In this study, we determined the efficacy of minocycline, a second generation tetracycline, in preventing male germ cell apoptosis after withdrawal of gonadotropins and intratesticular testosterone (T). Groups of 5 male rats received one of the following treatments daily for 5 days: (i) daily sc injection of GnRH-A (1.6 mg/kg BW), (ii) oral administration of 30% gum acacia as a vehicle control, and (iii) GnRH-A + oral administration of 50 or 100 mg/kg BW of minocycline. Minocycline at both 50 and 100 mg dose levels significantly (P < 0.05) prevented GnRH-A -induced germ cell apoptosis by 59.4% and 62.2%, respectively, and fully prevented PARP cleavage. Minocycline-mediated protection occurred at the mitochondria, involving the restoration of the BCL-2 levels and, in turn, suppression of cytochrome c and DIABLO release. Minocycline was also effective in preventing human male germ cell apoptosis induced by hormone free culture condition. 相似文献

Airborne pollen produced by Betulaceae and Corylaceaeis present in the Trieste area for a long period fromJanuary to June, but only in April it does representa considerable proportion (436 p/m3) of the totalpollen count (1193 p/m3). In the years considered (1995–1997), there was a gradual increase in thepollen count of Corylaceae and Betulaceae: frommaximum levels of 580 p/m3 in 1995 to 1218 p/m3 in 1997, and the taxon making the mostsignificant contribution to the pollen concentrationcurve was Ostrya carpinifolia.
Sensitization to Betulaceae and Corylaceae wasanalyzed in 2213 subjects visiting our clinic betweenJanuary 1st 1995 and December 31th 1997, withallergic symptoms believed to be IgE mediated. Of thegroup, 1292 (58.4%) were atopic by skin prick testand 328 of them (25.4%) were sensitized to Betulaceaeand Corylaceae. Of the 328 subjects, 72.6% werecosensitized to Gramineae, 56.1% to Oleaceae, 42.1%to Compositae, and just over half to house-dust mites(52.1%). Only ten cases were mono-sensitized (3%).Of the subjects sentitized to Betulaceae andCorylaceae, 163 complained of rhinitis (72%) and 110of asthma (33.6%), often in association withrhinitis; 177 (54%) had only seasonal symptoms.Sensitization to Betulaceae and Corylaceae is high,but its role in inducing allergic respiratorysymptoms is difficult to evaluate because almost allpatients were sensitized to other pollen types. Inconclusion, the role played by this family of trees indetermining allergic respiratory symptoms could becomeincreasingly important in this geographical area inthe future, if pollen levels continue to rise at theirpresent rates. For the moment, sensitivity toBetulaceae-Corylaceae among the population is presentalmost exclusively in subjects sensitized also toother pollen types. 相似文献

Ascidians, invertebrates belonging to the subphylum Urochordata (Tunicata), are renowned for their great chemical diversity,
and during the last 25 years, they have been shown to produce an array of cytotoxic molecules. Among the first six marine-derived
compounds that have reached clinical trials as antitumor agents, three are derived from ascidians, as evidence of the high
potential of these organisms as a new source of antitumor compounds. Reported in this communication are some recent results
on the chemistry of Mediterranean ascidians; a number of new molecules with different structural features but all endowed
with antiproliferative or cytotoxic activity are discussed. These results strongly evidence the highly significant role that

In general, hatchery salmonid smolts experience higher mortality during migration than wild smolts, which is suggested to
be due to domestication effects and that hatchery fish lack experience of the natural environment. However, possible differences
in feeding during smolt migration between hatchery and wild smolts have rarely been addressed. We compared the number of feeding
smolts and stomach fullness among wild Atlantic salmon smolts, hatchery-reared smolts released as 1-year-old parr, and hatchery-reared
smolts released as 2-year-old smolts during their descent to sea in River Tornionjoki. In addition, estimations of prey selection
among the smolt groups were conducted. A high proportion of wild smolts and smolts stocked as parr actively fed during the
smolt migration. A lower proportion of smolts stocked as smolts was feeding and their stomach fullness were much reduced in
comparison with the two other groups. The study also indicated that the feeding of migrating smolts is selective rather than
opportunistic. In conclusion, this study suggests that stocked 2-year-old smolts may enter sea with an inferior foraging behaviour
and it is a possibility that this may contribute to the observed low post-smolt survival in the Baltic Sea. 相似文献

G-protein-coupled receptor function involves interactions between the receptor, G-proteins and effectors in the cell plasma
membrane. The main biochemical processes have been individually identified but the mechanisms governing the successive protein–protein
interactions of this complex multi-molecular machinery have yet to be established. We discuss advances in understanding the
functional dynamics of the receptor resulting from diffusion measurements, and in the context of the plasma membrane organization.

Scandinavian moose (Alces alces) eat Scots pine (Pinus sylvestris) in winter. Although North American moose are known to eat conifers such as true firs (Abies spp.) in winter, substantial consumption of pine by moose in North America has not been documented. Here, we document short-term winter preferences of human-habituated northwestern moose (Alces alces andersoni) for branches of mature North American and European conifer species as determined by a cafeteria-style feeding trial. Moose selected for species such as Douglas fir (Pseudotsuga menziesii; from which they took the smallest bite diameters) while avoiding species such as lodgepole pine (Pinus contorta; from which they took the largest bites) and hybrid white spruce (Picea glauca × engelmanii). The amount of species-specific biomass consumed by moose was negatively correlated with bite diameters taken from branches of those species and did not appear to be significantly influenced by differences in twig morphology between species. Our trial suggests that northwestern moose readily consume conifers in winter and, from the species we tested, prefer Douglas fir. While no clear preference existed between Scots pine and lodgepole pine, moose avoided lodgepole pine, but not Scots pine, relative to Douglas fir. Our trial suggests that northwestern moose are more likely to feed on the branches of Douglas fir than pine, which may be of interest to foresters managing conifers within the North American range of moose, particularly where Scots pine are being considered for planting. 相似文献

Manganese is essential for several metabolic pathways but becomes toxic in excessive amounts. Manganese levels in the body are therefore tightly regulated, but the responsible protein(s) remain incompletely known. We studied two consanguineous families with neurologic disorders including juvenile-onset dystonia, adult-onset parkinsonism, severe hypermanganesemia, polycythemia, and chronic hepatic disease, including steatosis and cirrhosis. We localized the genetic defect by homozygosity mapping and then identified two different homozygous frameshift SLC30A10 mutations, segregating with disease. SLC30A10 is highly expressed in the liver and brain, including in the basal ganglia. Its encoded protein belongs to a large family of membrane transporters, mediating the efflux of divalent cations from the cytosol. We show the localization of SLC30A10 in normal human liver and nervous system, and its depletion in liver from one affected individual. Our in silico analyses suggest that SLC30A10 possesses substrate specificity different from its closest (zinc-transporting) homologs. We also show that the expression of SLC30A10 and the levels of the encoded protein are markedly induced by manganese in vitro. The phenotype associated with SLC30A10 mutations is broad, including neurologic, hepatic, and hematologic disturbances. Intrafamilial phenotypic variability is also present. Chelation therapy can normalize the manganesemia, leading to marked clinical improvements. In conclusion, we show that SLC30A10 mutations cause a treatable recessive disease with pleomorphic phenotype, and provide compelling evidence that SLC30A10 plays a pivotal role in manganese transport. Streptococcus suis Dpr is an iron-binding protein involved in oxidative stress resistance. It belongs to the bacterial Dps protein family whose members form dodecameric assemblies. Previous studies have shown that zinc and terbium inhibit iron incorporation in Listeria innocua Dps protein. In order to gain structural insights into the inhibitory effect of zinc and terbium, the crystal structures of Streptococcus suis Dpr complexes with these ions were determined at 1.8 A and 2.1 A, respectively. Both ions were found to bind at the ferroxidase center and in the same location as iron. In addition, a novel zinc-binding site formed by His40 and His44 was identified. Both His residues were found to be present within all known Streptococcus suis Dpr variants and in Streptococcus pneumoniae, Streptococcus gordonii, and Streptococcus sanguinis Dpr proteins. Amino acid sequence alignment of Dpr with other Dps family members revealed that His44 is highly conserved, in contrast to His40. The inhibitory effect of zinc and terbium on iron oxidation in Dpr was studied in vitro, and it was found that both ions at concentrations >0.2 mM almost completely abolish iron binding. These results provide a structural basis for the inhibitory effect of zinc and terbium in the Dps family of proteins, and suggest a potential role of the Dps proteins in zinc detoxification mechanisms involving the second zinc-binding site. 相似文献

We investigated the association of the -11,391G>A, -11,377G>C, +45T>G, and +276G>T adiponectin single-nucleotide polymorphisms (SNPs) and expected haplotypes with the insulin resistance (IR) state in overweight/obese children; by using the haplotype background analysis, we also assessed the effect of each SNP independently. GG genotype at the -11,391 locus was associated with higher fasting insulin levels and homeostasis model assessment-IR index and lower adiponectin levels compared with GA + AA genotypes (p = 0.01, 0.002, and 0.03, respectively). Those heterozygous and homozygous for G allele at the -11,377 locus showed higher fasting glucose (p = 0.001 for both), fasting insulin (p = 0.001 for both), homeostasis model assessment-IR index (p < 0.001 for both), and triglyceride levels (p = 0.02 and 0.03, respectively) and lower adiponectin levels (p = 0.002 and 0.02, respectively) compared with C homozygotes. The +45G carriers showed higher fasting and 2-hour glucose levels (p = 0.01 for both) and lower adiponectin levels (p = 0.02) compared with non-carriers. Haplotype analysis suggested that, considering the same haplotypic background, each of the three polymorphisms exerted an independent effect on investigated parameters. The -11,391G>A, -11,377C>G, and +45T>G SNPs are associated with IR syndrome in overweight/obese children; they independently influence the investigated variables. The effect of +45T>G SNP seems to be marginal compared with the promoter SNPs. The GGT haplotype is associated with the highest degree of IR. 相似文献
