The Southern‐fur‐seal belongs to the order Carnivora, suborder Pinnipedia, and Otariidae family. This species inhabits aquatic and terrestrial environments, thus presenting important morphophysiological adaptive changes, especially in the cardiac system. For this purpose, Southern‐fur‐seal (Arctocephalus australis) hearts were used from animals that died from natural causes. Gross morphology observations were supported by light, scanning and transmission electron microscopy. The heart was long and flat; it was lined by pericardium and partly covered by lungs. Structurally, atrium and ventricle muscle fibers exhibit typical features of cardiac fibers revealing myofibrils bundles, mitochondria, plate‐shaped junctions, anastomosis between myofibrils bundles, and electron‐dense granule natriuretic around the nucleus and mitochondria of atrium muscle cells. The Southern‐fur‐seal heart was structurally similar to other mammals; however, it presented morphological changes that assist in their adaptation to their environment.  相似文献

The melanocortin system is an important regulator of energy balance, and melanocortin 4 receptor (MC4R) deficiency is the most common monogenic cause of obesity. We investigated whether the relationship between melanocortin system activity and energy expenditure (EE) is mediated by brown adipose tissue (BAT) activity. Therefore, female APOE*3-Leiden.CETP transgenic mice were fed a Western-type diet for 4 weeks and infused intracerebroventricularly with the melanocortin 3/4 receptor (MC3/4R) antagonist SHU9119 or vehicle for 2 weeks. SHU9119 increased food intake (+30%) and body fat (+50%) and decreased EE by reduction in fat oxidation (−42%). In addition, SHU9119 impaired the uptake of VLDL-TG by BAT. In line with this, SHU9119 decreased uncoupling protein-1 levels in BAT (−60%) and induced large intracellular lipid droplets, indicative of severely disturbed BAT activity. Finally, SHU9119-treated mice pair-fed to the vehicle-treated group still exhibited these effects, indicating that MC4R inhibition impairs BAT activity independent of food intake. These effects were not specific to the APOE*3-Leiden.CETP background as SHU9119 also inhibited BAT activity in wild-type mice. We conclude that inhibition of central MC3/4R signaling impairs BAT function, which is accompanied by reduced EE, thereby promoting adiposity. We anticipate that activation of MC4R is a promising strategy to combat obesity by increasing BAT activity.  相似文献

In this paper, a new species of Vasseuromys, V. bergasensis sp. nov., from the locality of Bergasa (Ebro Basin, Spain), is described. Bergasa contains a fauna belonging to the Late Oligocene (zone MP30), composed—among other species—of Issiodoromys pseudanaema and Rhodanomys transiens. The main diagnostic features of V. bergasensis sp. nov. are the presence of a long centrolophid (fused or not to the mesoconid) in the lower molars, a large reduction in the number and length of extra ridges in the upper and lower molars, the absence of extra ridges between metalophid and centrolophid and between centrolophid and mesolophid, and the absence of the metatrope in more than half the specimens of the upper teeth M1 and M2. V. bergasensis sp. nov. is similar in size to V. elegans and smaller than the other members of the genus. The age and simple dental pattern of the new species of Vasseuromys allow us to hypothesize about relationships within the genus.  相似文献

Important players in triglyceride (TG) metabolism include the liver (production), white adipose tissue (WAT) (storage), heart and skeletal muscle (combustion to generate ATP), and brown adipose tissue (BAT) (combustion toward heat), the collective action of which determine plasma TG levels. Interestingly, recent evidence points to a prominent role of the hypothalamus in TG metabolism through innervating the liver, WAT, and BAT mainly via sympathetic branches of the autonomic nervous system. Here, we review the recent findings in the area of sympathetic control of TG metabolism. Various neuronal populations, such as neuropeptide Y (NPY)-expressing neurons and melanocortin-expressing neurons, as well as peripherally produced hormones (i.e., GLP-1, leptin, and insulin), modulate sympathetic outflow from the hypothalamus toward target organs and thereby influence peripheral TG metabolism. We conclude that sympathetic stimulation in general increases lipolysis in WAT, enhances VLDL-TG production by the liver, and increases the activity of BAT with respect to lipolysis of TG, followed by combustion of fatty acids toward heat. Moreover, the increased knowledge about the involvement of the neuroendocrine system in TG metabolism presented in this review offers new therapeutic options to fight hypertriglyceridemia by specifically modulating sympathetic nervous system outflow toward liver, BAT, or WAT.  相似文献

Background and aims

Nitrogen (N) availability affects water uptake from the roots, which decreases upon N deprivation and increases upon resupply. The aim of this study was to reveal possible mechanisms of regulation of water transport in roots through physiological and morphological adaptations to N availability.

Methods

The effects of continuous N deprivation and following resupply on root morphology, osmotic hydraulic conductivity, and expression of genes for aquaporins (water channels) were examined in rice (Oryza sativa L.) plants. The effect of local N availability was examined by using a split-root system.

Results

N deprivation decreased the expression of root-specific aquaporin genes, whereas N resupply increased their expression. Changes in aquaporin gene expression were correlated with changes in hydraulic conductivity. N deprivation increased dry matter allocation to the roots. In a split-root experiment, the expression of root-specific aquaporin genes was down-regulated in the N-deprived half, whereas it was up-regulated in the N-supplied half.

Conclusion

Our results suggest that expression of genes for root-specific aquaporins underlies the changes in conductivity during continuous N deprivation and resupply. Rice plants seem to adapt to N availability through coordinated adjustment of root proliferation and abundance of aquaporins.  相似文献

In mammals, two carotenoid cleaving oxygenases are known; beta-carotene 15,15′-monooxygenase (BCMO1) and beta-carotene 9′,10′-oxygenase (BCO2). BCMO1 is a key enzyme in vitamin A synthesis by symmetrically cleaving beta-carotene into 2 molecules of all-trans-retinal, while BCO2 is responsible for asymmetric cleavage of a broader range of carotenoids. Here, we show that the Atlantic salmon beta-carotene oxygenase (bco) gene family contains 5 members, three bco2 and two bcmo1 paralogs. Using public sequence databases, multiple bco genes were also found in several additional teleost species. Phylogenetic analysis indicates that bco2a and bco2b originate from the teleost fish specific genome duplication (FSGD or 3R), while the third and more distant paralog, bco2 like, might stem from a prior duplication event in the teleost lineage. The two bcmo1 paralogs (bcmo1 and bcmo1 like) appear to be the result of an ancient duplication event that took place before the divergence of ray-finned (Actinopterygii) and lobe-finned fish (Sarcopterygii), with subsequent nonfunctionalization and loss of one Sarcopterygii paralog. Gene expression analysis of the bcmo1 and bco2 paralogs in Atlantic salmon reveals regulatory divergence with tissue specific expression profiles, suggesting that the beta-carotene oxygenase subtypes have evolved functional divergences. We suggest that teleost fish have evolved and maintained an extended repertoire of beta-carotene oxygenases compared to the investigated Sarcopterygii species, and hypothesize that the main driver behind this functional divergence is the exposure to a diverse set of carotenoids in the aquatic environment.  相似文献

The development of complementary and/or alternative drugs for treatment of hepatitis C virus (HCV) infection is still needed. Antiviral compounds in medicinal plants are potentially good targets to study. Morinda citrifolia is a common plant distributed widely in Indo‐Pacific region; its fruits and leaves are food sources and are also used as a treatment in traditional medicine. In this study, using a HCV cell culture system, it was demonstrated that a methanol extract, its n‐hexane, and ethyl acetate fractions from M. citrifolia leaves possess anti‐HCV activities with 50%‐inhibitory concentrations (IC50) of 20.6, 6.1, and 6.6 μg/mL, respectively. Bioactivity‐guided purification and structural analysis led to isolation and identification of pheophorbide a, the major catabolite of chlorophyll a, as an anti‐HCV compound present in the extracts (IC50 = 0.3 μg/mL). It was also found that pyropheophorbide a possesses anti‐HCV activity (IC50 = 0.2 μg/mL). The 50%‐cytotoxic concentrations (CC50) of pheophorbide a and pyropheophorbide a were 10.0 and 7.2 μg/mL, respectively, their selectivity indexes being 33 and 36, respectively. On the other hand, chlorophyll a, sodium copper chlorophyllin, and pheophytin a barely, or only marginally, exhibited anti‐HCV activities. Time‐of‐addition analysis revealed that pheophorbide a and pyropheophorbide a act at both entry and the post‐entry steps. The present results suggest that pheophorbide a and its related compounds would be good candidates for seed compounds for developing antivirals against HCV.  相似文献

Accumulations of radionuclides in marine macroalgae (seaweeds) resulting from the Fukushima 1 Nuclear Power Plant (F1NPP) accident in March 2011 have been monitored for two years using high-purity germanium detectors. Algal specimens were collected seasonally by snorkeling at Nagasaki, Iwaki, Fukushima Prefecture (Pref.), Japan, ca. 50 km perimeter from the F1NPP. Additional collections were done at Soma, Hironocho, Hisanohama and Shioyazaki in Fukushima Pref. as well as at Chiba Pref. and Hyogo Pref. as controls. In May 2011, specimens of most macroalgal species showed 137Cs levels greater than 3,000 Bq kg?1 at Shioyazaki and Nagasaki. The highest 137Cs level recorded 7371.20 ± 173.95 Bq kg?1 in Undaria pinnatifida (Harvey) Suringar on 2 May 2011, whereas seawater collected at the same time at Shioyazaki and Nagasaki measured 8.41 ± 3.21 and 9.74 ± 3.43 Bq L?1, respectively. The concentration factor of marine macroalgae was estimated to be ca. 8–50, depending on taxa and considering a weight ratio of wet/dry samples of ca. 10. 137Cs level declined remarkably during the following 5–6 months. In contrast, the 137Cs level remained rather stable during the following 12–16 months, and maintained the range of 10–110 Bq kg?1. Contamination was still detectable in many samples in March 2013, 24 months after the most significant pollution.  相似文献

Having a deep genetic structure evolved during its domestication and adaptation, the Asian cultivated rice (Oryza sativa) displays considerable physiological and morphological variations. Here, we describe deep whole-genome sequencing of the aus rice cultivar Kasalath by using the advanced next-generation sequencing (NGS) technologies to gain a better understanding of the sequence and structural changes among highly differentiated cultivars. The de novo assembled Kasalath sequences represented 91.1% (330.55 Mb) of the genome and contained 35 139 expressed loci annotated by RNA-Seq analysis. We detected 2 787 250 single-nucleotide polymorphisms (SNPs) and 7393 large insertion/deletion (indel) sites (>100 bp) between Kasalath and Nipponbare, and 2 216 251 SNPs and 3780 large indels between Kasalath and 93-11. Extensive comparison of the gene contents among these cultivars revealed similar rates of gene gain and loss. We detected at least 7.39 Mb of inserted sequences and 40.75 Mb of unmapped sequences in the Kasalath genome in comparison with the Nipponbare reference genome. Mapping of the publicly available NGS short reads from 50 rice accessions proved the necessity and the value of using the Kasalath whole-genome sequence as an additional reference to capture the sequence polymorphisms that cannot be discovered by using the Nipponbare sequence alone.  相似文献
