Mesenchymal stem cells (MSCs) are being widely studied as potential cell therapy agents due to their immunomodulatory properties, which have been established by in vitro studies and in several clinical trials. Within this context, mesenchymal stem cell therapy appears to hold substantial promise, particularly in the treatment of conditions involving autoimmune and inflammatory components. Nevertheless, many research findings are still contradictory, mostly due to difficulties in characterization of the effects of MSCs in vivo. The purpose of this review is to report the mechanisms underlying mesenchymal stem cell therapy for acute graft-versus-host disease, particularly with respect to immunomodulation, migration, and homing, as well as report clinical applications described in the literature. 相似文献

Lipoteichoic acid (LTA) is an important cell wall component of Gram‐positive bacteria. In Staphylococcus aureus it consists of a polyglycerolphosphate‐chain that is retained within the membrane via a glycolipid. Using an immunofluorescence approach, we show here that the LTA polymer is not surface exposed in S. aureus, as it can only be detected after digestion of the peptidoglycan layer. S. aureus mutants lacking LTA are enlarged and show aberrant positioning of septa, suggesting a link between LTA synthesis and the cell division process. Using a bacterial two‐hybrid approach, we show that the three key LTA synthesis proteins, YpfP and LtaA, involved in glycolipid production, and LtaS, required for LTA backbone synthesis, interact with one another. All three proteins also interacted with numerous cell division and peptidoglycan synthesis proteins, suggesting the formation of a multi‐enzyme complex and providing further evidence for the co‐ordination of these processes. When assessed by fluorescence microscopy, YpfP and LtaA fluorescent protein fusions localized to the membrane while the LtaS enzyme accumulated at the cell division site. These data support a model whereby LTA backbone synthesis proceeds in S. aureus at the division site in co‐ordination with cell division, while glycolipid synthesis takes place throughout the membrane. 相似文献

The ovicidal action of the nematophagous fungus Pochonia chlamydosporia (VC4) was evaluated on Dioctophyma renale eggs under laboratory conditions (Assay A). Next, the enzymatic action of proteases and chitinases produced by P. chlamydosporia (VC4) was evaluated on D. renale eggs, under laboratory conditions (Assay B). At the end of the experiment, there was difference (p < 0.01) in the destruction of eggs in the four concentrations tested in relation to control group at each interval studied. On the other hand, no difference was observed (p > 0.01) among the concentrations in the destruction of eggs. However, there was a trend of increasing mortality with increased concentration. Then (Assay B), it was observed that in the 24-hour interval, the proteases and chitinases of P. chlamydosporia (VC4), either individually or together, caused a significant percentage reduction (p < 0.01) on the number of viable eggs of D. renale, compared to control, with the following reduction values: 27.8% (proteases), 29.4% (chitinases) and 43.4% (proteases + chitinases). Thus, the constant search for alternatives that may help combat the various infectious forms (or eggs and larvae) of potentially zoonotic nematodes is important, as in the use of fungi destroyers of eggs. Therefore, it is suggested that the application of P. chlamydosporia would be an approach in the biological control of nematodes. 相似文献

Introduction
Multiple myeloma (MM) development involves a series of genetic abnormalities and changes in the bone marrow (BM) microenvironment, favoring the growth of the tumor and failure of local immune control. T regulatory (Treg) cells play an important role in dampening anti-tumor immune responses while T-helper-17 (Th17) cells seem to be critical for the eradication of malignant cells. The aim of our study was to characterize the expression of Treg- and Th17-related genes in total myeloma BM samples to assess their role as biomarkers, prognostic factors, and possible therapeutic targets in this incurable disease.Methods
Expression of markers for Treg (FOXP3, CTLA4) and Th17 cells (RORγt) was determined by quantitative real-time PCR in BM aspirates of 46 MM patients, four patients with monoclonal gammopathy of undetermined significance, five solitary plasmacytomas, and five healthy BM donors. Gene expression was evaluated regarding an influence on the patients’ overall survival (OS).Results
FOXP3 and CTLA4 presented a sixfold (p = 0.02) and 30-fold higher expression (p = 0.03), respectively, in MM patients than in controls. RORγt expression was similar in MM patients and controls. Median OS of MM patients was 16.8 (range 4.5–29.1) months, and international staging system was the only independent prognostic factor for patients survival.Conclusions
Overexpression of FOXP3 and CTLA4 in total BM samples suggests a local accumulation of immunosuppressive Tregs, the MM tumor environment, possibly dampening anti-tumor host immune responses. Therapeutic approaches targeting Treg cells and restoring local anti-tumor immunity may provide new treatment strategies for this incurable malignancy. 相似文献805.

Vernonieae is one of the major tribes in Asteraceae (subfamily Cichorioideae) with ca. 1,100 species placed into 129 genera. Currently, 21 subtribes are recognized in Vernonieae and one of them is Lychnophorinae, almost entirely endemic to Brazil, containing 11 genera and ca. 100 species. About 42 % of Lychnophorinae genera are monophyletic, reflecting the poorly understood relationships among the members of the group. Trichomes are one of the most useful anatomical characters to be used in angiosperm taxonomy; they are diverse, exist in many taxa and are not difficult to study. This work intends to illustrate non-glandular leaf trichome diversity in Lychnophorinae and discuss this diversity in the light of the subtribe’s taxonomy. Sampled material included 67 species of 11 genera. Macerations and free hand sections were performed to be analyzed in the light microscope and photographed. A phenogram was generated using a matrix with 67 terminals (species) and 18 characters coded as binary. The subtribe Lychnophorinae displays a great diversity of non-glandular trichomes (5 types and 18 subtypes). The present study reveals the great diversity of non-glandular trichomes in Lychnophorinae. While trichome complement is of little use to distinguish genera, it appears to be a valuable characteristic at a lower taxonomic level to identify closely morphologically related species. 相似文献

The organization of knee articular cartilage of the bullfrog (Lithobates catesbeianus) differs in relation to morphofunctional adaptation in many aspects from similar structures in mammals. Thus, we investigated the structural organization and distribution of the extracellular matrix components in three articular cartilage regions in the distal epiphysis of the femur and proximal epiphysis of the tibia in male bullfrogs at 7, 540 and 1,080 days after metamorphosis. Cartilage thickness and cell density decreased in all regions with age. The basophilia differed among cartilage sites during aging. Calcium deposits were detected in growth cartilage of the femur and tibia in older animals. Immunohistochemical staining for chondroitin-6-sulfate was positive in the pericellular and territorial matrix in all samples. Positive immunostaining for type I collagen was observed in the superficial layer at all ages and in ossification centers of older animals. Reactivity to type II collagen was intense and was found throughout the stroma at all ages. Ultrastructural analysis of the epiphyseal region, in young animals, showed that the cytoplasm of chondrocytes was rich in rough endoplasmic reticulum, Golgi complex and mitochondria. In old animals, were observed a reduction in the size and number of mitochondria, disintegration of rough endoplasmic reticulum, and vacuolization of the Golgi complex. The bullfrog articular cartilage presented structural and organizational changes during aging which may contribute to the functional cartilage deterioration in old animals. 相似文献

Human–megafauna interaction in the Americas has great scientific and ethical interest because of its implications on Pleistocene extinction. The Arroyo del Vizcaíno site near Sauce, Uruguay has already yielded over 1000 bones belonging to at least 27 individuals, mostly of the giant sloth Lestodon. The assemblage shows some taphonomic features suggestive of human presence, such as a mortality profile dominated by prime adults and little evidence of major fluvial transport. In addition, several bones present deep, asymmetrical, microstriated, sharp and shouldered marks similar to those produced by human stone tools. A few possible lithic elements have also been collected, one of which has the shape of a scraper and micropolish consistent with usage on dry hide. However, the radiocarbon age of the site is unexpectedly old (between 27 and 30 thousand years ago), and thus may be important for understanding the timing of the peopling of America. 相似文献

A new species of Tovellia, T. aveirensis, is described on the basis of light (LM) and scanning electron microscopy (SEM) of motile cells and resting cysts, complemented with transmission electron microscopy (TEM) of flagellate cells and phylogenetic analysis of partial sequences of the large subunit ribosomal rRNA gene. Both vegetative cells and several stages of a life cycle involving sexual reproduction and the production of resting cysts were examined in cultures established from a tank in the University of Aveiro campus. Vegetative cells were round and little compressed dorsoventrally; planozygotes were longer and had a proportionally larger epicone. Chloroplast lobes were shown by TEM to radiate from a central, branched pyrenoid, although this was difficult to ascertain in LM. The amphiesma of flagellate cells had mainly 5 or 6-sided vesicles with thin plates, arranged in 5–7 latitudinal series on the epicone, 3–5 on the hypocone. The cingulum had 2 rows of plates, the posterior row extending into the hypocone and crossed by a series of small projecting knobs along the lower edge of the cingulum. A line of narrow amphiesmal plates extended over the cell apex, from near the cingulum on the ventral side to the middle of the dorsal side of the epicone. Eight or 9 narrow amphiesmal plates lined each side of this apical line of plates (ALP). Resting cysts differed from any described before in having numerous long, tapering spines with branched tips distributed over most of the surface. Most mature cysts showed an equatorial constriction. Neither cysts nor motile cells were seen to accumulate red cytoplasmic bodies in any stage of the cultures. The phylogenetic analysis placed, with high statistical support, the new species within the genus Tovellia; it formed a clade, with moderate support, with T. sanguinea, a species notable for its reddening cells. 相似文献
