The Mar Menor is a sheltered and hypersaline lagoon, with salinity ranges between 38 and 51 psu. The lagoon is threatened by several pressures and in the last decades detrimental impact on the natural community structure and dynamics have increased. In the watershed, agricultural practices are rapidly evolving from extensive dry crop farming to intensively irrigated crops, with increasing loads of nutrient and pollutants to the lagoon. Hydrological conditions, nutrients and chlorophyll a concentrations were analysed in 1997 and 2002–2003 in a grid of 20 stations in the lagoon. Different time scales, from daily to inter-annual, were considered. In the considered periods, the dissolved inorganic nitrogen (DIN) increased whilst phosphate decreased significantly. These contrasting patterns depended upon the increased agricultural loading for DIN and were due to the implementation of the wastewater works for phosphates. In 1997 and 2002, the highest nitrate concentrations were usually found on the west coast of the lagoon, close to the mouths of the main watercourses. In parallel, the lowest concentrations were detected at the inner coastline along “La Manga” sandy bar and “El Estacio” channel. Based on weekly data, correlations between chlorophyll a concentrations and environmental variables disagreed with traditional eutrophication models. Relationships between chlorophyll a and nutrients were negative, suggesting that in the short term phytoplankton controlled nutrient concentrations. Moreover, nitrate and phosphorous seemed to alternate as limiting factors. The relationships between chlorophyll a became positive when considering time lags and analysed at longer time scales (monthly or seasonal means), thus suggesting a very rapid response of primary producers to nutrient enrichment. A significant correlation between chlorophyll a concentration and fish larvae density was also found at all time scales analysed, suggesting a top-down control of the trophic web.  相似文献

BnSP-7 and BnSP-6, two Lys49-phospholipase A2 isolated from Bothrops neuwiedi pauloensis snake venom, were co-crystallized with alpha-tocopherol and X-ray diffraction data were collected for both complexes (2.2 and 2.6 A). Incubation of heat-denatured plasma from the rattlesnake Crotalus atrox with trypsin generated a bradykinin (BK) that contained two amino acid substitutions (Arg1 --> Val and Ser6 --> Thr) compared with mammalian BK. Bolus intra-arterial injections of synthetic rattlesnake BK (0.01-10 nmol/kg) into the anesthetized rattlesnake, Crotalus durissus terrificus, produced a pronounced and concentration-dependent increase in systemic vascular conductance (Gsys). This caused a fall in systemic arterial blood pressure (Psys) and an increase in blood flow. Heart rate and stroke volume also increased. This primary response was followed by a significant rise in Psys and pronounced tachycardia (secondary response). Pretreatment with N(G)-nitro-L-arginine methyl ester reduced the NK-induced systemic vasodilatation, indicating that the effect is mediated through increased NO synthesis. The tachycardia associated with the late primary and secondary response to BK was abolished with propranolol and the systemic vasodilatation produced in the primary phase was also significantly attenuated by pretreatment, indicating that the responses are caused, at least in part, by release of cathecholamines and subsequent stimulation of beta-adrenergic receptors. In contrast, the pulmonary circulation was relatively unresponsive to BK.  相似文献

Various chromosomal banding techniques were utilized on the catfish, Iheringichthys labrosus, taken from the Capivara Reservoir. C-banding regions were evidenced in telomeric regions of most of the chromosomes. The B microchromosome appeared totally heterochromatic. The restriction endonuclease AluI produced a banding pattern similar to C-banding in some chromosomes; the B microchromosome, when present, was not digested by this enzyme and remained stained. G-banding was conspicuous in almost all the chromosomes, with the centromeres showing negative G-banding. When the restriction endonuclease BamHI was used, most of the telomeres remained intact, while some centromeres were weakly digested. The B chromosome was also not digested by this enzyme. The first pair of chromosomes showed a pattern of longitudinal bands, both with G-banding and BamHI; this was more evident with G-banding. This banding pattern can be considered a chromosomal marker for this population of I. labrosus.  相似文献

Contamination of meat products with food-borne pathogens usually results from the carcass coming in contact with the feces of an infected animal during processing. In the case of Salmonella, pigs can become colonized with the organism during transport and lairage from contaminated trailers and holding pens, resulting in increased pathogen shedding just prior to processing. Increased shedding, in turn, amplifies the likelihood of carcass contamination by magnifying the amount of bacteria that enters the processing facility. We conducted a series of experiments to test whether phage therapy could limit Salmonella infections at this crucial period. In a preliminary experiment done with small pigs (3 to 4 weeks old; 30 to 40 lb), administration of an anti-Salmonella phage cocktail at the time of inoculation with Salmonella enterica serovar Typhimurium reduced Salmonella colonization by 99.0 to 99.9% (2- to 3-log reduction) in the tonsils, ileum, and cecum. To test the efficacy of phage therapy in a production-like setting, we inoculated four market-weight pigs (in three replicates) with Salmonella enterica serovar Typhimurium and allowed the challenged pigs to contaminate a holding pen for 48 h. Sixteen naïve pigs were randomly split into two groups which received either the anti-Salmonella phage cocktail or a mock treatment. Both groups of pigs were comingled with the challenged pigs in the contaminated pen. Treatment with the anti-Salmonella phage cocktail significantly reduced cecal Salmonella concentrations (95%; P < 0.05) while also reducing (numerically) ileal Salmonella concentrations (90%; P = 0.06). Additional in vitro studies showed that the phage cocktail was also lytic against several non-Typhimurium serovars.The U.S. Centers for Disease Control and Prevention report approximately 40,000 culture-confirmed cases of salmonellosis each year in the United States, which result in approximately 400 deaths (5). Many Salmonella outbreaks are associated with meat and poultry (20), with contamination usually resulting from the carcass coming into contact with the feces of a Salmonella-infected animal during processing (22).There is an association between pork products and Salmonella, as swine are generally considered to be the second largest reservoir of the organism among food animals after poultry. Although infections in adult swine are normally asymptomatic, once colonized, pigs can shed the organism in the feces for weeks and sometimes months (7).While a great deal of research has been done on developing on-farm anti-Salmonella intervention strategies, these methods are confounded by the fact that Salmonella prevalence in pigs often increases once the animals leave the farm as a result of (i) stress-induced reactivation of preexisting infections (14), (ii) new infections from contaminated transport trailers and processing facility holding pens (12, 15, 24, 31), or (iii) both. Consequently, animals with no history of previous Salmonella infection can begin shedding the organism just prior to processing, which is highly problematic in terms of food safety.We hypothesized that phage therapy could be developed as an effective means to counteract transport- and lairage-associated increases in Salmonella colonization in swine. Phage therapy has the advantage of being natural, nontoxic, and relatively inexpensive and could be used just prior to slaughter, unlike many antibiotics (18, 28). Here we describe a series of experiments demonstrating that treating market-weight pigs with an anti-Salmonella phage cocktail prior to their comingling with Salmonella-infected pigs in a highly contaminated environment resulted in reductions in Salmonella colonization. We further show that the phage cocktail could be effectively microencapsulated, making feed or water delivery possible.  相似文献

Traditional medicine provides strong guidance for scientific experiments involving plant products used by the Brazilian people. The species "cipó-cravo" (Tynnanthus fasciculatus) is a plant commonly used either to combat indigestion and stomach aches, or as a general stimulant and aphrodisiac. In this study, the effects of "cipó-cravo" infusion were investigated within the testicular parenchyma of adult Wistar rats. Rats were divided into 3 groups: a control (distilled water) and two treated groups, which received the plant infusion (100 and 200mg/animal/day). The 200mg dose promoted a significant increase of the testicular parenchyma weight and of the volume and total length of the seminiferous tubules, as well as in total daily sperm production and sperm production per gram of testis.  相似文献

In this article, we describe a novel methodology to extract semantic characteristics from protein structures using linear algebra in order to compose structural signature vectors which may be used efficiently to compare and classify protein structures into fold families. These signatures are built from the pattern of hydrophobic intrachain interactions using Singular Value Decomposition (SVD) and Latent Semantic Indexing (LSI) techniques. Considering proteins as documents and contacts as terms, we have built a retrieval system which is able to find conserved contacts in samples of myoglobin fold family and to retrieve these proteins among proteins of varied folds with precision of up to 80%. The classifier is a web tool available at our laboratory website. Users can search for similar chains from a specific PDB, view and compare their contact maps and browse their structures using a JMol plug-in.  相似文献

According to life-history theory, reproductive investments involve costs in terms of growth, future fecundity, and/or survival. However, studies to date have often failed to detect costs of reproduction, with survival costs among the less documented. We investigated the cost of reproduction in Helianthemum squamatum (Cistaceae), a short-lived perennial of semiarid Mediterranean environments. After experimental flower removal, we evaluated next season's growth, reproduction, and survival of the plants. We also monitored an indicator of plant physiological status (F(v)/F(m)) and leaf nutrient concentration at key phenological stages during reproduction. Survival rate in deblossomed plants was significantly higher than in control plants. As far as we know, this is the first experimental evidence of a survival cost of reproduction in a perennial plant. In contrast, no cost to growth or reproduction was found during the next season, and no significant differences in F(v)/F(m) or leaf nutrients were found between control and deblossomed plants. Helianthemum squamatum's success in semiarid Mediterranean ecosystems seems to rely on a persistent seed bank, combined with a sustained high reproductive output at the expense of survival. We conclude that this strategy might be more common than previously thought among short-lived shrubby plants growing in stressful Mediterranean areas.  相似文献

Summary A filamentous diatom Melosira italica was collected at the beginning of rainy season from a shallow lake in the tropical savanna region in Brazil. Even the sample taken from surface water contained empty cells in high percentages. The number of cells per filament of M. italica showed a peculiar pulse-like frequency distribution with peak values at 4, 8, 12 and 16. Evidences of the synchronous cell division in this planktonic diatom under natural environment are discussed.  相似文献

Germination of Beauveria bassiana (Bb) and Metarhizium anisopliae (Ma) conidia determined from a fast-rehydration (FR) protocol were compared to those obtained when dry conidia were subjected to slow rehydration (SR) by holding under high humidity conditions prior to aqueous suspension. Differences in viability estimates obtained using the FR vs. SR protocols increased markedly after conidia were exposed to various stress factors in storage (high aw, temperature, and O2 concentrations), with the SR protocol producing higher estimates of viability in all cases. After Bb conidia were stored under moist conditions for 21 days at 25 °C, the SR estimate of viability was >21% greater than the FR estimate. In jars flushed with different O2 concentrations and stored at 50 °C for 34 days, proportional differences between protocols varied, depending on water activity, from 18-44% in jars flushed with 0% O2 (100% N2) to as high as 63-93% when treated with 21-22% O2. For conidia stored over a broad range of moderate to high temperatures in the absence of O2, SR-FR differences were ?9% at 25-40 °C but 30% at 50 °C. Germination of stressed Bb and Ma conidia increased substantially when incubation time on the germination substrate was increased from 24 to 72 h, whereas germination of non-stressed conidia showed little change. Conidia debilitated by stress were characterized by hypersensitivity to lethal imbibitional damage (damage that is mitigated by slow rehydration) and slow germination. Viability protocols that may provide more reliable assessments of overall mycopesticide quality are discussed.  相似文献
