The genome of the cold-adapted bacterium Pseudoalteromonas haloplanktis TAC125 contains multiple genes encoding three distinct monomeric hemoglobins exhibiting a 2/2 ??-helical fold. In the present work, one of these hemoglobins is studied by resonance Raman, electronic absorption and electronic paramagnetic resonance spectroscopies, kinetic measurements, and different bioinformatic approaches. It is the first cold-adapted bacterial hemoglobin to be characterized. The results indicate that this protein belongs to the 2/2 hemoglobin family, Group II, characterized by the presence of a tryptophanyl residue on the bottom of the heme distal pocket in position G8 and two tyrosyl residues (TyrCD1 and TyrB10). However, unlike other bacterial hemoglobins, the ferric state, in addition to the aquo hexacoordinated high-spin form, shows multiple hexacoordinated low-spin forms, where either TyrCD1 or TyrB10 can likely coordinate the iron. This is the first example in which both TyrCD1 and TyrB10 are proposed to be the residues that are alternatively involved in heme hexacoordination by endogenous ligands.  相似文献

Background

In a globalized word, prevention of infectious diseases is a major challenge. Rapid detection of viable virus particles in water and other environmental samples is essential to public health risk assessment, homeland security and environmental protection. Current virus detection methods, especially assessing viral infectivity, are complex and time-consuming, making point-of-care detection a challenge. Faster, more sensitive, highly specific methods are needed to quantify potentially hazardous viral pathogens and to determine if suspected materials contain viable viral particles. Fourier transform infrared (FTIR) spectroscopy combined with cellular-based sensing, may offer a precise way to detect specific viruses. This approach utilizes infrared light to monitor changes in molecular components of cells by tracking changes in absorbance patterns produced following virus infection. In this work poliovirus (PV1) was used to evaluate the utility of FTIR spectroscopy with cell culture for rapid detection of infective virus particles.

Results

Buffalo green monkey kidney (BGMK) cells infected with different virus titers were studied at 1 - 12 hours post-infection (h.p.i.). A partial least squares (PLS) regression method was used to analyze and model cellular responses to different infection titers and times post-infection. The model performs best at 8 h.p.i., resulting in an estimated root mean square error of cross validation (RMSECV) of 17 plaque forming units (PFU)/ml when using low titers of infection of 10 and 100 PFU/ml. Higher titers, from 103 to 106 PFU/ml, could also be reliably detected.

Conclusions

This approach to poliovirus detection and quantification using FTIR spectroscopy and cell culture could potentially be extended to compare biochemical cell responses to infection with different viruses. This virus detection method could feasibly be adapted to an automated scheme for use in areas such as water safety monitoring and medical diagnostics.  相似文献

The individual and interactive effects of temperature, pH, NaCl, and aw on the proteolytic and lipolytic activities of Lactobacillus delbrueckii subsp. bulgaricus B397, Lactococcus lactis subsp. lactis T12, and Lb. plantarum 2739 were studied by quadratic response surface methodology. The effects on enzyme activities depended on the interactions among the independent variables, type of activity, substrate and, especially, species. The proteinase activity of strains B397 and T12 was affected differently by pH as individual or interactive terms depending on the type of substrate sl- or β-casein. The increase of NaCl concentration (2.5–7.5%) under cheese-like conditions had a negative effect on the proteinase activity of strain T12. The effect of NaCl was related to the corresponding decrease in aw. Aminopeptidases N and A, iminopeptidase and endopeptidase of Lc. lactis subsp. lactis T12 were strongly inhibited by pH 5–6 and NaCl concentration higher than 3.75%. The negative effects of these independent variables was in several cases enhanced by their interaction and/or by the interaction with the lowest temperatures. In contrast, the same peptidases of Lb. plantarum 2739 retained a high activity under the very hostile environmental conditions. Iminopeptidase and especially endopeptidase activities of strain 2739 were stimulated slightly by NaCl at concentrations up to 5%. Lipase/esterase activity of Lb. delbrueckii subsp. bulgaricus B397 was markedly inhibited under cheese-like conditions.  相似文献

Alcoholic extracts of bakers' yeast (Saccharomyces cerevisiae) have been used for over 60 years in over-the-counter medications for the treatment of hemorrhoids, burns, and wounds. Although previous studies suggested that small peptides were responsible for the medical observations, the peptides were never resolved into separate fractions and identified. In the present report, a protein fraction was prepared by RPC18 chromatography of the extract which enhances wound closure in both diabetic and non-diabetic littermates. The peptides are active in nanomolar amounts and are 600 times more active than the initial extract. SDS-PAGE and N-terminal amino acid sequencing identified 4 polypeptides in the extract. Three of the proteins were small molecular weight stress-associated proteins: copper, zinc superoxide-dismutase, ubiquitin, and glucose lipid regulated protein (HSP 12). The fourth protein, acyl-CoA binding protein II, has not been previously associated with stress proteins.  相似文献

Bosmina (Cladocera) populations, especially within the subgenus Eubosmina, show a variety of phenotypes that exhibit large differences in body size and shape and antennule length. In some populations, the morphological traits also vary during the season, with the most extreme forms occurring in periods with high densities of certain invertebrate predators. However, while temporal phenotypic variation in other cladocerans, as in the family Daphnidae, has been shown to be an adaptation to reduce the risk of predation by invertebrate predators, the reason for such changes in Bosmina is much less clear. We examined whether certain morphological traits in Bosmina species could act as a defence against invertebrate predators. We tested three Bosmina forms (subgenus Eubosmina), differing in morphology from each other, which are found in lakes together with the predator Leptodora kindtii (Cladocera). Bosmina (E.) longispina has a relatively low and elongated carapace with a caudal mucro, and short antennule, B. (E.) coregoni gibbera has a higher and more protruding carapace without caudal mucro, and a much longer antennule. Finally, B. (E.) coregoni retro extensa has a carapace like that of B. longispina but with no caudal mucro and a much longer antennule. In one experiment, B. longispina and B. gibbera were exposed for 12 h to Leptodora in Petri dishes. In a second experiment, we observed directly the escape efficiency of B. longispina, B. gibbera and B. retro extensa, and the handling time of Leptodora. The two Bosmina forms with more extreme morphological features had a lower death rate and higher escape efficiency than B. longispina. Prey that escaped did so, in most cases, within 5 min. Predator handling time was correlated to predator body length and antennule length of the prey. The results suggests that Bosmina species with extreme morphological traits may be less vulnerable to invertebrate predators. Received: 8 June 1998 / Accepted: 5 October 1998  相似文献

Natural killer (NK) cells are important effectors in resistance to viral infections. The role of NK cells in the acute response to human immunodeficiency virus 1 (HIV-1) infected cells was investigated in a mouse model based on a HIV-1/murine leukemia virus (MuLV) pseudovirus. Splenocytes infected with HIV-1/MuLV were injected intraperitoneally and local immunologic responses and persistence of infected cells were investigated. In vivo depletion with an anti-NK1.1 antibody showed that NK cells are important in resistance to virus infected cells. Moreover, NK cell frequency in the peritoneal cavity increased in response to infected cells and these NK cells had a more mature phenotype, as determined by CD27 and Mac-1 expression. Interestingly, after injection of HIV-1/MuLV infected cells, but not MuLV infected cells, peritoneal NK cells had an increased cytotoxic activity. In conclusion, NK cells play a role in the early control of HIV-1/MuLV infected cells in vivo.  相似文献

It has been shown that the propensity of a protein to form amyloid-like fibrils can be predicted with high accuracy from the knowledge of its amino acid sequence. It has also been suggested, however, that some regions of the sequences are more important than others in determining the aggregation process. Here, we have addressed this issue by constructing a set of “sequence scrambled” variants of the first 29 residues of horse heart apomyoglobin (apoMb1-29), in which the sequence was modified while maintaining the same amino acid composition. The clustering of the most amyloidogenic residues in one region of the sequence was found to cause a marked increase of the elongation rate (kagg) and a remarkable shortening of the lag phase (tlag) of the fibril growth, as determined by far-UV circular dichroism and thioflavin T fluorescence. We also show that taking explicitly into consideration the presence of aggregation-promoting regions in the predictive methods results in a quantitative agreement between the theoretical and observed kagg and tlag values of the apoMb1-29 variants. These results, together with a comparison between homologous segments from the family of globins, indicate the existence of a negative selection against the clustering of highly amyloidogenic residues in one or few regions of polypeptide sequences.  相似文献

The influence of hormones in water subphase on the stability of monolayers built of phospholipid mixtures extracted from embryogenic (PLE) and nonembryogenic (PLNE) wheat calli was examined. Additionally, experiments on individual lipids, dipalmitoylphosphatidylcholine (DPPC) and dipalmitoylphosphatidic acid (DPPA), were performed. DPPC was chosen because it was the main phospholipid present in both calli. Negatively charged DPPA could mimic a negatively charged natural mixture of lipids. As hormones, auxins (IAA and 2,4-D), cytokinins (zeatin and kinetin) and zearalenone were chosen. The time of monolayer stability for PLNE calli was much longer than for PLE calli. Kinetics of monolayer stability of PLNE was similar to DPPA, whereas that of PLE was similar to DPPC. Generally, hormones increased the time after which the monolayer stability was reached and decreased the surface pressure. The greatest effect was observed for auxins (especially IAA), whereas cytokinins affected the monolayer stability to a lesser degree.  相似文献
