In Low Arctic tundra, thermal erosion of ice‐rich permafrost soils (thermokarst) has increased in frequency since the 1980s. Retrogressive thaw slumps (RTS) are thermokarst disturbances forming large open depressions on hillslopes through soil wasting and vegetation displacement. Tall (>0.5 m) deciduous shrubs have been observed in RTS a decade after disturbance. RTS may provide conditions suitable for seedling recruitment, which may contribute to Arctic shrub expansion. We quantified in situ seedling abundance, and size and viability of soil seedbanks in greenhouse trials for two RTS chronosequences near lakes on Alaska's North Slope. We hypothesized recent RTS provide microsites for greater recruitment than mature RTS or undisturbed tundra. We also hypothesized soil seedbanks demonstrate quantity–quality trade‐offs; younger seedbanks contain smaller numbers of mostly viable seed that decrease in viability as seed accumulates over time. We found five times as many seedlings in younger RTS as in older RTS, including birch and willow, and no seedlings in undisturbed tundra. Higher seedling counts were associated with bare soil, warmer soils, higher soil available nitrogen, and less plant cover. Seedbank viability was unrelated to size. Older seedbanks were larger at one chronosequence, with no difference in percent germination. At the other chronosequence, germination was lower from older seedbanks but seedbank size was not different. Seedbank germination was positively associated with in situ seedling abundance at one RTS chronosequence, suggesting postdisturbance revegetation from seedbanks. Thermal erosion may be important for recruitment in tundra by providing bare microsites that are warmer, more nutrient‐rich, and less vegetated than in undisturbed ground. Differences between two chronosequences in seedbank size, viability, and species composition suggest disturbance interacts with local conditions to form seedbanks. RTS may act as seedling nurseries to benefit many Arctic species as climate changes, particularly those that do not produce persistent seed. 相似文献

Although peroxynitrite stimulates apoptosis in many cell types, whether peroxynitrite acts directly as an oxidant or the induction of apoptosis is because of the radicals derived from peroxynitrite decomposition remains unknown. Before undergoing apoptosis because of trophic factor deprivation, primary motor neuron cultures become immunoreactive for nitrotyrosine. We show here using tyrosine-containing peptides that free radical processes mediated by peroxynitrite decomposition products were required for triggering apoptosis in primary motor neurons and in PC12 cells cultures. The same concentrations of tyrosine-containing peptides required to prevent the nitration and apoptosis of motor neurons induced by trophic factor deprivation and of PC12 cells induced by peroxynitrite also prevented peroxynitrite-mediated nitration of motor neurons, brain homogenates, and PC12 cells. The heat shock protein 90 chaperone was nitrated in both trophic factor-deprived motor neurons and PC12 cells incubated with peroxynitrite. Tyrosine-containing peptides did not affect the induction of PC12 cell death by hydrogen peroxide. Tyrosine-containing peptides should protect by scavenging peroxynitrite-derived radicals and not by direct reactions with peroxynitrite as they neither increase the rate of peroxynitrite decomposition nor decrease the bimolecular peroxynitrite-mediated oxidation of thiols. These results reveal an important role for free radical-mediated nitration of tyrosine residues, in apoptosis induced by endogenously produced and exogenously added peroxynitrite; moreover, tyrosine-containing peptides may offer a novel strategy to neutralize the toxic effects of peroxynitrite. 相似文献

Dihydropyrimidine dehydrogenase enzyme (DPD) deficiency is a pharmacogenetic syndrome leading to severe side-effects in patients receiving therapies containing the anticancer drug 5-fluorouracil (5-FU). The aim of this population study is to evaluate gene variations in the coding region of the dihydropyrimidine dehydrogenase gene (DPYD) in the Tunisian population. One hundred and six unrelated healthy Tunisian volunteers were genotyped by denaturing HPLC (DHPLC). Twelve variants in the coding region of the DPYD were detected. Allele frequencies of DPYD*5 (A1627G), DPYD*6 (G2194A), DPYD*9A (T85C), A496G, and G1218A were 12.7%, 7.1%, 13.7%, 5.7%, and 0.5%, respectively. The DPYD alleles DPYD*2A (IVS 14+1g>1), DPYD*3 (1897 del C) and DPYD*4 (G1601A) associated with DPD deficiency were absent from the examined subjects. We describe for the first time a new intronic polymorphism IVS 6-29 g>t, found in an allelic frequency of 4.7% in the Tunisian population. Comparing our data with that obtained in Caucasian, Egyptian, Japanese and African-American populations, we found that the Tunisian population resembles Egyptian and Caucasian populations with regard to their allelic frequencies of DPYD polymorphisms. An unprecedented epidemic of chikungunya virus (CHIKV) infection recently started in countries of the Indian Ocean area, causing an acute and painful syndrome with strong fever, asthenia, skin rash, polyarthritis, and lethal cases of encephalitis. The basis for chikungunya disease and the tropism of CHIKV remain unknown. Here, we describe the replication characteristics of recent clinical CHIKV strains. Human epithelial and endothelial cells, primary fibroblasts and, to a lesser extent, monocyte-derived macrophages, were susceptible to infection and allowed viral production. In contrast, CHIKV did not replicate in lymphoid and monocytoid cell lines, primary lymphocytes and monocytes, or monocyte-derived dendritic cells. CHIKV replication was cytopathic and associated with an induction of apoptosis in infected cells. Chloroquine, bafilomycin-A1, and short hairpin RNAs against dynamin-2 inhibited viral production, indicating that viral entry occurs through pH-dependent endocytosis. CHIKV was highly sensitive to the antiviral activity of type I and II interferons. These results provide a general insight into the interaction between CHIKV and its mammalian host. 相似文献

Herein, we disclose the design of a model Ni(II) complex of glycine Schiff base possessing single‐nitrogen stereogenic center, which was successfully used for high‐performance liquid chromatography (HPLC)‐assisted assessment of its configurational stability. The major finding is that the configurational stability of the Ni(II)‐coordinated nitrogen is profoundly dependent on the reaction conditions used, in particular the solvent, and can range from inconsequential (t½ less than 5 min) to virtually completely stable (t½ 90 y).
