Recombinant proteins are of great commercial and scientific interest. However, most current production methods using mammalian cells involve the time- and labor-intensive step of creating stable cell lines. Although production methods based on transient gene expression could offer a significant improvement, transient transfection is currently still limited by low titers and low specific productivity compared to stable cell lines. To overcome these bottlenecks, we have explored the use of various growth factors to enhance specific productivity and titers in the context of transient gene expression. For that purpose, several growth factors were cloned and screened for their effect on transient gene expression in HEK293E and CHO-DG44 cells. In particular, acidic fibroblast growth factor (aFGF) was able to increase specific productivity by 60% and recombinant protein titers by 80% in HEK293E cells, while FGF9 increased titers by 250% in CHO-DG44 cells. 相似文献

The fruit mineral contents (K, Ca, Ba, Br, Zn, Co, Cr, Fe, Na, Rb, Cs, and Sr) of four native and two exotic naturalized shrubs
growing in different areas of the Andean Patagonian region of Argentina were investigated. Native species Berberis darwinii, Berberis microphylla (Berberidaceae), Aristotelia chilensis (Elaeocarpaceae) and Ribes magellanicum (Saxifragaceae) produce small berries while the fruits of the exotic species Rosa rubiginosa and Rosa canina (Rosaceae) are aggregates of aquenes. They are used to prepare jams, tea, liquors, and ice creams. Native shrub fruits had
higher content of Br, Zn, Co, Cr, Fe, Mo, and Na than those of the exotic naturalized species. Rosa species showed the highest contents of Ca and Ba in their fruits (the mean content doubled those of the native plant fruits).
The fruit nutrient content found in the studied species was similar or higher than other values reported for fruits of temperate

Production of polyhydroxyalkanoates (PHA) by mixed cultures has been widely studied in the last decade. Storage of PHA by
mixed microbial cultures occurs under transient conditions of carbon or oxygen availability, known respectively as aerobic
dynamic feeding and anaerobic/aerobic process. In these processes, PHA-accumulating organisms, which are quite diverse in
terms of phenotype, are selected by the dynamic operating conditions imposed to the reactor. The stability of these processes
during long-time operation and the similarity of the polymer physical/chemical properties to the one produced by pure cultures
were demonstrated. This process could be implemented at industrial scale, providing that some technological aspects are solved.
This review summarizes the relevant research carried out with mixed cultures for PHA production, with main focus on the use
of wastes or industrial surplus as feedstocks. Basic concepts, regarding the metabolism and microbiology, and technological
approaches, with emphasis on the kind of feedstock and reactor operating conditions for culture selection and PHA accumulation,

Natural killer (NK) cells are important effectors in resistance to viral infections. The role of NK cells in the acute response to human immunodeficiency virus 1 (HIV-1) infected cells was investigated in a mouse model based on a HIV-1/murine leukemia virus (MuLV) pseudovirus. Splenocytes infected with HIV-1/MuLV were injected intraperitoneally and local immunologic responses and persistence of infected cells were investigated. In vivo depletion with an anti-NK1.1 antibody showed that NK cells are important in resistance to virus infected cells. Moreover, NK cell frequency in the peritoneal cavity increased in response to infected cells and these NK cells had a more mature phenotype, as determined by CD27 and Mac-1 expression. Interestingly, after injection of HIV-1/MuLV infected cells, but not MuLV infected cells, peritoneal NK cells had an increased cytotoxic activity. In conclusion, NK cells play a role in the early control of HIV-1/MuLV infected cells in vivo. 相似文献

When far red light preincubated cells of Euglena gracilis are transferred to dark or light, chlorophyll fluorescence (F0 and Fm) decreases. Non-photochemical quenching in the dark is suggested to be induced partly by chlororespiration and partly by
changes in the distribution of excitation energy between the photosystems. Depending on the light intensities it was possible
to resolve the non-photochemical quenching into at least three different components. The slowest relaxation phase of non-photochemical
quenching occurred only after exposure to high light and was assigned to photoinhibition. The other two components were an
energy-dependent quenching (qE), and the one which we attribute to a spill over mechanism. We suggest that both photosystems
use a common antenna system consisting of LHC I and LHC II proteins. In contrast to higher plants, qE in Euglena gracilis is independent of the xanthophyll cycle and an aggregation of LHC II.

We evaluated whether nutritional vitamin A deficiency generates oxidative stress and inflammation in aorta. Wistar male rats (21 days old) were given free access to a control (8 mg retinol as retinyl palmitate/kg) or a vitamin A- deficient diet for three months. One group of deficient animals was fed with the control diet fifteen days before sacrifice. Thiobarbituric acid-reactive substances (TBARS) and nitrite concentration where both analyzed in serum and aorta. Aorta Copper-Zinc Superoxide dismutase (CuZnSOD), Glutathion peroxidase (GPx) and Catalase (CAT) activities were measured. In addition, binding activity of the nuclear factor- kB (NF-kB), inducible and endothelial Nitric Oxide synthase (iNOS and eNOS, respectively) and Ciclooxygenase-2 (COX-2) expressions were determinated in aorta. Rats fed the vitamin A- deficient diet were characterized by sub-clinical plasma retinol concentration and showed increased serum and aorta concentrations of TBARS compared to controls. Lower than control activities of CuZnSOD, GPx, and CAT were observed in aorta of the vitamin A- deficient group. The binding activity of NF- kB was higher in vitamin A- deficient animals than controls. In addition, NO production evaluated as nitrite concentration increased in aorta and serum, associated with a higher expression of iNOS, eNOS and COX-2 in aorta of vitamin A-deficient rats. The incorporation of vitamin A into the diet of vitamin A-deficient rats reverted the changes observed in TBARS level, CuZnSOD and GPx activities, nitrite concentration and also, iNOS, eNOS and COX-2 expression. Temporal trends in macrobenthic communities of the Valli di Comacchio, the largest lagoonal ecosystem in the Po River deltaic area, were examined at four sampling sites located along a putative stress gradient and characterized by different levels of disturbance. Trends of parameters characterizing macrobenthic community structure were analyzed using data collected quarterly from 1996 to 2002. The positive slopes in trends of proportion of abundance of opportunistic species observed at all the four study sites indicated the persistence of stressful conditions. Conversely, temporal trends of species richness and diversity showed a positive slope at three out of four sites, thus indicating an increase of community structural complexity. It is suggested that the increase in abundance of detritivorous opportunistic species, as well as the increase in diversity and species richness, is probably driven by the frequency of physical disturbance that resets the ‘successional clock’ of the benthic communities of the Valli. Redundancy was assessed through multivariate analyses and a ‘forward selection backward elimination’ algorithm. At the ecosystem level (i.e., merging counts from the four sites), two response units (a response unit is the smallest subset of species for which the Spearman rank correlation with sample similarities for the full species set exceeds a predetermined value) were obtained. Each of the response units encompassed quite a large taxonomic and functional spread, since all guilds were well represented. Conversely, at the local level, only one response unit was obtained. Species number and function is essential to long-term maintenance of community and ecosystem processes over a complete cycle of common disturbance events. Since redundancy reflects the resistance to change of community functioning, we hypothesize that the macrobenthic communities of the Valli di Comacchio are quite resistant to environmental changes, despite disturbances that generate fluctuations in species abundance and complement at the local level. 相似文献
