Prothrombin Barcelona is a varient of human prothrombin. Its activation by factor Xa is characterized by two abnormal features. The first one is the absence of one of the two Xa-catalyzed cleavages, and the second one the generation of a thrombin-like activity responsible for the thrombin-catalyzed cleavages on the prothrombin molecule and for the activity toward small substrates but without clotting activity.The molecular species exhibiting the thrombin-like activity have been characterized Diisopropyl phosphofluoridate incorporation experiments show that upon activation, two high molecular weight intermediates bearing an active site are formed: one has the molecular weight of prothrombin, the other of prethrombin 1. In the presence of hirudin, only the first intermediate is formed due to a single cleavage by factor Xa which is therefore responsible for the unmasking of the active site. 相似文献

Lipid patterns of embryonal carcinoma cell lines and their derivatives: Changes with differentiation
The lipid composition of several teratocarcinoma cell lines has been examined by biochemical and immunological methods in order to identify properties that might be correlated with the state of cell differentiation. The data indicate qualitative and quantitative changes in the phospholipid, cholesterol, and glycolipid composition. In particular, the ratios of cholesterol/phospholipid and of sphingomyelin/phosphatidylcholine are higher in differentiated cells. Gangliosides with short glycosidic chains (GM3 and GD3) are characteristic of undifferentiated, multipotent, embryonal carcinoma cell lines. More complex gangliosides (GM1 and GD1a) appear early during the course of differentiation. Each differentiated cell line presents a unique ganglioside map. Results are tentatively correlated with a stabilization of the membrane bilayer in differentiated cell lines, whereas a more fluid state of the membrane in embryonal carcinoma cell lines would allow maximal flexibility. Subtle differences in ganglioside composition among embryonal carcinoma cell lines are discussed in relation with their potentialities, and their developmental age. 相似文献

A new transposable element has been isolated from an unstable niaD mutant of the fungus Fusarium oxysporum. This element, called impala, is 1280 nucleotides long and has inverted repeats of 27 bp. Impala inserts into a TA site and leaves behind a “footprint” when it excises. The inserted element, impala-160, is cis-active, but is probably trans-defective owing to several stop codons and frameshifts. Similarities exist between the inverted repeats of impala and those of transposons belonging to the widely dispersed mariner and Tc1 families. Moreover, translation of the open reading frame revealed three regions showing high similarities with Tc1 from Caenorhabditis elegans and with the mariner element of Drosophila mauritiana. The overall comparison shows that impala occupies an intermediate position between the mariner and Tcl-like elements, suggesting that all these elements belong to the same superfamily. The degree of relatedness observed between these elements, described in different kingdoms, raises the question of their origin and evolution. 相似文献
