Summary
Clavispora opuntiae was the prevalent yeast associated with the feeding sites ofSigelgaita sp. larvae in the cactusPilosocereus arrabidae. Also associated with this habitat wereCandida sonorensis, Pichia cactophila, Pichia barkeri, Candida sp. A,Geotrichum sp.,Geotrichum sericeum and the yeast like organismsPrototheca zopfii andAcremonium sp. Atypical yeast biotypes that may represent new species ofPichia, Sporopachydermia andCandida were isolated. This article presents a comparative study between different colour models (RGB, HSI and CIEL*a*b*) applied to a very large microscopic image analysis. Such analysis of different colour models is needed in order to carry out a successful detection and therefore a classification of different regions of interest (ROIs) within the image. This, in turn, allows both distinguishing possible ROIs and retrieving their proper colour for further ROI analysis. This analysis is not commonly done in many biomedical applications that deal with colour images. Other important aspects is the computational cost of the different processing algorithms according to the colour model. This work takes these aspects into consideration to choose the best colour model tailored to the microscopic stain and tissue type under consideration and to obtain a successful processing of the histological image. 相似文献

Carex sect. Spirostachyae comprises 25 species displaying the centre of diversity in Eurasia. Phylogenetic analysis of the ITS nrDNA region of 20 species of sect. Spirostachyae, six species of sect. Ceratocystis, five species of sect. Elatae, and eight outgroup species reveals that neither sect. Spirostachyae nor sect. Elatae is monophyletic. With the exclusion of Carex cretica, the 19 species of sect. Spirostachyae studied form a clade with the five species of tropical-subtropical sect. Elatae. Taxonomy of the core Spirostachyae is not only mostly in agreement with our phylogenetic hypothesis, but also with ecological and new cytogenetic results. Two main groups with different chromosome numbers and edaphic preferences are identified in the core Spirostachyae. One includes primarily acidophilous species with high chromosome numbers (2n=(64)68–84), whereas the other one includes mainly basophilous species with lower chromosome numbers (2n=60–74(75)). Chromosome-number variation is extremely different in the core Spirostachyae, showing great stability in some widespread species (e.g. Carex extensa) but an active chromosome evolution – faster chromosomal rearrangements, fusion and fission events than ITS nucleotide substitutions – in more restricted species (e.g. Carex troodi). Biogeography of the two amphiatlantic pairs of species reveals two independent colonizations of South America from the European continent. The geographical barrier of the Strait of Gibraltar has played different roles in the course of evolution of this section, acting as an effective barrier to gene flow in one case (Carex helodes) but as a limited barrier or recent separation in two others (Carex distans, Carex punctata). 相似文献

This study builds on earlier quantitative ethnobotanical studies to develop an approach which represents local values for useful forest species, in order to explore factors affecting those values. The method, based on respondents ranking of taxa, compares favourably with more time-consuming quantitative ethnobotanical techniques, and allows results to be differentiated according to social factors (gender and ethnic origin), and ecological and socio-economic context. We worked with 126 respondents in five indigenous and five immigrant communities within a forest-dominated landscape in the Peruvian Amazonia. There was wide variability among responses, indicating a complex of factors affecting value. The most valued family is Arecaceae, followed by Fabaceae and Moraceae. Overall, fruit and non-commercialised construction materials predominate but women tend to value fruit and other non-timber species more highly than timber, while the converse is shown by men. Indigenous respondents tend to value more the species used for fruit, domestic construction and other NTFPs, while immigrants tend to favour commercialised timber species. Across all communities, values are influenced by both markets and the availability of the taxa; as the favourite species become scarce, others replace them in perceived importance. As markets become more accessible, over-exploitation of the most valuable species and livelihood diversification contribute to a decrease in perceived value of the forest. 相似文献

During the course of radical oxidation, cholesterol may exert seemingly contradictory effects. In order to gain a better understanding of the relationship between cholesterol levels and membrane susceptibility to oxidative damage induced by reactive oxygen species (ROS), here we analyze the integrity and structural stability of cholesterol-modified (enriched or depleted) and unmodified (control) erythrocytes exposed to tert-butyl hydroperoxide. The oxidant significantly increased ROS production, with almost complete oxidation of hemoglobin and a reduction in GSH content in the different erythrocyte groups at 2 mM concentration. These changes were accompanied by losses of cholesterol and total phospholipids, the main decreases being in phosphatidylethanolamine and phosphatidylcholine. The highest lipid loss was found in the cholesterol-depleted group. Fatty acid analyses revealed changes only in peroxidized cholesterol-modified erythrocytes, with decreases in linoleic and arachidonic acids. Fluorescence anisotropy studies showed an increase in the fluidity of the negatively charged surface of peroxidized control erythrocytes. Increased hemolysis and a positive correlation between cellular osmotic fragility and malondialdehyde contents were found in all peroxidized groups. These findings provide evidence that the modification of cholesterol levels in the erythrocyte membrane has provoking effects on peroxidation, with corresponding increases in oxidative damage in the treated cell, possibly as a consequence of lipid bilayer destabilization. 相似文献

To investigate the relationship between P-glycoprotein (Pgp), glutathione S-transferase π (GST-π) and topoisomerase II (Topo II) expression and human gastric cancer chemoresistance in vitro. Primary single-cell suspensions were prepared from fresh specimens of primary gastric cancer and exposed to hydroxycamptothecin (HCPT), cisplatin (CDDP), 5-fluorouracil (5-FU), adriamycin (ADM) and mitomycin (MMC) for 48 h. Cell metabolic activity and rate of inhibition were evaluated using tetrazolium (MTT) assay. Pgp, GST-π and Topo II expression was determined in gastric carcinoma tissue samples using immunohistochemistry. Chemosensitivity of the gastric cancer cells varied; the rates of inhibition of cells exposed to HCPT, CDDP and 5-FU were significantly higher than that of cells exposed to ADM and MMC (p?0.05). Gastric cancer cells with Pgp expression were resistant to ADM and HCPT (p?=?0.008 and p?=?0.011, respectively). Cells resistant to 5-FU, CDDP and MMC had significantly higher GST-π expression (p?0.05). Topo II expression was significantly lower in cells resistant to HCPT, ADM and MMC (p?0.05). Pgp and GST-π expression may contribute to primary resistance of gastric cancer cells to some chemotherapeutic drugs, while Topo II expression may indicate HCPT, ADM and MMC sensitivity. Pgp, GST-π and Topo II detection and the MTT assay could be used as predictors in chemotherapeutic drug administration and for identifying drug resistance in gastric carcinoma. The present study evaluated the susceptibility of three different batches of whiteleg shrimp Litopenaeus
vannamei from Mexico to an inoculum of infectious hypodermal and haematopoietic necrosis virus (IHHNV). Each of the three shrimp batches came from a different hatchery. Because of their origin, it was possible that the genetic makeup of these batches was different among each other. The three batches tested showed differences in IHHNV susceptibility. Here, susceptibility is defined as the capacity of the host to become infected, and it can be measured by the infectivity titer. Susceptibility to IHHNV was observed in decreasing order in shrimp from batch 1 (hatchery from El Rosario, Sinaloa), batch 3 (hatchery from Nayarit) and batch 2 (hatchery from El Walamo, Sinaloa), respectively. The largest susceptibility difference between batches was 5012 times, and that between early and late juveniles from the same batch was 25 times. These results indicate that within a species, susceptibility to a pathogen such as IHHNV can have large differences. Susceptibility to pathogens is an important trait to consider before performing studies on pathogenesis. It may influence virological parameters such as speed of replication, pathogenicity and virus titer. In order to evaluate the potential use of IHHNV as a natural control agent against white spot syndrome virus (WSSV), it is necessary to know host susceptibility and the kinetics of IHHNV infection. These features can help to determine the conditions in which IHHNV could be used as antagonist in a WSSV infection. 相似文献

The holocentric structure promotes chromosome rearrangements by fission, fusion, translocation, and inversion, which have been thought to promote differentiation and speciation. The Carex laevigata group (Cyperaceae) comprises four species: two restricted endemics from the western Mediterranean (Carex camposii, 2n = 72, and Carex paulo-vargasii, 2n = 74–75), and two more widespread species, found mostly in Western Europe (C. laevigata, 2n = 69–84, and Carex binervis, 2n = 72–74). We tested the selection for chromosome number by climatic variables by controlling for the non-independence of the data using generalized linear mixed model (GLMM). We obtained chromosome counts as well as DNA sequences for the 5′ trnK intron and the trnV-ndhC intergenic spacer in the chloroplast genome from 181 individuals from 53 populations representing these four species. We also climatically characterized the sites where the 53 populations were found using the WorldClim database. Our results show that the best predictor of chromosome number variation is the climatic environment rather than neutral evolutionary processes like founder events and migration patterns. These results support the adaptive value of the holocentric chromosomes and their role in promoting differentiation and eventually speciation. 相似文献
