Model ecosystems were grown in 12 sunlit, climate-controlled chambers to gain insight into the effects of elevated (+3°C) air temperature (Tair) on temperate grasslands. In this study, the hypothesis of delayed senescence in response to elevated Tair was tested for Rumex acetosa L. and Plantago lanceolata L. During the autumn of the first treatment year, frequent measurements were made of leaf chlorophyll a (Chl a ) fluorescence transients. Chl fluorescence images of individual leaves as well as digital colour images of these ecosystems were captured. Chl fluorescence variables, such as the maximum quantum yield of primary photochemistry (Fv/Fm), indicated a decreasing efficiency with time. Despite no treatment effect on Fv/Fm, other variables derived from the Chl fluorescence transients showed a strong trend towards a positive effect of a 3°C temperature increase on the photosynthetic performance of R. acetosa and P. lanceolata in the first year. After mid-September, the initial positive treatment effect disappeared for R. acetosa , strongly suggesting that leaf lifespan of this species was shortened by higher Tair. One possible explanation is more intense drought stress in the elevated compared to the ambient temperature treatments. Second-year measurements were possibly too limited in time to confirm this trend. These results show that temperate grassland species may take advantage of a future increase in Tair during autumn. This will ultimately depend on the species' degree of acclimation to a temperature change and on the resistance to drought stress.  相似文献

A series of copper(I) complexes of N,N′-disubstituted thioureas, [C6H5CONHCSNHR]Cu(I)Cl where R = C6H5 (1a), 2-ClC6H4 (2a), 3-ClC6H4 (3a), 4-ClC6H4 (4a), 2,3-Cl2C6H3 (5a), 2,4-Cl2C6H3 (6a), 2,5-Cl2C6H3 (7a), 2,6-Cl2C6H3 (8a), 3,4-Cl2C6H3 (9a) and 3,5-Cl2C6H3 (10a) have been synthesized. These complexes (1a–10a) have been characterized by elemental analyses, IR, 1H and 13C NMR spectroscopy, cyclic voltammetry and single crystal XRD for 1a and 8a, and for ligand 7. The X-ray crystal structures reveal that the complexes 1a and 8a are mononuclear in the solid state in which the copper atoms adopt a distorted tetrahedral geometry. In both the cases, the neutral N,N′-disubstituted thiourea ligands have been coordinated to the Cu(I) through the sulphur atom in a terminal mode. The complexes have been screened for their in vitro cytotoxic activity in human cell lines carcinomas A498 (Renal), EVSA-T (Breast), H226 (Lung), IGROV (Ovarian), M19 (Melanoma-Skin), MCF-7 (Breast) and WIDR (Colon). They show a moderate cytotoxicity against these seven human cancer cell lines comparable to that of the less active standard chemotherapeutic drugs used for comparison. They were also screened for their anti-bacterial activity and were found less active than the standard drug Imipenem.  相似文献

Background and Aims

The interaction between forest fragmentation and predicted climate change may pose a serious threat to tree populations. In small and spatially isolated forest fragments, increased homozygosity may directly affect individual tree fitness through the expression of deleterious alleles. Climate change-induced drought stress may exacerbate these detrimental genetic consequences of forest fragmentation, as the fitness response to low levels of individual heterozygosity is generally thought to be stronger under environmental stress than under optimal conditions.

Methods

To test this hypothesis, a greenhouse experiment was performed in which various transpiration and growth traits of 6-month-old seedlings of Quercus robur differing in multilocus heterozygosity (MLH) were recorded for 3 months under a well-watered and a drought stress treatment. Heterozygosity–fitness correlations (HFC) were examined by correlating the recorded traits of individual seedlings to their MLH and by studying their response to drought stress.

Key Results

Weak, but significant, effects of MLH on several fitness traits were obtained, which were stronger for transpiration variables than for the recorded growth traits. High atmospheric stress (measured as vapour pressure deficit) influenced the strength of the HFCs of the transpiration variables, whereas only a limited effect of the irrigation treatment on the HFCs was observed.

Conclusions

Under ongoing climate change, increased atmospheric stress in the future may strengthen the negative fitness responses of trees to low MLH. This indicates the necessity to maximize individual multilocus heterozygosity in forest tree breeding programmes.  相似文献

The C-type lectin-like receptor CD161 is a well-established marker for human IL17-producing T cells, which have been implicated to contribute to the development of graft-versus-host disease (GVHD) after allogeneic stem cell transplantation (allo-SCT). In this study, we analyzed CD161+ T cell recovery, their functional properties and association with GVHD occurrence in allo-SCT recipients. While CD161+CD4+ T cells steadily recovered, CD161hiCD8+ T cell numbers declined during tapering of Cyclosporine A (CsA), which can be explained by their initial growth advantage over CD161neg/lowCD8+ T cells due to ABCB1-mediated CsA efflux. Interestingly, occurrence of acute and chronic GVHD was significantly correlated with decreased levels of circulating CD161+CD4+ as well as CD161hiCD8+ T cells. In addition, these subsets from transplanted patients secreted high levels of IFNγ and IL17. Moreover, we found that CCR6 co-expression by CD161+ T cells mediated specific migration towards CCL20, which was expressed in GVHD biopsies. Finally, we demonstrated that CCR6+ T cells indeed were present in these CCL20+ GVHD-affected tissues. In conclusion, we showed that functional CD161+CCR6+ co-expressing T cells disappear from the circulation and home to GVHD-affected tissue sites. These findings support the hypothesis that CCR6+CD161-expressing T cells may be involved in the immune pathology of GVHD following their CCL20-dependent recruitment into affected tissues.  相似文献

The mutagenicity of several test compounds was verified by the Salmonella/microsome mutagenicity test (Ames test), using both human liver and rat liver (untreated or pretreated with Aroclor 1254) S9 under identical experimental conditions. Aflatoxin B1, 3-methylcholanthrene, and cigarette-smoke condensate were less mutagenic in the presence of human-liver S9 than in the presence of rat-liver S9 (particularly after treatment with Aroclor 1254). The opposite was observed with 2-aminonanthracene and to a lesser degree with 2-aminofluorene; correlation studies indicate that the two compounds were activated by the same or by very similar enzymes, probably cytochrome P-450s. These results clearly indicate that human-liver S9, as an activating system, behaves differently than rat-liver S9; therefore, it may constitute a useful, additional tool for the study of mutagenicity and probably, carcinogenicity in man.  相似文献
