Stability of Lanatoside C content in thein vitro propagatedDigitalis lanata clones

In vitro culture was established from shoot tips ofDigitalis lanata cotyledonous plants. The propagated plant material was rooted, transplanted into soil and grown under field conditions. Lanatoside C content was determined in a total of 20 clones and statistically evaluated by means of variance analysis of unequal-sized samples.In vitro clonal propagation ofD. lanata was found not to affect lanatoside C content. Drug level was dependent on a plant genotype.     

Mesotocin and vasotocin in the brain of the lizard Gekko gecko

Summary The distribution of mesotocin and vasotocin was studied in the brain of the lizard Gekko gecko with antisera specific for either peptide. Both mesotocinergic and vasotocinergic perikarya are found in the paraventricular and supraoptic nuclei of the hypothalamus, whereas vasotocinergic neurons are exclusively present in the bed nucleus of the stria terminalis and in a cell group of the rhombencephalon. The distributional pattern of the mesotocinergic fibers corresponds closely to that of the vasotocinergic fibers. However, throughout the entire brain the mesotocinergic innervation is less dense than the vasotocinergic innervation. Molecular cloning of a pair of human pepsinogen A genes which differ by a Glu→Lys mutation in the activation peptide

Summary Three human cosmid clones containing pepsinogen A (PGA) encoding sequences were isolated from a genomic bank derived from a single individual. One cosmid contains two PGA genes in tandem in a head-to-tail orientation, while the other two cosmids each contain a single PGA gene. The three cosmids were characterized by restriction mapping and sequence analysis (exons 1 and 2 and flanking regions). As judged from these data, three of the four PGA genes isolated appear to be nearly identical, but one of the tandem genes is clearly different from the other genes. The first exon of all four genes codes for the same amino acid sequence. However, in the second exon of one of the tandem genes we found a nucleotide substitution giving rise to a GluLys substitution of the 43rd amino acid residue of the activation peptide, leading to a charge difference of the corresponding isozymogens. The presence of two distinct PGA genes in the isolated gene pair conclusively proves the multigene structure of the PGA system. These genes might be responsible for at least part of the electrophoretic polymorphism at the protein level.     

Interaction of allosteric effectors (ATP,CO2, H+) modulating oxygen affinity of the hemoglobin in the carp,Cyprinus carpio,in vitro

Summary The interaction of allosteric effectors (CO₂, ATP, H⁺) with respect to the oxygen affinity of carp hemoglobin was analyzed by determining oxygen binding curves spectrophotometrically in dilute solutions of stripped hemoglobin at 20°C. The pH range studied was 6.8–8.2.P _CO2 was 0, 10 and 70 mmHg (0, 1.33 and 9.3 kPa). ATP/Hb₄ was 0, 8 and 24. In the presence of either CO₂ or ATP, the effects of the cofactors onP ₅₀ were as expected over the whole pH range. In contrast to other published data, each cofactor also had a significant effect onP ₅₀ in the presence of the other cofactor. Evidence was obtained that oxylabile carbamate is formed by carp hemoglobin and that the formation of carbamate persists at a lower level in the presence of ATP. The results support the view that the binding of ATP to carp hemoglobin requires only one terminal amino group, leaving the other N-terminal of the -chain free to react with CO₂.     

Neuromedin U-immunoreactivity in the nervous system of the small intestine of the pig and its coexistence with substance P and CGRP

Summary In the small intestine of the pig, neuromedin U (NMU)-immunoreactivity was mainly confined to the nerve plexus of the inner submucosal and mucosal regions. After colchicine treatment, a high number of immunoreactive nerve cell bodies was observed in the plexus submucosus internus (Meissner), whereas only a low number was found in the plexus submucosus externus (Schabadasch). The plexus myentericus as well as the aganglionic nerve meshworks in the circular and longitudinal smooth muscle layers almost completely lacked NMU-immunoreactivity. Double-labeling experiments demonstrated the occurrence of distinct NMU-containing neuron populations in the plexus submucosus internus: (1) relatively large type-II neurons revealing immunoreactivity for NMU and calcitonin gene-related peptide (CGRP) and/or substance P (SP); (2) a group of small NMU- and SP-immunoreactive neurons; (3) a relatively low number of small neurons displaying immunoreactivity for NMU but not for SP. Based on its distributional pattern, it is concluded that NMU plays an important role in the regulation and control of mucosal functions.     

Functional role of newly formed pore complexes in postmitotic nuclear reorganization

Many nuclear proteins are released into the cytoplasm at prometaphase and are transported back into the daughter nuclei at the end of mitosis. To determine the role of this reentry in nuclear remodelling during early interphase, we experimentally manipulated nuclear protein uptake in dividing cells. Recently we and others have shown that signal-dependent, pore complex-mediated uptake of nuclear protein is blocked in living cells on microinjection of the lectin wheat germ agglutinin (WGA), or of antibodies such as PI1 that are directed against WGA-binding pore complex glycoproteins. In the present study, we microinjected mitotic PtK₂ cells with WGA or antibody PI1 and followed nuclear reorganization of the daughter cells by immunofluorescence and electron microscopy. The inhibitory effect on nuclear protein uptake was monitored by co-injection of the karyophilic protein nucleoplasmin. When injected by itself early in mitosis, nucleoplasmin became sequestered into the daughter nuclei as they entered telophase. In contrast, nucleoplasmin was excluded from the daughter nuclei in the presence of WGA or antibody PI1. Although PtK₂ cells with blocked nuclear protein uptake completed cytokinesis, their nuclei showed a telophaselike organization characterized by highly condensed chromatin surrounded by a nuclear envelope containing a few pore complexes. These findings suggest that pore complexes become functional as early as telophase, in close coincidence with nuclear envelope reformation. They further indicate that the extensive structural rearrangement of the nucleus during the telophase-G1 transition is dependent on the influx of karyophilic proteins from the cytoplasm through the pore complexes, and is not due solely to chromosome-associated components.Abbreviations WGA wheat germ agglutinin - GlcNAc N-acetylglucosamine     

Use of pheromone chemistry to identify Grapholita lobarzewskii as an occasional pest of apple and plum

As shown by chemical analysis and field trapping, the sex pheromone of Grapholita lobarzewskii consists of a blend of (E)-8-dodecenyl acetate and (Z)-8-dodecenyl acetate. Maximum attractiveness was found at 80–95%. Dodecyl acetate and tetradecyl acetate, both present in the female gland, did not affect trap catch. G. lobarzewskii has recently gained importance as a pest of apple and plum, but has so far been referred to as Grapholita janthinana. The latter is attracted to the same two compounds, but in reversed portions (20% E).

Zusammenfassung Chemische Analysen und Feldversuche zeigen, dass das Sexualpheromon des Kleinen Fruchtwicklers, G. lobarzewskii aus (E)-8-Dodecenylacetat (Z8-12:Ac) und (Z)-8-Dodecenylacetat (Z8-12:Ac) besteht. Die grösste Lockwirkung wurde mit einem Gemisch der beiden Substanzen bei einem E-Anteil von 80–95% erzielt. Dodecylacetat (12:Ac) und Tetradecylacetat (14:Ac) wurden ebenfalls in den weiblichen Pheromondrüsen nachgewiesen, hatten aber im Freiland keinen Einfluss auf die Lockwirkung. G. lobarzewskii tritt seit einiger Zeit in der Schweiz als Schädling von Apfel und Zwetschge auf und ist an ihrem charakteristischem Frassgang leicht erkennbar. Die Art wurde bisher irrtümlich als G. janthinana bezeichnet. G. janthinana lebt auf Rosaceen, und wird vom gleichen Substanzpaar angelockt, allerdings bei umgekehrtem Mischungsverhältnis (20% E).

     

Extracellular ATP stimulates poly(inositol phospholipid) hydrolysis and eicosanoid synthesis in mouse peritoneal macrophages in culture

The effects of extracellular ATP on inositol phospholipid breakdown and synthesis of eicosanoids were studied in mouse peritoneal macrophages. Addition of ATP to intact cells labelled with [3H]inositol stimulated a rapid (within 10 s) formation of inositol 1,4,5-trisphosphate and inositol 1,3,4,5-tetrakisphosphate. In parallel there was also a substantial accumulation of inositol 1,3,4-trisphosphate and the monophosphate and bisphosphate derivatives of inositol. Within 10 s after the addition of 30 microM ATP there was a twofold increase in inositol trisphosphate (InsP3), which declined over 2 min. The ED50 for ATP-stimulated generation of InsP3 was approximately 12 microM. ADP and GTP showed only weak effects on InsP3 formation, while AMP and adenosine were completely ineffective at 30 microM. Furthermore, the rank order of potency of ATP analogues was ATP greater than ATP[S] greater than AdoPP[NH]P = AdoPP[CH2]P greater than AdoP[CH2]PP thus, indicating the presence of a P2y-purinergic receptor. Cells labelled with [3H]arachidonic acid showed a 50% increase of label in 1,2-diacylglycerol after 15 s upon stimulation with ATP. In parallel to the stimulation of inositol phospholipid hydrolysis, ATP also caused a marked synthesis of prostaglandin E2 (PGE2) and leukotriene C4 (LTC4) in mouse peritoneal macrophages. The rank order of potency of ATP analogues was identical with that of InsP3 generation. The effect on eicosanoid synthesis could be mimicked by the calcium ionophore A23187 and the phorbol ester 12-O-tetradecanoylphorbol 13-acetate. These results suggest that ATP-induced activation of P2y-purinergic receptors in mouse peritoneal macrophages triggers inositol phospholipid breakdown and eicosanoid synthesis.     

Evidence for a novel growth factor in Xenopus oocytes

Xenopus oocytes contain a novel growth factor, as determined by its effect on the anchorage-dependent and -independent growth of various rat cells and on a Xenopus cell line; it is destroyed by trypsin, acidification (pH 2.0), heating (100 degrees C, 3 min), 8 M urea but not by dithiothreitol. Gel filtration of this activity in nondissociating conditions suggests the existence of aggregates and the presence of a very high (approximately 2000 Kd) and a much lower (approximately 30 Kd) form.
