Protoplast isolation from apple leaf tissue of in-vitro cultured shoots is possible if very young leaves from buds are used. Digestible leaves are found in apple shoots of the subculture kerö 14 days after transferring the shoot and of M26 some days later.The yield of protoplasts is considerably improved if the apple shoots are cultured in a medium containing L-methionine (0.5 mM).  相似文献

A trial of the efficacy of a plain whole cell pertussis vaccine was conducted in Sweden. In this non-blinded trial 525 infants aged 2 months who were born on days with an even number received three doses of vaccine one month apart and 615 infants of the same age who were born on days with an odd number were enrolled as controls. During the 18 months of follow up there were 55 cases of pertussis. The attack rate was 1·5% (8/525) among the vaccinated children and 7·6% (47/615) among the unvaccinated children (p<0·001). The estimated efficacy of the vaccine was 80% (95% confidence interval 58 to 90).The estimated efficacy of pertussis vaccine was similar to that observed in British trials over 30 years ago.  相似文献

Low-temperature fluorescence emission spectra of epicotyls of 6.5-day-old dark-grown seedlings of pea ( Pisum sativum L.) showed the dominance of short-wavelength protoch lorophyllide forms with emission maxima at 629 and 636 nm, respectively. The presence of long-wavelength protochlorophyllide with emission maxima around 650 nm was just detectable. Accordingly, irradiation with millisecond flashes gave a minute formation of chlorophyllide. The chlorophyll(ide) formation varied along the epicotyl. Irradiation with continuous light for 1.5 h resulted in an evident accumulation of chlorophyll(ide) in the upper part of the epicotyl. Only small amounts accumulated in the middle section. The conversion of protochlorophyllide to chlorophyllide was temperature dependent and almost arrested at 0°C. The chlorophyll(ide) formed had one dominating fluorescence peak at 681 nm. Irradiation for 24 h gave almost 100 times more chlorophyll in the upper part of the epicotyl than in the lower part. Electron micrographs from the upper part of the epicotyl irradiated for 6 h showed plastids with several developing thylakoids, while the plastids in the lower part of the epicotyl had only a few thylakoids. The dominance of short-wavelength protochlorophyllide forms indicated the presence of protochlorophyllide not bound to the active site of NADPH-protochlorophyllide oxidoreductase (EC 1.3.1.33). The inability of the short-wavelength form to transform into chlorophyllide with flash light denotes a dislocation from the active site. The time and temperature dependence of the chlorophyll(ide) formation in continuous light indicates that a relocation is required of the short-wavelength protochlorophyllide before chlorophyllide formation can occur.  相似文献

Two so-called Ter sites, which bind the Escherichia coli Tus protein, are located near the replication origin of plasmid R1. Inactivation of the tus gene caused a large decrease in the stability of maintenance of the R1 mini-derivative pOU47 despite the presence of a functional partition system on the plasmid. Deletion of the right Ter site caused a drop in stability similar to that observed after inactivation of the tus gene. Substitution of 2 bp required for Tus binding also caused unstable plasmid maintenance, whereas no effects on stability were observed when the left Ter site was deleted. Inactivation of the tus gene was coupled to an increased occurrence of multimeric plasmid forms as shown by gel electrophoresis of pOU47 DNA. Inactivation of the recA gene did not increase plasmid stability, suggesting that the multimerization was not mediated by RecA. Plasmid DNA was isolated from the tus strain carrying plasmid pOU47 and from a wild-type strain carrying pOU47 in which the right Ter site had been inactivated; in both cases, electron microscopy revealed the presence of multimers as well as rolling-circle structures with double-stranded tails. Thus, the right Ter site in plasmid R1 appears to stabilize the plasmid by preventing multimerization and shifts from theta to rolling-circle replication.  相似文献

Histones are abundant cellular proteins but, if not incorporated into chromatin, they are usually bound by histone chaperones. Here, we identify Arabidopsis NASP as a chaperone for histones H3.1 and H3.3. NASP interacts in vitro with monomeric H3.1 and H3.3 as well as with histone H3.1–H4 and H3.3–H4 dimers. However, NASP does not bind to monomeric H4. NASP shifts the equilibrium between histone dimers and tetramers towards tetramers but does not interact with tetramers in vitro. Arabidopsis NASP promotes [H3–H4]2 tetrasome formation, possibly by providing preassembled histone tetramers. However, NASP does not promote disassembly of in vitro preassembled tetrasomes. In contrast to its mammalian homolog, Arabidopsis NASP is a predominantly nuclear protein. In vivo, NASP binds mainly monomeric H3.1 and H3.3. Pulldown experiments indicated that NASP may also interact with the histone chaperone MSI1 and a HSC70 heat shock protein.  相似文献

Ancient Persia witnessed one of its most prosperous cultural and socio-economic periods between 550 bc and ad 651, with the successive domination of the Achaemenid, Seleucid, Parthian and Sassanian Empires. During this period agricultural activities increased on the Iranian plateau, as demonstrated by a remarkable arboricultural expansion. However, available data are not very informative about the spatial organization of agricultural practices. The possible links between climate conditions and agricultural activities during this millennium of continuous imperial domination are also unclear, due to the lack of parallel human-independent palaeoclimatic proxies. This study presents a new late Holocene pollen-based vegetation record from Lake Parishan, SW Iran. This record provides invaluable information regarding anthropogenic activities before, during and after the empires and sheds light on (i) spatial patterning in agricultural activities and (ii) possible climate impacts on agro-sylvo-pastoral practices during this period. Results of this study indicate that arboriculture was the most prominent form of agricultural activity in SW Iran especially during the Achaemenid, Seleucid and Parthian periods. Contrary to the information provided by some Greco-Roman written sources, the record from Lake Parishan shows that olive cultivation was practiced during Achaemenid and Seleucid times, when olive cultivation was significant, at least in this basin located close to the capital area of the Achaemenid Empire. In addition, pollen from aquatic vegetation suggests that the period of the latter centuries of the first millennium bc was characterized by a higher lake level, which might have favoured cultural and socio-economic prosperity.  相似文献

Periampullary adenocarcinoma, including pancreatic cancer, is a heterogeneous group of tumours with dismal prognosis, for which there is an urgent need to identify novel treatment strategies. The human epithelial growth factor receptors EGFR, HER2 and HER3 have been studied in several tumour types, and HER-targeting drugs have a beneficial effect on survival in selected types of cancer. However, these effects have not been evident in pancreatic cancer, and remain unexplored in other types of periampullary cancer. The prognostic impact of HER-expression in these cancers also remains unclear. The aim of this study was therefore to examine the expression and prognostic value of EGFR, HER2 and HER3 in periampullary cancer, with particular reference to histological subtype. To this end, protein expression of EGFR, HER2 and HER3, and HER2 gene amplification was assessed by immunohistochemistry and silver in situ hybridization, respectively, on tissue microarrays with tumours from 175 periampullary adenocarcinomas, with follow-up data on recurrence-free survival (RFS) and overall survival (OS) for up to 5 years. EGFR expression was similar in pancreatobiliary (PB) and intestinal (I) type tumours, but high HER2 and HER3 expression was significantly more common in I-type tumours. In PB-type cases receiving adjuvant gemcitabine, but not in untreated cases, high EGFR expression was significantly associated with a shorter OS and RFS, with a significant treatment interaction in relation to OS (pinteraction = 0.042). In I-type cases, high EGFR expression was associated with a shorter OS and RFS in univariable, but not in multivariable, analysis. High HER3 expression was associated with a prolonged RFS in univariable, but not in multivariable, analysis. Neither HER2 protein expression nor gene amplification was prognostic. The finding of a potential interaction between the expression of EGFR and response to adjuvant chemotherapy in PB-type tumours needs validation, and merits further study.  相似文献

Summary Male guinea pigs were given a single subcutaneous injection of estradiol, which induces formation of Kurloff cells, and serial sections of thymus were examined after 10, 12, 15 and 21 days. Kurloff cells were found in large numbers in lymphatic vessels, both outside the thymus, in the interlobular tissue, at the cortical surface and inside the cortex, suggesting migration via such structures. Large extrathymic or interlobular lymphatics communicated with a previously undescribed thymic structure-the lymphatic centre-surrounded by a marginal sinus. The orientation of lymphatic valves, and the concentration of Kurloff cells within this lymphatic centre at an early time after the administration of estradiol, indicate the existence of an afferent migratory pathway. The different morphology at different times after estradiol suggest that the treatment caused a dynamic remodeling of thymic lymphatic structures.  相似文献
