Deficits in auditory processing are among the best documented endophenotypes in schizophrenia, possibly due to loss of excitatory synaptic connections. Dendritic spines, the principal post-synaptic target of excitatory projections, are reduced in schizophrenia. p21-activated kinase 1 (PAK1) regulates both the actin cytoskeleton and dendritic spine density, and is a downstream effector of both kalirin and CDC42, both of which have altered expression in schizophrenia. This study sought to determine if there is decreased auditory cortex PAK1 protein expression in schizophrenia through the use of quantitative western blots of 25 schizophrenia subjects and matched controls. There was no significant change in PAK1 level detected in the schizophrenia subjects in our cohort. PAK1 protein levels within subject pairs correlated positively with prior measures of total kalirin protein in the same pairs. PAK1 level also correlated with levels of a marker of dendritic spines, spinophilin. These latter two findings suggest that the lack of change in PAK1 level in schizophrenia is not due to limited sensitivity of our assay to detect meaningful differences in PAK1 protein expression. Future studies are needed to evaluate whether alterations in PAK1 phosphorylation states, or alterations in protein expression of other members of the PAK family, are present in schizophrenia. 相似文献

The knee is often a site of injury that can often lead to a chronic disease known as osteoarthritis (OA). The disease may be initiated, in part, by acute injuries to joint cartilage and its cells. In a recent study by this laboratory, using Flemish Giant rabbits, an impact compressive load on the tibial femoral joint was shown to cause significant levels of acute damage to chondrocytes in cartilage of the medial and lateral tibial plateaus. In the current study, using the same model, histological and mechanical data from the plateaus were documented at 6 and 12 months post impact, and compared to the unimpacted control limbs and a limb from unimpacted, control animals. The mechanical properties of cartilage were measured with indentation relaxation tests on the medial and lateral plateaus in regions covered and uncovered by the meniscus. The histological studies on impacted limbs showed surface lesions on both plateaus, thickening of the underlying subchondral bone at 12 months and numerous occult microcracks at the calcified cartilage–subchondral bone interface at 6 and 12 months, without significant changes in cartilage thickness or its mechanical properties versus controls. Yet, there was an increase in both the matrix and fiber moduli and a decrease in the permeability of uncovered, medial plateau cartilage in both limbs of impacted animals between 6 and 12 months post impact that was not documented in control animals. 相似文献

The syntheses and comparative studies of the spectral, voltammetry and spectroelectrochemical properties of new manganese phthalocyanine complexes, tetra-substituted with diethylaminoethanethio at the peripheral (complex 3a) and non-peripheral positions (complex 3b) are reported. Solution electrochemistry of complex 3a showed quasi-reversible metal-based (MnIIIPc−2/MnIIPc−2, E1/2 = −0.07 V vs. Ag|AgCl) and ring-based (MnIIPc−2/MnIIPc−3, E1/2 = −0.78 V vs. Ag|AgCl) reductions, but no ring-based oxidation. However, complex 3b showed weak irreversible ring-oxidation signal (Ep = +0.86 vs. Ag|AgCl). Reversible metal-based (MnIIIPc−2/MnIIPc−2, E1/2 = −0.04 V vs. Ag|AgCl) and ring-based (MnIIPc−2/MnIIPc−3, E1/2 = −0.68 V vs. Ag|AgCl) reductions were also observed for complex 3b. Spectroelectrochemistry was used to confirm these processes. Reduction process involving the metal (MnIIIPc−2/MnIIPc−2) was associated with the formation of manganese μ-oxo complex in complex 3a. 相似文献

There have been few studies quantifying litterfall, standing litterstock and gross litter decomposition following forest conversion to plantation crops such as cocoa. Additionally, an assessment of changing processes occurring in forest floor litter systems with plantation age is lacking. We investigated litterfall production, standing litter changes and litter decomposition along a chronosequence of shaded cocoa farm fields (secondary forest, 3, 15 and 30-year-old) in the moist semi-deciduous forest belt in the Ashanti Region of Ghana in West Africa over 24 months. Mean annual litterfall production differed significantly among study sites and ranged from 5.0 to 10.4 Mg DM ha?1. Similarly, standing litter differed significantly between land-use /plot ages. The results showed significant differences in quality between litter from forest and litter from cocoa plantations. Litterfall from forests had higher concentrations of nitrogen and lower concentration of soluble polyphenols and lignin compared to litter from cocoa systems. Annual decomposition coefficients (k L ) were similar in cocoa systems (0.221–0.227) but higher under secondary forests (0.354). Correlations between litter quality parameters and the decomposition coefficient showed nitrogen and lignin concentrations as well as ratios that include nitrogen are the best predictors of decomposition for the litters studied. Our results confirm the hypothesis that decomposition decreases following forest conversion to shaded cocoa systems because of litter quality changes and that decomposition rates correlate to litter quality differences between forest and cocoa ecosystems. The study also showed that standing litter pools and litterfall production in recently converted cocoa plantations are low compared to secondary forests or mature cocoa systems. Management strategies involving the introduction of upper canopy species during plantation development with corresponding replacement of tree mortality with diverse fast growing species will provide high quality and quantity litter resources. 相似文献

In birds, the period spent brooding or guarding young chicks is highly variable, but such variation has seldom been studied. Previous single‐year studies of Antarctic petrels Thalassoica antarctica and grey‐headed albatrosses Thalassarche chrysostoma revealed a pronounced seasonal decline in brood‐guarding duration and gave rise to the ‘synchronisation hypothesis’, which suggests that some of the variation in the length of the brood‐guarding stage is related to predictable seasonal changes in the risk of chick predation. We tested the predictions of this and three other hypotheses in a two‐site, four‐year study of the black‐browed albatross T. melanophris. The existence of a pronounced seasonal decline in brood‐guarding duration was apparent at both sites, and in years of contrasting food availability, providing further support for the ‘synchronisation hypothesis’. Alternative explanations for this pattern are that short brood‐guarding periods for late‐hatched chicks result from a seasonal decline in food availability or from the fact that early nesting birds are of higher individual quality. However, these explanations are at odds with the absence of a seasonal decline in early chick growth or in probability of chick survival. Furthermore, adult quality (measured as past reproductive performance) had a weak and inconsistent effect on the duration of brood‐guarding. Weather changes explained some of the variation in brood‐guarding, but there were no differences between regions of contrasting climates. Individual pairs displayed a degree of inter‐annual consistency in brood‐guarding duration and, at least in some years, longer brood‐guarding resulted in higher fledging probability. We speculate that a higher investment in brood‐guarding increases the cost of reproduction, which counteracts other selective pressures that would otherwise lead to longer brood‐guarding durations. 相似文献

The acalyptrate fly superfamily Opomyzoidea, as currently recognized, is a poorly-known group of 14 families. The composition of this group and relationships among included families have been controversial. Furthermore, the delimitation of two opomyzoid families, Aulacigastridae and Periscelididae, has been unstable with respect to placement of the genera Stenomicra, Cyamops, and Planinasus. To test the monophyly of Opomyzoidea, previously proposed relationships between families, and the position of the three problematic genera, we sequenced over 3300 bp of nucleotide sequence data from the 28S ribosomal DNA and CAD (rudimentary) genes from 29 taxa representing all opomyzoid families, as well as 13 outgroup taxa. Relationships recovered differed between analyses, and only branches supporting well-established monophyletic families were recovered with high support, with a few exceptions. Opomyzoidea and its included subgroup, Asteioinea, were found to be non-monophyletic. Stenomicra, Cyamops, and Planinasus group consistently with Aulacigastridae, contrary to recent classifications. Xenasteiidae and Australimyzidae, two small, monogeneric families placed in separate superfamilies, were strongly supported as sister groups. 相似文献

The human amniotic membrane (HAM) is an abundant and readily obtained tissue that may be an important source of scaffold for
transplanted chondrocytes in cartilage regeneration in vivo. To evaluate the potential use of cryopreserved HAMs as a support
system for human chondrocytes in human articular cartilage repair. Chondrocytes were isolated from human articular cartilage,
cultured and grown on the chorionic basement membrane side of HAMs. HAMs with chondrocytes were then used in 44 in vitro human
osteoarthritis cartilage repair trials. Repair was evaluated at 4, 8 and 16 weeks by histological analysis. Chondrocytes cultured
on the HAM revealed that cells grew on the chorionic basement membrane layer, but not on the epithelial side. Chondrocytes
grown on the chorionic side of the HAM express type II collagen but not type I, indicating that after being in culture for
3–4 weeks they had not de-differentiated into fibroblasts. In vitro repair experiments showed formation on OA cartilage of
new tissue expressing type II collagen. Integration of the new tissue with OA cartilage was excellent. The results indicate
that cryopreserved HAMs can be used to support chondrocyte proliferation for transplantation therapy to repair OA cartilage. 相似文献
