Summary The ultrastructural localization of acetyl-CoA carboxylase activity was studied in two strains of the ascomycetous fungus Claviceps purpurea differing in the ergot alkaloid synthesis. Mycelia were harvested by centrifugation of saprophytic submerged cultures, fixed in cold 3% glutaraldehyde in 0.05 M cacodylate buffer pH 7.2 and washed repeatedly in the same buffer. The incubation medium of Yates et al. (1969) had to be modified in the molarity of ATP. The best results were obtained with a medium of the following composition: 50 mM cacodylate buffer pH 7.2, 4 mM ATP, 3.5 mM lead nitrate, 13.5 mM sodium citrate, 3.75 mM sodium bicarbonate, 1.25 mM manganese chloride, 0.4 mM acetyl-CoA and 2 mM biotin. The fixation is a prerequisite for a distinct localization. The enzyme activity was detected only in cells producing high amount of clavine alkaloids. It was confined to the membranes of endoplasmic reticulum and their derivatives: tonoplast of vacuoles, tiny vesicles and amorphous material inside vacuoles. The reaction product was very fine and localized in both leaflets of the membranes. The specificity of the reaction was confirmed by negative results in control preparations: boiled cells incubated in the complete medium, cells incubated in the medium supplemented with avidin or in the media from which either ATP, or acetyl CoA, or sodium bicarbonate, or biotin were omitted. It is suggested that the activity of acetyl-CoA carboxylase is linked to the synthesis of clavine alkaloid precursors which occurs in the endoplasmic reticulum and its derivatives.With technical assistance of J. Barley embryos were treated by 0.1% colchicine for 30 min. Samples of root tips were fixed after 4 h, 8 h and 12 h. In the first sample,c-metaphases, normal metaphases and anaphases were present jointly. Inc-metaphases, chromosomes sometimes tended to make two groups with 7 chromosomes in each. In anaphases, lagging chromosomes, tripolar and multipolar anaphasos were found. No chromosomal aberrations were detected in anaphases and metaphases. The influence of the mitotic organizing centers, the kinetochores and the polar organizers, in controlling the dynamic spindle form and function has been investigated in the primary spermatocytes of two grasshoppers, Arphia xanthoptera and Melanoplus differentialis. A new measure of the total birefringent material in the spindle is introduced—volume-birefringence. This measure avoids many of the problems associated with the traditional retardation measurements of spindle organization.—The number of chromosomes (and their kinetochores) in a spindle can be altered with a piezoelectric micromanipulator in three ways: 1) chromosomes can be removed permanently from the cell, 2) chromosomes can be detached from the spindle and allowed to reenter the spindle at a later time, and 3) chromosomes can be transferred from one spindle to another in cells containing two spindles. Such operations show the volume-birefringence of the spindle is proportional to the number of chromosomes in the spindle. A residual volume-birefringence is seen and attributed to the contribution of the polar organizers to spindle structure. The relative polar contribution differs in the two species. Chromosome motion and spindle elongation in anaphase are unaffected by the number of chromosomes in the spindle. The proportion of volume-birefringence associated with a kinetochore is used to estimate the number of microtubules one might expect to see if the birefringence of the spindle is of microtubular origin. These calculations predict about twice the number of microtubules per kinetochore than seen with the electron microscope. Reasons are suggested to explain this discrepancy.— It is argued that chromosome detachment releases spindle component subunits into the total subunit pool, but that these excess subunits do not influence the metaphase form nor the anaphase function of the spindle; therefore, spindle dynamics are under the direct control of the kinetochores and the polar organizing centers. 相似文献

Adiaspiromycosis was identified in 6 animals in the examination of the lungs of 90 large carnivores. Emmonsia crescens (Chrysosporium parvum var. crescens) was demonstrated as the causative agent in 5 cases of disease — in the badger (Meles meles), the otter (Lutra lutra) and the fox (Vulpes vulpes). E. parva was demonstrated in the remaining case of disease in a fox. The badger is a new, up to the present unknown host of E. crescens. The sporadic occurrence of adiaspiromycosis in the fox and the otter classifies this disease among rare diseases of these animals. 相似文献

The paper deals with the problem of the significance of Ca2+ for the growth of algae, its participation in the process of nitrate and ammonium nitrogen utilization, and with the possibility of substituting Sr2+ for it. Summary Uptake and incidence of microperoxidase (Sigma) in the rat ova during cleavage at the 1-, 2-, 8-cell and blastocyst stages were studied after 30 min incubation with the enzyme (molecular weight of 1,900 and size of molecule of 2 nm).Evidence was furnished of the presence of a small amount of reaction product of microperoxidase in the zona pellucida and its local concentration in some parts of the perivitelline space. The larger part of surface of the ovum, however, was free of microperoxidase. In the cytoplasm microperoxidase was found in pinocytotic vesicles, less frequently in large vacuoles and starting with the eight-cell stage in secondary lysosomes. The largest amount of microperoxidase was ascertained at the stage of blastocyst, chiefly in the cells of the trophoblast. In the cells of the embryoblast, on the contrary, microperoxidase was found but occasionally. The reaction product of microperoxidase was also present in the intercellular space of ova in negligible amount, and likewise on the side of blastocysts' cavity. Ultrathin sectioning of submerged mycelium of Claviceps purpurea Tul. producing clavine alkaloids revealed yeast-like budding resulting in asexual sporesblastospores. These deciduous spores were born by extended hyphal cells and retained the same ultrastructure of cell organelles. Barley seeds were treated for 3 h at 25°C with 240 mM ethyl methanesulphonate (EMS), washed for 18 h, treated with various concentrations of unbuffered sodium azide (pH 6.7–7.3) for 3 h at 25°C, re-dried to 30% water content and either sown immediately or stored at 25°C for 12 days and then sown. The synergistic action of sodium azide post-treatment has been demonstrated only for the EMS-induced M1 germination reduction, while the EMS-induced M1 sterility and the yield of M2 chlorophyll mutants were unaffected. The ?storage” recovery from EMS-induced mutagenic effects was insensitive to sodium azide post-treatment. The 12 day-seed storage at 25°C brought about an improvement of M1 germination, M1 survival, M1 fertility and a decrease in the amount of M2 mutants, regardless of whether sodium azide post-treatment was applied or not. 相似文献
