The Work Productivity and Activity Impairment (WPAI) questionnaire is a well validated instrument to measure impairments in work and activities. However, its validation among patients with rheumatoid arthritis (RA) has not been well established. The entry of retroviruses into cells depends on receptor recognition by the viral envelope surface subunit SU followed by membrane fusion, which is thought to be mediated by a fusion peptide located at the amino terminus of the envelope transmembrane subunit TM. Several fusion determinants have been previously identified in murine leukemia virus (MLV) envelopes, but their functional interrelationships as well as the processes involved in fusion activation upon retroviral receptor recognition remain unelucidated. Despite both structural and functional similarities of their envelope glycoproteins, ecotropic and amphotropic MLVs display two different postbinding properties: (i) while amphotropic MLVs fuse the cells at neutral pH, penetration of ecotropic MLVs is relatively acid pH dependent and (ii) ecotropic envelopes are more efficient than amphotropic envelopes in inducing cell-to-cell fusion and syncytium formation. By exploiting the latter characteristic in the analysis of chimeras of ecotropic and amphotropic MLV envelopes, we show here that substitution of the ecotropic MLV proline-rich region (PRR), located in the SU between the amino-terminal receptor binding domain and the TM-interacting SU carboxy-terminal domains, is sufficient to revert the amphotropic low-fusogenic phenotype into a high-fusogenic one. Furthermore, we have identified potential β-turns in the PRR that control the stability of SU-TM associations as well as the thresholds required to trigger either cell-to-cell or virus-to-cell fusion. These data, demonstrating that the PRR functions as a signal which induces envelope conformational changes leading to fusion, have enabled us to derive envelopes which can infect cells harboring low levels of available amphotropic receptors.  相似文献

Significant interspecific differences in avian vision occur, even in congeneric species, and these have been correlated with differences in the perceptual challenges associated with foraging. Although diurnal raptors are assumed to be mainly visually guided in their foraging, they differ markedly in their foraging tactics and this may result in different visual demands. Among the Falconidae (Falconiformes), most falcons forage mainly on the wing for highly mobile prey, whereas caracaras forage on the ground for carrion and insects. We assessed whether Saker Falcon Falco cherrug and Southern Caracara Caracara plancus differ in their visual abilities by determining the visual fields and foveal characteristics of both species. Using an ophthalmoscopic reflex technique, we found a higher degree of binocular overlap in the caracaras than in the falcons. The high binocular overlap (47°) of the Southern Caracara may facilitate object manipulation (e.g. moving rocks) when foraging. We used an ultra‐high resolution spectral‐domain optical coherence tomography to determine foveal characteristics. We found two foveas (depressions in the retina where high visual resolution is expected) in the falcons (one central and one temporal) but only a central fovea in the caracaras. The presence of a shallower temporal fovea in Saker Falcons may help to fixate visually upon a highly mobile prey item during pursuit. We conclude that these differences in visual field configurations and foveal characteristics reflect different foraging demands, suggesting that the extraction of visual information is finely tuned to the demands of their foraging tactics.  相似文献

Olfactory signals constitute an important mechanism in interspecific interactions, but little is known regarding their role in communication between predator species. We analyzed the behavioral responses of a mesopredator, the red fox (Vulpes vulpes), to an olfactory cue (scat) of an apex predator, the lynx (Lynx lynx) in Bia?owie?a Primeval Forest, Poland, using video camera traps. Red fox visited sites with scats more often than expected and the duration of their visits was longer at scat sites than at control sites (no scat added). Vigilant behavior, sniffing and scent marking (including over-marking) occurred more often at scat sites compared to control sites, where foxes mainly passed by. Vigilance was most pronounced during the first days of the recordings. Red fox behavior was also influenced by foxes previously visiting scat sites. They sniffed and scent marked (multiple over-marking) more frequently when the lynx scat had been over-marked previously by red fox. Fox visits to lynx scats may be seen as a trade-off between obtaining information on a potential food source (prey killed by lynx) and the potential risk of predation by an apex predator.  相似文献

In an effort to identify HDAC isoform selective inhibitors, we designed and synthesized novel, chiral 3,4-dihydroquinoxalin-2(1H)-one and piperazine-2,5-dione aryl hydroxamates showing selectivity (up to 40-fold) for human HDAC6 over other class I/IIa HDACs. The observed selectivity and potency (IC50 values 10–200 nM against HDAC6) is markedly dependent on the absolute configuration of the chiral moiety, and suggests new possibilities for use of chiral compounds in selective HDAC isoform inhibition.  相似文献
