The Sooty Shearwater Ardenna grisea, an abundant but declining petrel, is one of many seabird species that construct breeding burrows, presumably because these confer protection from predators and the elements. Little is known about the causes of variation in Sooty Shearwater burrow architecture, which can differ markedly both within and between breeding sites. We hypothesize that burrow architecture varies in response to habitat type and competition for space. To address these hypotheses, we recorded Sooty Shearwater burrow dimensions on Kidney Island, the largest Sooty Shearwater colony in the Falkland Islands, South Atlantic, and modelled these as functions of burrow density (a proxy for competition) and habitat indices. Our models suggest that Sooty Shearwaters burrow further underground in response to competition for breeding space, and that soil underlying dense tussac grass Poa flabellata is more easily excavated than other substrates, indicating how vegetation restoration could aid the conservation of this species. 相似文献

Isolates of the rice blast fungus Magnaporthe grisea that carry the gene encoding Avirulence Conferring Enzyme1 (ACE1) are specifically recognized by rice (Oryza sativa) cultivars carrying the resistance gene Pi33. This recognition enables resistant plants to activate a defense response. ACE1 was isolated by map-based cloning and encodes a putative hybrid between a polyketide synthase and a nonribosomal peptide synthetase, enzymes involved in microbial secondary metabolism. ACE1 is expressed exclusively during fungal penetration of host leaves, the time point at which plant defense reactions are triggered. Ace1 appears to be localized in the cytoplasm of the appressorium. Mutation of the putative catalytic site of the beta-ketoacyl synthase domain of Ace1 abolishes recognition of the fungus by resistant rice. This suggests that Ace1 biosynthetic activity is required for avirulence. Our results are consistent with the hypothesis that the fungal signal recognized by resistant rice plants is the secondary metabolite whose synthesis depends on Ace1. 相似文献

Metal toxicity for living organisms involves oxidative and/or genotoxic mechanisms. Plant protection against metal toxicity occurs, at least in part, through control of root metal uptake and of long distance metal transport. Inside cells, proteins such as ferritins and metallothioneins, and glutathion-derived peptides named phytochelatins, participate in excess metal storage and detoxification. Low molecular weight organic molecules, mainly organic acids and amino acids and their derivatives, also play an important role in plant metal homeostasis. When these systems are overloaded, oxidative stress defense mechanisms are activated. Molecular and cellular knowledge of these processes will be necessary to improve plant metal resistance. Occurrence of naturally tolerant plants which hyperaccumulate metals provides helpful tools for this research. 相似文献

Effects of the calcium channel agonist, BAY K 8644, on electrical activity in mouse pancreatic B-cells. 总被引:4,自引:0,他引:4
We studied the effects of the dihydropyridine derivative BAY K 8644 on the membrane potential of B-cells in mouse pancreatic islets. BAY K 8644, in a dose-dependent manner, decreased the spike frequency but increased the duration of the spikes elicited by glucose with or without quinine or tetraethylammonium (TEA). These effects were antagonized by cobalt and nifedipine but not by tetrodotoxin. The interval between spikes was proportionate to the duration of the spikes and the ratio of the interval to the spike duration was constant at all concentrations of BAY K 8644 tested. Peak inward current, estimated from the derivative of the action potential recorded in the presence of TEA, was increased by BAY K 8644 and decreased by nifedipine. BAY K 8644 elicited spike activity when the membrane was moderately depolarized by either 5.6 mM glucose or 15 mM K+, but did not change the membrane potential of the resting hyperpolarized B-cell. These results suggest that BAY K 8644 acts on the open Ca2+-channels. The threshold occurs at a membrane potential of -50 mV. Also, the modifications of the shape of the spikes appear to reflect specific changes in Ca2+ entry. We propose the existence of a Ca2+-induced Ca2+-channel inactivation process in the pancreatic B-cell. 相似文献

Tenuazonic acid (TA) is a phytotoxin produced by a fungal pathogen of rice, Pyricularia oryzae. We have synthesized and characterized the metal complexes of TA with copper (II), iron (III), nickel (II), and magnesium (II). The stoichiometry of the complexes determined by microanalysis and mass spectroscopy (D/CI) are Cu(II)TA2, Fe(III)TA3, Ni(II)TA2, and Mg(TA)2. Voltammograms of Fe(III)TA3, and Cu(II)TA2 in methanolic solutions confirmed this stoichiometry. Ni(II)TA2 paramagnetism and visible absorption data suggest an octahedral geometry. Fe(III)TA3 showed a characteristic visible absorption at 450 nm. Addition of Fe(III)Cl3 and Mg(II)Cl2 did not reverse the toxicity of NaTA to rice and bacterial cells, showing that this toxicity is not due to the privation of the cells of these metals essential for cell growth. 相似文献

Secondary effects of the entomopathogenic hyphomycete Beauveria bassiana (Bals.) Vuill. were investigated on adults of Leptinotarsa decemlineata Say (Coleoptera:Chrysomelidae) treated as newly moulted 4th-instar larvae. Fecundity and egg hatching were monitored in surviving females mated with surviving males. When reared at 22°C, adult survivors showed a lessening of their reproductive potential during their whole life. According to the fungal inoculum dosage, the reductions of the total number of eggs laid per female and the mean number of eggs per egg mass ranged from 20% to 56% and from 18% to 46%, respectively. In contrast, at 25°C, the fecundity of survivors was not affected by the fungal infection. This variability of the secondary effects of B. bassiana according to temperature conditions might be related to the lower infection level of treated larvae at 25°C. Moreover, at both temperatures, eggs laid by females surviving fungal infection as larvae were as fertile as eggs laid by control insects. It is concluded that secondary effects of B. bassiana on the fecundity of the Colorado Potato Beetle are temperature-dependent and that they could become negligible at high temperatures under field conditions.
Résumé Les effets secondaires de l'hyphomycète entomopathogène, Beauveria bassiana (Bals.) Vuill. sur la fécondité et la fertilité des insectes survivant à l'infection fongique ont été étudiés chez les adultes du doryphore Leptinotarsa decemlineata Say (Coleoptera:Chrysomelidae) issus de larves contaminées en début de 4ème stade. Les insectes maintenus à 22°C ont présenté une réduction de leur potentiel reproductif. Ainsi, suivant la dose d'inoculum fongique, le nombre total d'oeufs pondus par femelle et le nombre moyen d'oeufs par opplaque ont baissé de 20% à 56% et de 18% à 46%, respectivement. En revanche, à 25°C la fécondité des adultes survivants n'a pas été affectée par la maladie. Cette variabilité en fonction des conditions thermiques peut être liée à la diminution de l'effet-dose du champignon sur les larves à 25°C par rapport à 22°C. Par ailleurs, quelles que soient les conditions thermiques (22°C ou 25°C) on ne constate aucun changement significatif de la fertilité des oeufs pondus par les femelles survivantes. Les auteurs concluent que les effets secondaires de B. bassiana sur la fécondité du doryphore dépendent des conditions thermiques et qu'ils pourraient devenir négligeables à des températures élevées en plein champ.相似文献

Many successful studies on genome editing in plants have been reported and one of the popular genome editing technology used in plants is Zinc Finger Nucleases (ZFN), which are chimeric proteins composed of synthetic zinc finger-based DNA binding domain and a DNA cleavage domain. The objective of this research was to utilize ZFNs to induce a double-stranded break in SSIVa, a soluble starch synthase involved in starch biosynthesis pathway, leading to the regulation of the SSIVa expression. The isoform SSIVa is not yet well studied, thus, by modifying the endogenous loci in SSIVa, we can explore on the specific roles of this gene in starch biosynthesis and other possible functions it might play. In this study, we used ZFN-mediated targeted gene disruption in the coding sequence of the SSIVa rice gene in an effort to elucidate the functional role of the gene. Generation of transgenic plants carrying premature stop codons and substitution events, revealed no SSIVa mRNA expression, low starch contents and dwarf phenotypes. Remarkably, based on our analysis SSIVa gene disruption had no effect on other starch synthesis related genes as their expression remained at wild type levels. During cell cultivation processes for the production of biopharmaceuticals, good process performance and good product quality can be ensured by online monitoring of critical process parameters (e.g. temperature, pH, or dissolved oxygen). These data can be used in real‐time for process control, as suggested by the process analytical technology (PAT) initiative. Today, solutions for real‐time monitoring of parameters such as concentrations of cells, main nutrients, and metabolism by‐products are developing, but applications of these more complex tools in industrial settings are still limited. Here, we evaluated the use of dielectric spectroscopy (DS) and near‐infrared spectroscopy (NIRS) as PAT tools for a perfusion PER.C6® cultivation process. We showed that DS enabled predictions of viable cell density from the cultivation vessel, with a root mean square error of prediction (RMSEP) of 4.4% of the calibration range. Additionally, predictions of glucose and lactate concentrations from the cultivation vessel (RMSEP of 10 and 14%, respectively) and from the perfusion stream (RMSEP of 12 and 10%, respectively) were achieved with NIRS. We also showed that the perfusion stream offers great opportunities for noninvasive, yet frequent process monitoring. Accurate online monitoring of critical process parameters with PAT tools is the essential first step toward increased control of process output. 相似文献
