A flu-like disease spread among chimpanzees (Pan troglodytes schweinfurthii) of the M group at Mahale Mountains National Park, Tanzania, from June to July 2006. This epizootic or epidemic killed up to 12 chimpanzees. The obvious evidence of their deaths came from finding the bodies of three infants who had previously shown some symptoms of the disease. At least one of these infants died of pneumonia. In addition, nine chimpanzees were missing after the outbreak. These individuals were assumed to have been killed by this epizootic because most of them had contact with the infected individuals on the last days they were observed. We also found two dead bodies during this period, which were thought to be those of two missing individuals. We confirmed 23 (35.4%) of 65 individuals of the M group showed some symptoms of the disease, although most of them (20/23) did not die. More than half of them (14/23) had kin showing symptoms. Since this epizootic may have been caused by contact with humans, it will be necessary to establish and follow appropriate protocols for researchers, tourists, and park staff to observe chimpanzees, and to explore the mechanism of disease transmission from humans to chimpanzees and among chimpanzees.  相似文献

Feeding conditions, competitive regime, and female social relationships of Japanese macaques (Macaca fuscata) on Yakushima were compared between the two habitats at two different altitudes (coniferous forest, 1,000–1,200 m and coastal forest, 0–200 m). Fruit availability was higher in the coastal forest. There was no consistent difference in the frequency of agonistic interactions within a group during feeding between the two habitats. The coastal forest evoked stronger inter-group contest competition compared to the coniferous forest as evidenced by a higher inter-group encounter rate and a higher proportion of aggressive encounters to non-aggressive ones. Birth rate was higher in larger groups compared to smaller ones in the coastal forest, but did not differ in the coniferous forest. In spite of these differences in competitive regime, no variation in female social relationships was observed, such as direction and concentration on particular individuals in grooming, linearity in dominance rank, counter-attack, and support of juvenile kin during agonistic interactions. The present results indicate that the female social relationships of Japanese macaques are robust and do not change according to changes in the current environment.  相似文献

To elucidate the mechanism of the high aluminum (Al) resistance of a Myrtaceae tree, Melaleuca cajuputi Powell, we investigated the responses of root tips to Al and compared them with those of an Al-sensitive species, M. bracteata F. Muell. Roots of seedlings of both species were treated with a calcium solution (pH 4.0) containing 0 or 1 mM AlCl3. After 3 h of Al treatment, inhibition of root elongation and deposition of callose and lignin in root tips, typical signs of Al injury, were induced in M. bracteata but not in M. cajuputi, yet Al accumulation in root tips was similar in both species. These results indicate that internal Al tolerance mechanisms, not Al exclusion mechanisms, are responsible for the Al resistance of M. cajuputi. After 3 h of Al treatment, amount of Al tightly bound to root tips, Al remaining after washing with a desorbing solution, was less in M. cajuputi than in M. bracteata. In M. bracteata, 6 h of Al treatment triggered the accumulation of hydrogen peroxide (H2O2) in root tips despite the upregulation of antioxidant mechanisms, activity of peroxidase and concentration of reduced glutathione. In M. cajuputi, 6 h of Al treatment did not affect the concentration of H2O2, but decreased activity of peroxidase, and increased concentration of reduced glutathione in root tips. These results suggest that the less Al tightly bound to root tips is involved in the suppressing the H2O2 accumulation and the internal Al tolerance in M. cajuputi, and that the H2O2 accumulation or changes in cellular environment that bring about H2O2 accumulation despite the upregulation of antioxidant mechanisms results in Al-induced inhibition of root elongation in M. bracteata.  相似文献

We investigated the roles of Al-binding ligands in Al exclusion from roots and in internal Al detoxification in roots as Al resistance mechanisms in two Al-resistant Myrtaceae trees, Eucalyptus camaldulensis Dehnh. and Melaleuca cajuputi Powell. The amounts of ligands secreted from roots and contained in root tips of these species were compared with those of an Al-sensitive species, Melaleuca bracteata F. Muell., after the roots were exposed to 0 or 1 mM AlCl3 solution. Secretion of well-known ligands (citrate, oxalate, and malate) from roots under Al treatment was low in all species. However, in E. camaldulensis, the Al-binding capacity of root exudates under Al treatment was considerable and was higher than that in M. bracteata. Gel filtration chromatography revealed that a low-molecular-weight Al-binding ligand was secreted from roots in response to Al only in E. camaldulensis. On the other hand, the Al-binding capacity of cell sap in root tips under Al treatment was similar for the resistant and sensitive species. These results suggest that Al exclusion by secretion of the unknown low-molecular-weight Al-binding ligand from roots contributes to the Al resistance of E. camaldulensis, whereas M. cajuputi has developed Al-resistance mechanisms other than secretion of ligands from roots or concentration of internal ligands in root tips.  相似文献

We have developed a method to improve the transformation efficiency in genome-sequenced bacteria, using ‘Plasmid Artificial Modification’ (PAM), using the host''s own restriction system. In this method, a shuttle vector was pre-methylated in Escherichia coli cells, which carry all the putative genes encoding the DNA modification enzymes of the target microorganism, before electroporation was performed. In the case of Bifidobacterium adolescentis ATCC15703 and pKKT427 (3.9 kb E. coli-Bifidobacterium shuttle vector), introducing two Type II DNA methyltransferase genes lead to an enhancement in the transformation efficiency by five orders of magnitude. This concept was also applicable to a Type I restriction system. In the case of Lactococcus lactis IO-1, by using PAM with a putative Type I methyltransferase system, hsdMS1, the transformation efficiency was improved by a factor of seven over that without PAM.  相似文献

In plants, the effective mobilization of seed nutrient reserves is crucial during germination and for seedling establishment. The Arabidopsis H+-PPase-loss-of-function fugu5 mutants exhibit a reduced number of cells in the cotyledons. This leads to enhanced post-mitotic cell expansion, also known as compensated cell enlargement (CCE). While decreased cell numbers have been ascribed to reduced gluconeogenesis from triacylglycerol, the molecular mechanisms underlying CCE remain ill-known. Given the role of indole 3-butyric acid (IBA) in cotyledon development, and because CCE in fugu5 is specifically and completely cancelled by ech2, which shows defective IBA-to-indoleacetic acid (IAA) conversion, IBA has emerged as a potential regulator of CCE. Here, to further illuminate the regulatory role of IBA in CCE, we used a series of high-order mutants that harbored a specific defect in IBA-to-IAA conversion, IBA efflux, IAA signaling, or vacuolar type H+-ATPase (V-ATPase) activity and analyzed the genetic interaction with fugu5–1. We found that while CCE in fugu5 was promoted by IBA, defects in IBA-to-IAA conversion, IAA response, or the V-ATPase activity alone cancelled CCE. Consistently, endogenous IAA in fugu5 reached a level 2.2-fold higher than the WT in 1-week-old seedlings. Finally, the above findings were validated in icl–2, mls–2, pck1–2 and ibr10 mutants, in which CCE was triggered by low sugar contents. This provides a scenario in which following seed germination, the low-sugar-state triggers IAA synthesis, leading to CCE through the activation of the V-ATPase. These findings illustrate how fine-tuning cell and organ size regulation depend on interplays between metabolism and IAA levels in plants.  相似文献

8-硝基鸟嘌呤(8-nitroguanine, 8-NitroG)和8-羟基脱氧鸟苷(8-hydroxy-2′-deoxyguanosine, 8-OHdG)是2个氧化性DNA损伤生物标志物,而诱导型一氧化氮合酶(iNOS)在病理状态下催化细胞合成与氧化性DNA损伤有关的 氧自由基NO.本研究通过检测鼻咽癌组织中8-NitroG、8-OHdG和iNOS的免疫反应强度,初步探究鼻咽癌的发生和发展是否与氧化性DNA损伤有关以及8-NitroG、8-OHdG与iNOS表达的关系.利用多克隆抗体8-NitroG和单克隆抗体8-OHdG、iNOS,采用双色荧光免疫组织化学方法检测鼻咽癌组织中8-NitroG、8-OHdG和iNOS的免疫反应,秩和检验统计学方法分析鼻咽癌和慢性咽炎鼻咽组织之间8-NitroG、8-OHdG和iNOS免疫反应强度的差异.结果显示,19例鼻咽癌组织细胞中,8-NitroG、8-OHdG和iNOS均为强免疫反应,8-NitroG和8-OHdG阳性率100%,iNOS阳性率94.7 %,与13例慢性咽炎组织比较差异显著(P.<0.05).结果提示,鼻咽癌的发生和发展与氧化性DNA损伤有关,其原因与炎症等病理刺激下鼻咽组织高表达的iNOS催化细胞合成氧自由基NO引起的8-NitroG和8-OHdG DNA损伤密切相关.另外,8-NitroG和8-OHdG有望成为辅助鼻咽癌诊断的生物标志物.  相似文献

Caffeine (1,3,7-trimethylxanthine) and theobromine (3,7-dimethylxanthine) are purine alkaloids that are present in high concentrations in plants of some species of Camellia. However, most members of the genus Camellia contain no purine alkaloids. Tracer experiments using [8-14C]adenine and [8-14C]theobromine showed that the purine alkaloid pathway is not fully functional in leaves of purine alkaloid-free species. In five species of purine alkaloid-free Camellia plants, sufficient evidence was obtained to show the occurrence of genes that are homologous to caffeine synthase. Recombinant enzymes derived from purine alkaloid-free species showed only theobromine synthase activity. Unlike the caffeine synthase gene, these genes were expressed more strongly in mature tissue than in young tissue. The nucleotide sequence data reported here have been deposited in the GenBank database under the accession numbers AB297451 (CjCS1), AB362882 (CgCS1), AB362883 (CgCS2), AB362884 (CkCS1), AB362885 (ClCS1), and AB362886 (CcCS2).  相似文献
