Low frequency of CYP2B deletions in Brazilian patients with congenital adrenal hyperplasia due to 21-hydroxylas deficiency

The frequency of large mutations was determined in 131 Brazilian patients with different clinical forms of 21-hydroxylase deficiency, belonging to 116 families. DNA samples were examined by Southern blotting hybridization with genomic CYP21 and C4cDNA probes after Taql and Bg/II restriction. Large gene conversions were found in 6.6% and CYP21B deletions in 4.4% of the alleles. The breakpoint in these hybrid genes occurred after exon 3 in 92% of the alleles. All rearrangements involving CYP21B gene occurred in the heterozygous form, except in a patient with simple virilizing form who presented homozygous CYP21B deletion. Our data showed that in these Brazilian patients, CYP21B deletions were less frequent than in most of the large series previously reported.     

Platelets induce human umbilical vein endothelial cell proliferation through P-selectin

We studied whether platelets could participate in the endothelial cell monolayer regeneration in the case of a vessel damage. Incorporation of [3H]-thymidine into the DNA of human umbilical vein endothelial cells (HUVECs) was measured after 48 h of co-incubation with platelets. The effect of platelets was compared to that of platelet-free supernatants from thrombin-activated platelets that had secreted their active granule constituents. Platelets dose-dependently induced HUVEC proliferation. Platelets preactivated by thrombin induced similar proliferation as did unactivated platelets (proliferation factor = 7 - 8), indicating that preactivation of platelets was not required. Platelets fixed with paraformaldehyde had no effect, suggesting that the platelet mitogenic effect required a mobile, alive membrane. Ketanserine and suramin reduced by at most 30 % the platelet-induced proliferation; supernatants of thrombin-activated platelets caused only minor proliferation (proliferation factor = 2), suggesting that secreted 5-hydroxytryptamine and growth factors poorly contributed to the proliferative effect. When the co-incubation was performed in the presence of an anti P-selectin antibody, the platelet-induced HUVEC proliferation was inhibited. The results suggest that platelet adhesion participate in the control of the endothelial regeneration and that platelet P-selectin is a molecular determinant of the proliferative signal.     

Distinction of males of the Lutzomyia intermedia (Lutz & Neiva, 1912) species complex by ratios between dimensions and by an artificial neural network (Diptera: Psychodidae, Phlebotominae)

The females of the two species of the Lutzomyia intermedia complex can be easily distinguished, but the males of each species are quite similar. The ratios between the extra-genital and the genital structures of L. neivai are larger than those of L. intermedia s. s., according to ANOVA. An artificial neural network was trained with a set of 300 examples, randomly taken from a sample of 358 individuals. The input vectors consisted of several ratios between some structures of each insect. The model was tested on the remaining 58 insects, 56 of which (96.6%) were correctly identified. This ratio of success can be considered remarkable if one takes into account the difficulty of attaining comparable results using traditional statistical techniques.     

Cadmium chloride-induced oxidative stress in skeletal muscle cells in vitro

The effects of cadmium chloride (CdCl(2)) on oxidative stress in the skeletal muscle cell line C(2)C(12) were investigated. Myoblast cells that differentiated into myotubes were treated with CdCl(2) (1, 3, 5, 7.5, 10, and 12.5 microM) for 24, 48, and 72 h. Subsequent assay of cell homogenates for MTT (3-(4,5-dimethylthiozol-2-yl)-2,5-diphenyltetrazolium bromide) reduction, neutral red uptake and nucleic acid content showed that cadmium was toxic to C(2)C(12) cells in a concentration-dependent manner. Glutathione-S-transferase activity (nmol microg of protein(-1) min(-1)) was increased with 1 and 3 microM CdCl(2) (36.9 +/- 5.6 and 32.1 +/- 6.0, respectively) compared to control cells (21.8 +/- 1.5), but decreased at higher concentrations (7.5 microM = 15.9 +/- 3.3, 10 microM = 15.9 +/- 4.6, and 12.5 microM = 10.5 +/- 2.8). An increase in malondialdehyde content (nmol microg of protein(-1)), especially at high CdCl(2) concentrations (control = 7.3 +/- 0.5; CdCl(2): 7.5 microM = 11.2 +/- 3.1, 10 microM = 14.6 +/- 3.8, and 12.5 microM = 20.5 +/- 6.5) indicated that there was enhanced lipid peroxidation. Light and scanning electron microscopy showed that there was a concentration-dependent loss of adherent cells and the formation of vesicles indicative of cell death. These results indicated that CdCl(2) increased oxidative stress in C(2)C(12) cells, and this stress probably compromised cell adhesion and the cellular antioxidant defense mechanisms.     

Atrial supersensitivity to noradrenaline in stressed female rats

Stress can change the responses to catecholamines in many tissues. The aim of this study was to investigate the influence of the estrous cycle on the sensitivity of right atria to noradrenaline in female rats subjected to acute swimming stress. Female Wistar rats in proestrus, estrus, metestrus or diestrus were submitted to a 50 min-swimming session. Immediately after the exercise, the rats were killed and their right atria were mounted for isometric recording of the spontaneous beating rate. Concentration-effect curves to noradrenaline were obtained before and after the inhibition of neuronal uptake with phenoxybenzamine (10 microM) and of extraneuronal uptake with estradiol (5 microM). Acute swimming stress did not change the right atrial sensitivity to noradrenaline in rats in estrus, metestrus and diestrus. However, swimming stress produced supersensitivity to noradrenaline in proestrus (pD(2) control: 7.14 +/- 0.03 vs. pD(2) swimming: 7.55 +/- 0.04; p<0.05). This supersensitivity was still observed after uptake inhibition. When catecholamine uptake was inhibited, the concentration-effect curve to noradrenaline was shifted to the left 2.5-fold in the proestrus control group and 1.7-fold in the proestrus stress group (p<0.05). In conclusion, the estrous cycle influenced the acute stress-induced atrial supersensitivity to noradrenaline.     

Analysis of fish communities along a rural–urban gradient in a neotropical stream (Piracicaba River Basin,São Paulo,Brazil)

This study investigated the fish fauna of a stream in southeastern Brazil that runs through sugar cane-growing and urban areas that are subject to sewage discharge. It aimed to determine the importance of spatial and environmental factors on fish distributions and to assess the use of fish as indicators of the environmental conditions. A longitudinal pattern of chemical water quality was observed, with more disturbed conditions at the urban sites. Several opportunistic or tolerant and omnivorous species such as Astyanax altiparanae, Poecilia reticulata, and Astyanax fasciatus were numerically abundant at these urban sites, which resulted in a consistent increase in species density and biomass along the stream. Redundancy analysis extracted three axes, identifying a nutrient–productivity gradient associated with different species composition patterns, as also detected by the indicator species analysis. No species could be considered a potential indicator of the agricultural area in the dry season, whereas four different species were judged potential indicators of three urban sites. In order to explain species distribution, variation in species abundance was partitioned into spatial and environmental components. The latter was more important, explaining 25.4% of the variation. We concluded that the fish assemblage of the Guamium stream is undergoing the initial stages of disturbance, with pronounced effects at the urbanized area, but no detectable changes at the sugar cane sites. Therefore, fish assemblages might be used as ecological indicators to detect urban impacts in similar rivers. However, the potential use of fish to diagnose stressors in areas of sugar cane plantations should be complementary to more comprehensive studies aimed at understanding the influence of this crop on freshwater resources.     

Intermediate Tyrosyl Radical and Amyloid Structure in Peroxide-Activated Cytoglobin

We characterized the peroxidase mechanism of recombinant rat brain cytoglobin (Cygb) challenged by hydrogen peroxide, tert-butylhydroperoxide and by cumene hydroperoxide. The peroxidase mechanism of Cygb is similar to that of myoglobin. Cygb challenged by hydrogen peroxide is converted to a Fe⁴⁺ oxoferryl π cation, which is converted to Fe⁴⁺ oxoferryl and tyrosyl radical detected by direct continuous wave-electron paramagnetic resonance and by 3,5-dibromo-4-nitrosobenzene sulfonate spin trapping. When organic peroxides are used as substrates at initial reaction times, and given an excess of peroxide present, the EPR signals of the corresponding peroxyl radicals precede those of the direct tyrosyl radical. This result is consistent with the use of peroxide as a reducing agent for the recycling of Cygb high-valence species. Furthermore, we found that the Cygb oxidation by peroxides leads to the formation of amyloid fibrils. This result suggests that Cygb possibly participates in the development of degenerative diseases; our findings also support the possible biological role of Cygb related to peroxidase activity.     

Comparison of techniques for the measurement of skin temperature during exercise in a hot,humid environment

Exercising or working in a hot, humid environment can results in the onset of heat-related illness when an individual''s temperature is not carefully monitored. The purpose of the present study was to compare three techniques (data loggers, thermal imaging, and wired electrodes) for the measurement of peripheral (bicep) and central (abdominal) skin temperature. Young men and women (N = 30) were recruited to complete the present study. The three skin temperature measurements were made at 0 and every 10-min during 40-min (60% VO₂max) of cycling in a hot (39±2°C), humid (45±5% RH) environment. Data was statistically analyzed using the Bland-Altman method and correlation analysis. For abdominal skin temperature, the Bland-Altman limits of agreement indicated that data loggers (1.5) were a better index of wired than was thermal imaging (3.5), For the bicep skin temperature the limits of agreement was similar between data loggers (1.9) and thermal (1.9), suggesting the both were suitable measurements. We also found that when skin temperature exceeded 35°C, we observed progressively better prediction between data loggers, thermal imaging, and wired skin sensors. This report describes the potential for the use of data loggers and thermal imaging to be used as alternative measures of skin temperature in exercising, human subjects.     

Hysteretic behavior of proprotein convertase 1/3 (PC1/3)

The proprotein convertases (PCs) are calcium-dependent proteases responsible for processing precursor proteins into their active forms in eukariotes. The PC1/3 is a pivotal enzyme of this family that participates in the proteolytic maturation of prohormones and neuropeptides inside the regulated secretory pathway. In this paper we demonstrate that mouse proprotein convertase 1/3 (mPC1/3) has a lag phase of activation by substrates that can be interpreted as a hysteretic behavior of the enzyme for their hydrolysis. This is an unprecedented observation in peptidases, but is frequent in regulatory enzymes with physiological relevance. The lag phase of mPC1/3 is dependent on substrate, calcium concentration and pH. This hysteretic behavior may have implications in the physiological processes in which PC1/3 participates and could be considered an additional control step in the peptide hormone maturation processes as for instance in the transformation of proinsulin to insulin.
