Seasonality often influences multiple aspects of vector-borne plant diseases. With respect to the management of plant disease vectors, an understanding of the factors driving seasonal changes in vector flight activity may improve management outcomes by facilitating more proactive application of control measures. One of the most challenging issues for citrus Huanglongbing management is to forecast when disease primary spread by bacteriliferous Diaphorina citri will occur. We monitored the temporal patterns of immigrating psyllids, using upwards of 1,200 traps spread among 9 citrus farms and checked on a weekly basis over 4 years. This dataset was analysed with a set of hierarchical models to estimate the effect of climatic variables on citrus foliage production (i.e. flush), and effects of climatic variables and citrus foliage dynamics on D. citri catches over the season. The results showed substantial seasonal variability in immigrating D. citri abundance, with the critical dispersal/migration period occurring between end of winter and spring. During this period, 65% of the total psyllids were collected on sticky traps. Seasonality of immigrating D. citri coincided with changes in certain climate variables, with negative effects of humidity and daily maximum temperatures, and a positive effect of rainfall amount in prior weeks. Maximum temperature and both daily minimum temperature and rainfall during prior weeks were also associated with new citrus flush production, which itself was positively related to immigrating D. citri abundance. Based on these results, citrus growers should be aware and intensify the frequency of psyllid control tactics (i.e. insecticide and/or kaolin sprays) during this period in order to prevent Ca. L. asiaticus transmission. These results are an important step towards developing the predictive framework needed to refine D. citri and huanglongbing management. 相似文献
Although branched-chain amino acids (BCAA) are commonly used as a strategy to recover nutritional status of critically ill patients, recent findings on their role as immunonutrients have been associated with unfavorable outcomes, especially in obese patients. The present study aimed to explore the effects of different BCAA supplementation protocols in the inflammatory response of LPS-stimulated RAW 264.7 macrophages. Cell cultures were divided into five groups, with and without BCAA supplementation, (2 mmol/L of each amino acid). Then, cell cultures followed three different treatment protocols, consisting of a pretreatment (PT), an acute treatment (AT), and a chronic treatment (CT) with BCAA and LPS stimulation (1 µg/mL). Cell viability was analyzed by MTT assay, NO production was assessed by the Griess reaction and IL-6, IL-10, TNF-α and PGE2 synthesis, was evaluated by ELISA. BCAA significantly increased cell viability in AT and CT protocols, and NO and IL-10 synthesis in all treatment protocols. IL-6 synthesis was only increased in PT and CT protocols. TNF-α and PGE2 synthesis were not altered in any of the protocols and groups. BCAA supplementation was able to increase both pro and anti-inflammatory mediators synthesis by RAW 264.7 macrophages, which was influenced by the protocol applied. Moreover, these parameters were significantly increased by isoleucine supplementation, highlighting a potential research field for future studies.
Maximizing biodiversity persistence in heterogeneous human-modified landscapes is hindered by the complex interactions between habitat quality and configuration of native and non-native habitats. Here we examined these complex interactions considering avian diversity across 26 sampling sites, each of which comprised of three sampling points located across a gradient of disturbance: core native habitat fragment, fragment edge, and non-native adjacent matrix. The 78 sampling points were further nested within three neotropical biomes—Amazonia, Cerrado and Pantanal—in central-western Brazil. Matrix type consisted of cattle pastures in the Amazon and teak plantations in the Pantanal and Cerrado. We considered the interactive effects of (1) disturbance-context: fragment core, edge and adjacent matrix, (2) matrix type: tree plantation or cattle pastures, both subject to varying land-use intensity, and (3) native habitat configuration (fragment size, shape and isolation) on bird species richness, abundance and composition. Based on point-count surveys, we recorded 210 bird species. Bird species richness and abundance declined across the disturbance gradient, while genus composition only differed within the adjacent matrix, particularly cattle-pastures. The effect of native habitat area was positive but only detected at fragment edges. Overall bird diversity increased at sites characterized by higher availability of either relict trees within pasture landscapes or old-growth trees within teak plantation landscapes. The core of native fragments played a primary role in ensuring the persistence of bird diversity, regardless of fragment size. In contrast to pastures, tree plantations likely harbour a higher proportion of forest-dependent species while bird diversity can be further enhanced by reduced management intensity in both matrix types. Strategies to maximize avian persistence should not only include retaining native habitats, but also maximizing the size of core native habitats. Likewise, more structurally complex matrix types should be encouraged while maintaining low levels of land-use intensity.

Structural heterogeneity and the dynamics of the complexes of enzymes with substrates can determine the selectivity of catalysis; however, fully characterizing how remains challenging as heterogeneity and dynamics can vary at the spatial level of an amino acid residue and involve rapid timescales. We demonstrate the nascent approach of site-specific two-dimensional infrared (IR) spectroscopy to investigate the archetypical cytochrome P450, P450cam, to better delineate the mechanism of the lower regioselectivity of hydroxylation of the substrate norcamphor in comparison to the native substrate camphor. Specific locations are targeted throughout the enzyme by selectively introducing cyano groups that have frequencies in a spectrally isolated region of the protein IR spectrum as local vibrational probes. Linear and two-dimensional IR spectroscopy were applied to measure the heterogeneity and dynamics at each probe and investigate how they differentiate camphor and norcamphor recognition. The IR data indicate that the norcamphor complex does not fully induce a large-scale conformational change to a closed state of the enzyme adopted in the camphor complex. Additionally, a probe directed at the bound substrate experiences rapidly interconverting states in the norcamphor complex that explain the hydroxylation product distribution. Altogether, the study reveals large- and small-scale structural heterogeneity and dynamics that could contribute to selectivity of a cytochrome P450 and illustrates the approach of site-selective IR spectroscopy to elucidate protein dynamics. 相似文献
In southern Brazil, we investigated shark and ray capture records in an integral protection marine protection area (MPA) that allows fishing in that area. We found 10 shark and nine ray species, of which 88.5% and 66.2%, respectively, are endangered. Female adults and neonates of both sexes are abundant in the area, with records of stranding of large specimens, postcapture abortions and ovigerous capsules with fresh embryos on the beach. We suggest the application of continuous fisheries monitoring and an increase in guidelines on the capture of endangered species. 相似文献
