielded more than non-inoculated plants. G. intraradices and A. longula were favoured by buffer addition. Beneficial buffer effects on this plant resulted in lower pH, better plant P and K uptake, and higher biomass production. The best plant yield was obtained at pH 7.0 in both experiments. G. clarum and G. manihotis yielded the highest biomass production. Under the proposed conditions, AMF improved P. phaseoloides growth and pH regulation. These growth conditions may be used for the evaluation and propagation of some AMF species adding MES buffer in the nutrient solution.  相似文献

Two single nucleotide polymorphisms (SNPs) in the Human Hemochromatosis (HFE) gene, C282Y and H63D, are the major variants associated to altered iron status and it is well known that these mutations are in linkage disequilibrium with certain Human Leukocyte Antigen (HLA)-A alleles. In addition, the C282Y SNP has been previously suggested to confer susceptibility to acute lymphoblastic leukemia (ALL). We have aimed to assess the diagnosis utility of these polymorphisms in a population of Spanish subjects with suspicion of hereditary iron overload and to evaluate the effect of their associations with HLA-A alleles on the susceptibility to ALL. Both the 63DD [OR = 4.31 (1.7–11.2)] and 282YY (p for trend = 0.02) genotypes were more frequently found among subjects with suspicion of iron overload than among controls. 282YY carriers displayed significantly higher transferrin saturation index (TSI) values (p < 0.001) as well as serum iron (p = 0.01) and ferritin (p = 0.01) levels. In addition, transferrin levels were lower in these subjects (p = 0.01). Likewise, patients who were carriers of the compound heterozygous diplotype (282CY/63HD) showed significantly higher TSI and serum iron and ferritin concentrations. The H63D SNP did not significantly affect the analytical parameters measured. All 282YY carriers and 69.2% of compound heterozygotes showed an altered biochemical index. The frequencies of the HFE SNPs in ALL pediatric patients were lower than those found in controls, whereas the HLA-A*24 allele was significantly overrepresented in the patients group [OR = 3.76 (1.9–7.3)]. No HFE-HLA-A associations were found to modulate the ALL risk. These results suggest that it may be useful to test for both HFE H63D and C282Y polymorphisms in patients with iron overload, as opposed to just genotyping for the C282Y SNP, which is customary in some healthcare centers. These HFE variants and their associations with HLA-A alleles were not observed to be relevant for the susceptibility to ALL in our population.  相似文献

Aims

To test predictions of ecosystem theory for changes in P cycling over primary succession, we determined soil phosphorus (P) in labile, primary mineral, organic, and occluded forms along a chronosequence of five wave cut terraces known as the “Ecological Staircase”. The Ecological Staircase terraces (T1-T5) transition naturally from fertile native coastal forests in California, USA, to diminutive pygmy vegetation over the span of?>?500,000 years of pedogenesis.

Methods

Soil P fractions were quantified to a depth of 40 cm on T1-T5 using a modified Hedley P fractionation procedure.

Results

Overall results confirmed the Walker and Syers Model of Phosphorus Transformations During Pedogenesis: total P declined from youngest (194 mg/kg P) to oldest (127 mg/kg P) sites; primary-mineral P decreased sharply from T1 to older sites; and occluded P dominated P pools at the oldest pygmy sites (T3-T5). In addition, foliar P concentrations declined markedly in the pygmy forest, and N/P of vegetation (T1: 6.03, T5: 14.4) and N/Porganic of mineral soils (T1: 6.10, T5: 25.3) increased significantly over time.

Conclusions

Results point to P as the primary limiting nutrient in the pygmy forest, exemplifying the terminal steady-state of ecosystem retrogression that underlies the persistence of this unique ecosystem.  相似文献

The present study compares fin damages in gilthead seabream (Sparus aurata) and European seabass (Dicentrarchus labrax) according to their wild, escaped or farmed origins. In addition, the potential applicability of fin condition indices (Fin Erosion Index ‘FEI’ and Fin Splitting Index ‘FSI’) as identification tools is discussed. Farmed seabream fins evidenced more erosion and splitting (FEI ± SD: 2.1 ± 0.3; FSI ± SD: 1.9 ± 0.6) than wild seabream fins (FEI: 0.8 ± 0.6; FSI: 1.2 ± 0.9), a result of farming conditions in open‐sea cages. Escaped seabream fin erosion was between that of farmed and wild seabream (FEI: 1.6 ± 0.4), which could indicate that fins in farmed fish recover over time from farming abrasions once they are in the wild. However, the fins of escaped seabream seem to be weaker than those of the wild fish, and therefore might be more susceptible to suffer other types of erosion such as splitting or nipping (FSI: 2.3 ± 0.7). No significant differences were found in seabass FEI according to their origins, although wild seabass presented significantly more split caudal fins (FSI: 3.3 ± 2.8) than the farmed seabass (FSI: 1.2 ± 1.1) and escapees (FSI: 2.5 ± 1.6). Therefore, FEI for seabream could be used as tools not only to distinguish between wild and farmed fish, but also to identify recent escapees, improving further assessments on the contribution of seabream escapees in fishery landings. However, the healing potential of damaged fins must be considered for the proper identification of escapees. Use of fin condition indices from both species could be helpful for aquaculture management, to assess fish welfare in fish farms stocks, and improve the knowledge of handling, stock densities and open‐sea cage environment conditions.  相似文献

Annona lutescens Saff. (Annonaceae) grows as a native tree in Chiapas, Mexico in Tropical Dry Forest habitat. Like most Annonaceae, it biosynthesizes benzylisoquinoline alkaloids, mostly liriodenine. To determine the influence of seasonal changes in the accumulation of liriodenine, the monthly variation of liriodenine content in roots, stems and leaves of mature and young trees was observed. These parts of young and mature A. lutescens trees were collected monthly over a 1 year period and the alkaloids were extracted; the liriodenine was quantified by high-resolution liquid chromatography. The phenological stages of the species were also assessed (leaf development, flowering and fruiting) using the Biologische Bundesanstalt, Bundessortenamt und Chemische Industrie (BBCH) scale. The analysis of both young and mature trees showed a significant increase in the liriodenine concentration occurs within roots during the dry season, which coincides with leaf fall. A significant decrease also occurred at the beginning of the rainy season (the period of leaf growth); the liriodenine content for the next rainy season did not reach the levels of the previous dry season. The climatic variation induced phenological and physiological changes in this species.  相似文献

Introduction: Unwanted platelet activation is associated with numerous diseases, mainly thrombosis-related. In this context, proteomics has emerged as a novel tool with potential for drug target discovery and to scrutinize the effects of antiplatelet drugs.

Areas covered: The present review presents the main findings of platelet proteomic studies to date in the context of drug target discovery and perspectives for the future ahead. It includes data and evidences obtained from literature searches on PubMed as well as commentaries derived from the authors’ experience and opinions.

Expert commentary: Platelet proteomics applied to drug target discovery is a young field. Recent studies have shown promising data, especially in the context of coronary artery disease. However, challenges remain such as establishing definitive guidelines for blood collection and platelet isolation, essential to guarantee data reproducibility. Recent advances in quantitative platelet proteomics should lead to novel studies with higher clinical impact in the near future.  相似文献

Plasma‐derived extracellular vesicles (EVs) have been extensively described as putative biomarkers in different diseases. Interestingly, increased levels of EVs subpopulations are well known to associate with obesity. The goal of this study is to identify EVs‐derived biomarkers in plasma from obese patients in order to predict the development of pathological events associated with obesity. Samples are obtained from 22 obese patients and their lean‐matched controls are divided into two cohorts: one for a 2D fluorescence difference gel electrophoresis (2D‐DIGE)‐based study, and the other one for a label free LC–MS/MS‐based approach. EVs are isolated following a serial ultracentrifugation protocol. Twenty‐two and 23 differentially regulated features are detected from 2D‐DIGE and label free LC–MS/MS, respectively; most of them involve in the coagulation and complement cascades. Remarkably, there is an upregulation of complement C4, complement C3, and fibrinogen in obese patients following both approaches, the latter two also validated by 2D‐western‐blotting in an independent cohort. These results correlate with a proinflammatory and prothrombotic state of those individuals. On the other hand, a downregulation of adiponectin leading to an increased risk of suffering cardiovascular diseases has been shown. The results suggest the relevance of plasma‐derived‐EVs proteins as a source of potential biomarkers for the development of atherothrombotic events in obesity.  相似文献
