By assessing the development of Y-linked autoimmune acceleration (Yaa) gene-induced systemic lupus erythematosus in C57BL/6 (B6) x (New Zealand Black (NZB) x B6.Yaa)F(1) backcross male mice, we mapped three major susceptibility loci derived from the NZB strain. These three quantitative trait loci (QTL) on NZB chromosomes 1, 7, and 13 differentially regulated three different autoimmune traits: anti-nuclear autoantibody production, gp70-anti-gp70 immune complex (gp70 IC) formation, and glomerulonephritis. Contributions to the disease traits were further confirmed by generating and analyzing three different B6.Yaa congenic mice, each carrying one individual NZB QTL. The chromosome 1 locus that overlapped with the previously identified Nba2 (NZB autoimmunity 2) locus regulated all three traits. A newly identified chromosome 7 locus, designated Nba5, selectively promoted anti-gp70 autoantibody production, hence the formation of gp70 IC and glomerulonephritis. B6.Yaa mice bearing the NZB chromosome 13 locus displayed increased serum gp70 production, but not gp70 IC formation and glomerulonephritis. This locus, called Sgp3 (serum gp70 production 3), selectively regulated the production of serum gp70, thereby contributing to the formation of nephritogenic gp70 IC and glomerulonephritis, in combination with Nba2 and Nba5 in NZB mice. Among these three loci, a major role of Nba2 was demonstrated, because B6.Yaa Nba2 congenic male mice developed the most severe disease. Finally, our analysis revealed the presence in B6 mice of an H2-linked QTL, which regulated autoantibody production. This locus had no apparent individual effect, but most likely modulated disease severity through interaction with NZB-derived susceptibility loci.  相似文献

Antibodies named TcCRA “Trypanosoma cruzi Cross Reactive Antibodies” were detected in 47% of blood donors from French population unexposed to the parasite. In order to evaluate the passive or active transmissibility of TcCRA and further characterize its role and etiology, we have conducted a study in a cohort of 47 patients who underwent allogeneic Hematopoietic Stem Cell Transplantations (allo-HSCT). Donors and recipients were tested for TcCRA prior to transplantation. Recipients were further tested during follow-up after transplantation. Demographical, clinical and biological data were collected. Our primary end-point was to assess the risk of TcCRA acquisition after transplantation. During this initial analysis, we observed no seroconversion in patients receiving cells from TcCRA negative donors (n = 23) but detected seroconversion in 4 out of 24 patients who received hematopoietic stem cells from positive donors. Here, we are discussing possible scenarios to explain TcCRA-immune status in recipient after transplantation.  相似文献

Variable Selection for Clustering with Gaussian Mixture Models   总被引:3,自引:0,他引:3  
Summary .  This article is concerned with variable selection for cluster analysis. The problem is regarded as a model selection problem in the model-based cluster analysis context. A model generalizing the model of Raftery and Dean (2006,  Journal of the American Statistical Association   101, 168–178) is proposed to specify the role of each variable. This model does not need any prior assumptions about the linear link between the selected and discarded variables. Models are compared with Bayesian information criterion. Variable role is obtained through an algorithm embedding two backward stepwise algorithms for variable selection for clustering and linear regression. The model identifiability is established and the consistency of the resulting criterion is proved under regularity conditions. Numerical experiments on simulated datasets and a genomic application highlight the interest of the procedure.  相似文献

The need for cold, well-oxygenated waters significantly reduces the habitat available for lake trout (Salvelinus namaycush) during stratification of small temperate lakes. We examined the spatial and pelagic distribution of lake trout over two consecutive summers and winters and tested whether winter increased habitat availability and access to littoral regions in a boreal shield lake in which pelagic prey fish are absent. In winter, lake trout had a narrowly defined pelagic distribution that was skewed to the upper 3 m of the water column and spatially situated in the central region of the lake. Individual core areas of use (50% Kernel utilization distributions) in winter were much reduced (75%) and spatially non-overlapping compared to summer areas, but activity levels were similar between seasons. Winter habitat selection is in contrast to observations from the stratified season, when lake trout were consistently located in much deeper waters (>6 m) and widely distributed throughout the lake. Winter distribution of lake trout appeared to be strongly influenced by ambient light levels; snow depth and day length accounted for up to 69% of the variation in daily median fish depth. More restricted habitat use during winter than summer was in contrast to our original prediction and illustrates that a different suite of factors influence lake trout distribution between these seasons.  相似文献

The Pacific oyster Crassostrea gigas is a successive not systematic protandric hermaphrodite. Searching for an ortholog to Dmrt1, a conserved sex determinism factor, we have identified the first complete cDNA of a DM factor in Lophotrochozoa which we have called Cg-DMl (Crassostrea gigas DMRT-like). It is 359aa long, with the DM domain common to all the family factors, and one DMA domain specific to members such as Dmrt4 and Dmrt5. Its gene presents one intron of 598 bp. Real time PCR and in situ hybridization have shown that Cg-DMl was expressed in both sexes, with a significantly higher expression in male than in female gonads at the end of the adult gametogenetic cycle and that a significant peak of expression was observed in spat between 1 and 2 months of age. These results suggest that Cg-DMl may be involved in the development of the gonad and may constitute preliminary clues for future work in order to better understand DM protein evolution.  相似文献

The diversification of species and their interactions during the course of evolution has produced ecological networks with a complex topology. This topology influences the current functioning of ecosystems. It is therefore important to investigate whether the species introduced recently by human activities have merged seamlessly into recipient ecological networks by developing interactions quantitatively and qualitatively similar to those of native species, or whether their establishment has altered the topology of the networks. We tackled this issue in the case of a well resolved interaction network between 51 forest tree taxa and 154 pathogenic fungal species. We found that alien and native species with similar phylogenetic histories and life-history strategies had similar types and numbers of interactions. Our results also suggest that the clustered architecture of the network has not been altered by the integration of alien species. It therefore seems that a few centuries have been sufficient for the network to assimilate the newly introduced species. This rapid integration was unexpected for a plant-pathogen network, because selection acts continually on plants, favouring the emergence of defences against new pathogens and impeding the development of new interactions. However, it was recently shown that perturbation of the structure of ecological networks might be overlooked if species interactions are not quantified. The tree-parasitic fungus network considered in this study is binary. We might therefore end up with different results by using quantitative data.  相似文献
