In an outdoor mesocosm experiment of 80 weeks, the effect of nitrogen and phosphorus addition was tested on growth of Typha domingensis Presl. rhizomes in a matrix of Schoenoplectus americanus (Pers.) Volkart ex Schinz and Keller, under loading rates of 0.23 gm−2 d−1 of nitrogen, 0.17 gm−2 d−1 of P, both nutrient together and control conditions, to assess the potential expansion of T. domingensis in response to nutrient inputs. 相似文献

The objective of this study was to evaluate the efficacy of two commercially available modified live virus vaccines for preventing the reproductive and early postnatal consequences of infecting (challenging) pregnant gilts with virulent porcine reproductive and respiratory syndrome virus (PRRSV). For this purpose 21 crossbred gilts were allocated to one or another of four groups (Groups A-D). Group A comprised four gilts neither vaccinated nor challenged; Group B comprised five gilts that were challenged but not vaccinated; Group C comprised seven gilts that were vaccinated (AmervacPRRS) and challenged; Group D comprised five gilts that were vaccinated (Pyrsvac-183) and challenged. Vaccination was 24 days before conception, and challenge was at 90 days of gestation. Both vaccine viruses and the challenge virus were European strains but differed in part from one another on the basis of their genetic (nucleotide) sequence. After challenge PRRSV was isolated from five (100%), four (57%), and two (40%) of the gilts of Groups B, C and D, respectively. Although vaccination failed to prevent a detectable viremia in all of the gilts of Groups C and D after they were challenged (or congenital infection of some of their pigs), it did provide a statistically significant level of protection in regard to the incidence of congenital infection, reproductive performance, and pig health and viability. Namely, for Groups C and D the numbers of liveborn pigs/litter and healthy pigs/litter throughout the early postnatal period were similar to those of Group A (nonvaccinated and nonchallenged) and far exceeded those of Group B (nonvaccinated and challenged). 相似文献

Herbivore leaf consumption of various mangrove species in relation to environmental factors and leaf hardness were studied in the Dagua river estuary, Colombia. Leaf consumption and damage were assessed by measuring the percentage of area attacked by herbivores, distinguishing between consumption and damage. The species that suffered the highest consumption, such as Avicennia germinans (Avicenniaceae) and Laguncularia racemosa (Combretaceae), had softer leaves and less herbivore species when compared with Rhizophora spp. (Rhizophoraceae) and Pelliciera rhizophorae (Theaceae). The abundance and diversity of leaf grazing and its variability among mangrove species in the Dagua River estuary, show the importance of the trophic dynamics of live vegetable matter, in spite of their relatively low contribution to removing organic matter. 相似文献

Every organism on Earth must cope with a multitude of species interactions both directly and indirectly throughout its life cycle. However, how selection from multiple species occupying different trophic levels affects diffuse mutualisms has received little attention. As a result, how a given species amalgamates the combined effects of selection from multiple mutualists and antagonists to enhance its own fitness remains little understood. We investigated how multispecies interactions (frugivorous birds, ants, fruit flies and parasitoid wasps) generate selection on fruit traits in a seed dispersal mutualism. We used structural equation models to assess whether seed dispersers (frugivorous birds and ants) exerted phenotypic selection on fruit and seed traits in the spiny hackberry (Celtis ehrenbergiana), a fleshy‐fruited tree, and how these selection regimes were influenced by fruit fly infestation and wasp parasitoidism levels. Birds exerted negative correlational selection on the combination of fruit crop size and mean seed weight, favouring either large crops with small seeds or small crops with large seeds. Parasitoids selected plants with higher fruit fly infestation levels, and fruit flies exerted positive directional selection on fruit size, which was positively correlated with seed weight. Therefore, higher parasitoidism indirectly correlated with higher plant fitness through increased bird fruit removal. In addition, ants exerted negative directional selection on mean seed weight. Our results show that strong selection on phenotypic traits may still arise in perceived diffuse species interactions. Overall, we emphasize the need to consider diverse direct and indirect partners to achieve a better understanding of the mechanisms driving phenotypic trait evolution in multispecies interactions. 相似文献

The Tn antigen (GalNAcα-O-Ser/Thr) is a well-established tumor-associated marker which represents a good target for the design of anti-tumor vaccines. Several studies have established that the binding of some anti-Tn antibodies could be affected by the density of Tn determinant or/and by the amino acid residues neighboring O-glycosylation sites. In the present study, using synthetic Tn-based vaccines, we have generated a panel of anti-Tn monoclonal antibodies. Analysis of their binding to various synthetic glycopeptides, modifying the amino acid carrier of the GalNAc(*) (Ser* vs Thr*), showed subtle differences in their fine specificities. We found that the recognition of these glycopeptides by some of these MAbs was strongly affected by the Tn backbone, such as a S*S*S* specific MAb (15G9) which failed to recognize a S*T*T* or a T*T*T* structure. Different binding patterns of these antibodies were also observed in FACS and Western blot analysis using three human cancer cell lines (MCF-7, LS174T and Jurkat). Importantly, an immunohistochemical analysis of human tumors (72 breast cancer and 44 colon cancer) showed the existence of different recognition profiles among the five antibodies evaluated, demonstrating that the aglyconic part of the Tn structure (Ser vs Thr) plays a key role in the anti-Tn specificity for breast and colon cancer detection. This new structural feature of the Tn antigen could be of important clinical value, notably due to the increasing interest of this antigen in anticancer vaccine design as well as for the development of anti-Tn antibodies for in vivo diagnostic and therapeutic strategies. 相似文献

The eggshell oospecies from India and Argentina are compared and reviewed in detail. These eggshells resemble each other in having a nodular outer surface ornamentation and clearly arched growth lines of the shell units. Microstructurally, the eggshell oospecies belonging to the oofamily Megaloolithidae shows fan-like shell units, which are sharply separated from each other throughout the thickness of the eggshell and can be traced up to the surface of the eggshell. Comparisons between four oospecies from India and Argentina reveal three groupings, which show similarities between megaloolithids of both countries: (1) Megaloolithus jabalpurensis, M. matleyi and M. patagonicus; (2) M. cylindricus, M. rahioliensis and Tipo 1d; and (3) M.megadermus and Tipo 1e. The other two types of eggshell oospecies from India and Argentina show partially fused external nodes and shell units. As a result, growth lines enter into the adjacent shell units with a marked concavity. A new oogenus Fusioolithus have been erected due to fusion between shell units and tubospherulitic morphotype, which include two new oospecies F. baghensis and F. berthei. Till date, morphostructurally, a total of 15 eggshell oospecies belonging to different oofamilies have been recorded from India and seven oospecies from Argentina. 相似文献

We have previously shown that plasma membrane calcium ATPase (PMCA) pump activity is affected by the membrane protein concentration (Vanagas et al., Biochim Biophys Acta 1768:1641–1644, 2007). The results of this study provided evidence for the involvement of the actin cytoskeleton. In this study, we explored the relationship between the polymerization state of actin and its effects on purified PMCA activity. Our results show that PMCA associates with the actin cytoskeleton and this interaction causes a modulation of the catalytic activity involving the phosphorylated intermediate of the pump. The state of actin polymerization determines whether it acts as an activator or an inhibitor of the pump: G-actin and/or short oligomers activate the pump, while F-actin inhibits it. The effects of actin on PMCA are the consequence of direct interaction as demonstrated by immunoblotting and cosedimentation experiments. Taken together, these findings suggest that interactions with actin play a dynamic role in the regulation of PMCA-mediated Ca2+ extrusion through the membrane. Our results provide further evidence of the activation–inhibition phenomenon as a property of many cytoskeleton-associated membrane proteins where the cytoskeleton is no longer restricted to a mechanical function but is dynamically involved in modulating the activity of integral proteins with which it interacts. 相似文献

The exposure of the plasma membrane calcium pump (PMCA) to the surrounding phospholipids was assessed by measuring the incorporation of the photoactivatable phosphatidylcholine analog [(125)I]TID-PC/16 to the protein. In the presence of Ca(2+) both calmodulin (CaM) and phosphatidic acid (PA) greatly decreased the incorporation of [(125)I]TID-PC/16 to PMCA. Proteolysis of PMCA with V8 protease results in three main fragments: N, which includes transmembrane segments M1 and M2; M, which includes M3 and M4; and C, which includes M5 to M10. CaM decreased the level of incorporation of [(125)I]TID-PC/16 to fragments M and C, whereas phosphatidic acid decreased the incorporation of [(125)I]TID-PC/16 to fragments N and M. This suggests that the conformational changes induced by binding of CaM or PA extend to the adjacent transmembrane domains. Interestingly, this result also denotes differences between the active conformations produced by CaM and PA. To verify this point, we measured resonance energy transfer between PMCA labeled with eosin isothiocyanate at the ATP-binding site and the phospholipid RhoPE included in PMCA micelles. CaM decreased the efficiency of the energy transfer between these two probes, whereas PA did not. This result indicates that activation by CaM increases the distance between the ATP-binding site and the membrane, but PA does not affect this distance. Our results disclose main differences between PMCA conformations induced by CaM or PA and show that those differences involve transmembrane regions. 相似文献
