Endogenous expression of nNOS protein in several neuronal cell lines   总被引:3,自引:0,他引:3  
Several neuronal cell lines were screened for endogenous expression of neuronal nitric oxide synthase (nNOS) protein using Western blot analysis. Detectable levels of the nNOS protein were evident in the SK-N-SH, SH-SY5Y, and N1E-115 neuroblastoma cell lines, as well as the NG108-15 neuroblastoma x glioma hybrid. Only trace amounts were visible in Neuro2A human neuroblastoma cells. The presence of endogenously expressed nNOS in these cells may allow for the study of the interaction between nNOS and the endogenous receptor systems expressed in the same cells.  相似文献

In search of factors that regulate the phenotype of the peroxisomal compartment in wild-type liver parenchymal cells, we compared hepatocyte polarity to peroxisome differentiation, using adult liver as the standard. Differentiation parameters were evaluated in a three-dimensional culture model (spheroid), in 'sandwich' and monolayer primary hepatocyte cultures, and in 15.5 and 18.5-day-old foetal rat liver.Peroxisomes, studied by immunohistochemistry, enzyme histochemistry, and catalase specific activity, were better differentiated depending on foetal age (day 18.5 > day 15.5) and culture type (spheroid > sandwich > monolayer). The hepatocyte polarity markers ATP-, ADP-, and AMP-hydrolysing activities were, in all models, mislocalized at the lateral plasma membrane, whereas in contrast the multidrug resistance-associated protein 2 (mrp2) antigen was always correctly immunolocalized at the apical membrane domain. In cultures, the correct secretion of fluorescein (mrp2-mediated) into bile canaliculi was observed. Bile canaliculi (branching, ultrastructure and immunolocalization of the tight-junction associated protein ZO-1), were better differentiated in 18.5 than in 15.5-day-old foetal liver and in spheroid > sandwich > monolayer cultures.Our results show a parallelism between changes of the peroxisomal compartment and bile canalicular structure together with mrp2-mediated secretory function. Distinct polarization characteristics do not necessarily change simultaneously, suggesting different regulatory mechanisms.  相似文献

Vascular endothelial (VE) cadherin is an endothelial specific cadherin that plays a major role in remodeling and maturation of vascular vessels. Recently, we presented evidence that the extracellular part of VE cadherin, which consists of five homologous modules, associates as a Ca(2+)-dependent hexamer in solution (Legrand, P., Bibert, S., Jaquinod, M., Ebel, C., Hewat, E., Vincent, F., Vanbelle, C., Concord, E., Vernet, T., and Gulino, D. (2001) J. Biol. Chem. 276, 3581-3588). In an effort to identify which extracellular modules are involved in the elaboration and stability of this hexameric structure, we expressed various VE cadherin-derived fragments overlapping individual or multiple successive modules as soluble proteins, purified each to homogeneity, and tested their propensity to self-associate. Altogether, the results demonstrate that, as their length increases, VE cadherin recombinant fragments generate increasingly complex self-associating structures; although single module fragments do not oligomerize, some two or three module-containing fragments self-assemble as dimers, and four module-containing fragments associate as hexamers. Our results also suggest that, before elaborating a hexameric structure, molecules of VE cadherin self-assemble as intermediate dimers. A synergy between the extracellular modules of VE cadherin is thus required to build homotypic interactions. Placed in a cellular context, this particular self-association mode may reflect the distinctive biological requirements imposed on VE cadherin at adherens junctions in the vascular endothelium.  相似文献

The synthetic alkyl-lysophospholipid (ALP), 1-O-octadecyl-2-O-methyl-rac-glycero-3-phosphocholine, is an antitumor agent that acts on cell membranes and can induce apoptosis. We investigated how ALP is taken up by cells, how it affects de novo biosynthesis of phosphatidylcholine (PC), and how critical this is to initiate apoptosis. We compared an ALP-sensitive mouse lymphoma cell line, S49, with an ALP-resistant variant, S49(AR). ALP inhibited PC synthesis at the CTP:phosphocholine cytidylyltransferase (CT) step in S49 cells, but not in S49(AR) cells. Exogenous lysophosphatidylcholine, providing cells with an alternative way (acylation) to generate PC, rescued cells from ALP-induced apoptosis, indicating that continuous rapid PC turnover is essential for cell survival. Apoptosis induced by other stimuli that do not target PC synthesis remained unaffected by lysophosphatidylcholine. Using monensin, low temperature and albumin back-extraction, we demonstrated that ALP is internalized by endocytosis, a process defective in S49(AR) cells. This defect neither involved clathrin-coated pit- nor fluid-phase endocytosis, but depended on lipid rafts, because disruption of these microdomains with methyl-beta-cyclodextrin or filipin (sequestering cholesterol) or bacterial sphingomyelinase reduced uptake of ALP. Furthermore, ALP was found accumulated in isolated rafts and disruption of rafts also prevented the inhibition of PC synthesis and apoptosis induction in S49 cells. In summary, ALP is internalized by raft-dependent endocytosis to inhibit PC synthesis, which triggers apoptosis.  相似文献

Multiheme cytochromes c have been found in a number of sulfate- and metal ion-reducing bacteria. Geobacter sulfurreducens is one of a family of microorganisms that oxidize organic compounds, with Fe(III) oxide as the terminal electron acceptor. A triheme 9.6 kDa cytochrome c(7) from G. sulfurreducens is a part of the metal ion reduction pathway. We cloned the gene for cytochrome c(7) and expressed it in Escherichia coli together with the cytochrome c maturation gene cluster, ccmABCDEFGH, on a separate plasmid. We designed two constructs, with and without an N-terminal His-tag. The untagged version provided a good yield (up to 6 mg/l of aerobic culture) of the fully matured protein, with all three hemes attached, while the N-terminal His-tag appeared to be detrimental for proper heme incorporation. The recombinant protein (untagged) is properly folded, it has the same molecular weight and displays the same absorption spectra, both in reduced and in oxidized forms, as the protein isolated from G. sulfurreducens and it is capable of reducing metal ions in vitro. The shape parameters for the recombinant cytochrome c(7) determined by small angle X-ray scattering are in good agreement with the ones calculated from a homologous cytochrome c(7) of known structure.  相似文献

The research is focused on an ecologically sound and highly productive cultivation system for fodder and/or biomass for thermal power generation on the basis of winter legumes and maize as subsequent summer crop, managed without additional nitrogen fertiliser. Therefore the yield of biomass and N-fixing capacity of a winter pea (Pisum sativum L.) and crimson clover (Trifolium incarnatum L.) monocropped and intercropped with rye (Secale cereale L.) were examined for five years in a field trial. In mid-June above-ground biomass of winter crops was removed and maize transplanted. The winter crops achieved maximum dry matter yield about three to five weeks before maturity. Mixed stands yielded more biomass than pure stands and exhibited greater yield stability. The relative advantage of intercropping, expressed as land equivalent ratio (LER), determined for intercropped winter pea/rye were 1.1 to 1.2 and for crimson clover/rye 1.3. At maturity, the amount of fixed nitrogen ranged between 178 kg N for crimson clover and 242 kg N ha-1 for winter pea, respectively. At the end of anthesis (middle of June, harvesting stage for silage fodder) 75% and 88% of the total fixed nitrogen was achieved, for clover and pea, respectively. In intercropping the amount of fixed nitrogen was lower than in pure stands due to a lower seed density of the legume; however, the N-fixing efficiency was greater than in pure stands. N-release of the winter pea in a pure stand produced a maximum yield in maize (Zea mays L.) without additional N-fertiliser. An additional N mineral fertilisation of 75 to 150 kg N and 75 to 225 kg N was necessary to achieve maximum yields in maize following intercropped winter pea and crimson clover, respectively. Legumes in mixed stands with rye resulted in lower amounts of residual nitrogen after maize harvest. The beneficial effect of legumes on maize can be divided into N-effects and rotation effects. Both effects were positive regarding winter pea. The rotation effect of crimson clover in pure stands on maize was negative. Allelopathic effects and the high sensitivity of crimson clover to mineral nitrogen in the soil, released by residues of the preceding crop, winter rape (Brassica rapa L.), were discussed as the reason for this observation. The combination of the winter pea in pure stand and maize achieved the highest total biomass yield from winter and summer crops, unfertilised (156 dt ha-1 dry). The combinations of intercropped legumes and maize produced biomass yields of 142 to 145 dt ha-1. Because winter pea is highly susceptible to lodging, intercropping with low seed density of rye is recommended (3/4 winter pea, 1/4 rye). The rye crop prevents lodging by providing support and high rates of N-fixation are achieved with high seed density of pea. Intercropping with crimson clover and rye should be based on high seed densities of legumes, too because rye is highly competitive within those mixtures. In a coastal grassland area the influence of plant and corolla size on insect visitation rate, pollen deposition on stigmas and seed set was measured in a natural population of Geranium sanguineum. Visits by bumble bees, the primal pollinators, were significantly affected by plant size measured as the number of flowers, and differences in visit rates among plants were explained by size differences among these. On average, larger plants had bigger corollas. The mean corolla area affected pollen deposition positively whereas deposition did neither affect seed set (number per flower) nor seed weight. Insects reaction to corolla size was dependent on plant size, and thus we found the effect of corolla size on pollen deposition to decline with plant size. Instead pollen deposition per plant seemed to be correlated to overall floral display. We argued that resource status of the individual plants could explain corolla size and seed set variation. The finding of some plant setting seeds while other plant with comparable size did not set any seeds could not be explained satisfactory by resource limitation.  相似文献

In the past decade, ecological surveys emphasized rats and dogs as the most significant animal reservoirs for Schistosoma japonicum (S.j) in the Philippines. However, recent studies demonstrated 51–91% prevalence of schistosomiasis among water buffalo using qPCR in the Sj endemic regions in the Philippines. In order to resolve the inconsistency of reported surveys regarding Sj endemicity among carabao, a domestic water buffalo that is the most important draught animal, we introduced 42 schistosome negative water buffalo to Macanip, Jaro municipality, Leyte, the Philippines, a subsistence rice-farming village that has been the focus of schistosomiasis japonica studies of our group for the past 20 years. We conducted perfusion to the remaining 34 buffalo that survived 10 months of nature exposure and Typhoon Haiyan. Thirty-three water buffalo were found to be positive with at least 1 pair of worms from the mesenteric vein. The infection rate is 97%, with the worm burden of 94 (95% confidence interval, 49–138 worms) worms. To our knowledge, this is the first report about S. japonicum worm burden in naturally infected water buffalo in the Philippines. The fact that with less than one-year of exposure, in this human schistosomiasis endemic area, only 1 out of 34 water buffalo was uninfected is striking. Urgent attention is needed for a cost-effective technique for monitoring Sj infection in animals and humans. Meanwhile, intervention implementation, including water buffalo treatment and vaccination, should be taken into consideration.  相似文献
