Molecular recognition processes between cell surface elements are discussed with special reference to cell surface pattern formation of membrane-bound integral proteins. The existence, as detected by flow cytometric resonance energy transfer (Appendix), and significance of cell surface patterns involving the interleukin-2 receptor, the T-cell receptor–CD3 system, the intercellular adhesion molecule ICAM-1, and the major histocompatilibilty complex class I and II molecules in the plasma membrane of lymphocytes are described. The modulation of antigen presentation by transmembrane potential changes is discussed, and a general role of transmembrane potential changes, and therefore of icon channel activities, adduced as one of the major regulatory mechanisms of cell–cell communications. A general role in the mediation and regulation of intercellular interactions is suggested for cell-surface macromolecular patterns. The dynamic pattern of protein and lipid molecules in the plasma membrane is generated by the genetic code, but has a remarkable flexibility and may be one of the major instruments of accomodation and recognition processes at the cellular level.  相似文献

To establish a protocol suited for the molecular characterization of root induction the influences of explant position, etiolation state and orientation as well as temperature and light intensity on root and callus formation were investigated. Depending on these factors stem discs of Malus incubated on indole-3-butyric acid containing medium and subsequently on hormone-free medium regenerated roots and callus of filamentous and smooth texture to a varying extent. Concentration and incubation duration of indole-3butyric acid strongly affected rooting and the production of smooth callus. Moreover smooth callus was profuse at the low light levels applied during rooting. Rooting efficiency decreased and filamentous callus increased between 19°C and 25°C. Explants close to the shoot apex displayed reduced rooting efficiency and profuse filamentous callus. Summary The morphology of the bovine Sertoli cell was studied during 6 different phases of the spermatogenetic cycle. Tubular dimensions do not vary significantly during the phases. Sertoli cells occupy 27.0% (phase 4) to 38.4% (phase 8) of the tubular epithelium. Sertoli cells of phase 1 are approximately 20% larger than during the other phases. 30–35% of Sertoli cell volume consists of organelles. Mitochondrial (about 5.0%) and nuclear (about 5.7%) volume densities remain remarkably stable during the cycle, irrespective of changes in Sertoli cell size. Phagocytic capacity of bovine Sertoli cells is only moderate. Elimination of excess spermatid cytoplasm occurs to a large extent prior to spermiation. The majority of spermatid residual bodies undergoes autolytic decay while attached to the Sertoli cell apical surface. Aggregates of densely packed cisternae of the smooth endoplasmic reticulum (ER) located in a basal position and associated with the acrosome-phase and maturation-phase spermatids contribute between 14 and 17% to Sertoli cell volume. During phase 3 the ER pinches off a large number of small, smooth-walled vesicles filled with flocculent content. The contact area between Sertoli cells and other tubular constituents changes considerably during the different phases. It is concluded that the blood-testis barrier is particularly impassable during phases 1 and 8. A lipid cycle does not exist in the bovine testicular tubular epithelium.  相似文献

Data on the effects of selenium supplementation on clinical signs and metabolic profiles in women at risk for intrauterine growth restriction (IUGR) are scarce. This study was designed to assess the effects of selenium supplementation on clinical signs and metabolic status in pregnant women at risk for IUGR. This randomized double-blind placebo-controlled clinical trial was performed among 60 women at risk for IUGR according to abnormal uterine artery Doppler waveform. Participants were randomly assigned to intake either 100 μg selenium supplements as tablet (n = 30) or placebo (n = 30) for 10 weeks between 17 and 27 weeks of gestation. After 10 weeks of selenium administration, a higher percentage of women in the selenium group had pulsatility index (PI) of <1.45) (P = 0.002) than of those in the placebo group. In addition, changes in plasma levels of total antioxidant capacity (TAC) (P < 0.001), glutathione (GSH) (P = 0.008), and high-sensitivity C-reactive protein (hs-CRP) (P = 0.004) in the selenium group were significant compared with the placebo group. Additionally, selenium supplementation significantly decreased serum insulin (P = 0.02), homeostasis model of assessment-estimated insulin resistance (HOMA-IR) (P = 0.02), and homeostatic model assessment for B-cell function (HOMA-B) (P = 0.02) and significantly increased quantitative insulin sensitivity check index (QUICKI) (P = 0.04) and HDL-C levels (P = 0.02) compared with the placebo. We did not find any significant effect of selenium administration on malondialdehyde (MDA), nitric oxide (NO), fasting plasma glucose (FPG), and other lipid profiles. Overall, selenium supplementation in pregnant women at risk for IUGR resulted in improved PI, TAC, GSH, hs-CRP, and markers of insulin metabolism and HDL-C levels, but it did not affect MDA, NO, FPG, and other lipid profiles.Clinical trial registration number http://www.irct.ir: IRCT201601045623N64.  相似文献

Several independent studies have supported the association of DYX1C1 with dyslexia, but its role in general reading development remains unclear. Here, we investigated the contribution of this gene to reading, with a focus on orthographic skills, in a sample of 284 unrelated Chinese children aged 5 to 11 years who were participating in the Chinese Longitudinal Study of Reading Development. We tested this association using a quantitative approach for Chinese character reading, Chinese character dictation, orthographic judgment, and visual skills. Significant or marginally significant associations were observed at the marker rs11629841 with children''s orthographic judgments at ages 7 and 8 years (all P values<0.020). Significant associations with Chinese character dictation (all P values<0.013) were also observed for this single-nucleotide polymorphism (SNP) at ages 9, 10, and 11 years. Further analyses revealed that the association with orthographic skills was specific to the processing of specific components of characters (P values<0.046). No association was found at either SNP of rs3743205 or rs57809907. Our findings suggest that DYX1C1 influences reading development in the general Chinese population and supports a universal effect of this gene.  相似文献
