The solubility and reactivity of the Folch-Pi proteolipid from bovine CNS have been studied in reverse micelles of sodium bis(2-ethylhexyl)sulfosuccinate, isooctane, and water. Such a membrane-mimetic system resembles the aqueous spaces of the native myelin sheath in terms of its physicochemical properties. Although the proteolipid is completely insoluble in water, it can be inserted into the water-containing micellar system. In contrast, the lipid-depleted protein failed to be incorporated into these organized assemblies. The lipid requirements for insertion of the proteolipid were studied, therefore, after delipidation by several precipitations with isooctane, a nondenaturing solvent. Novel extraction procedures and quantitative analyses by HPLC of the protein-bound lipids revealed the persistence of a lipid-protein complex (6 +/- 1 mol of lipid/mol of protein) displaying optimal micellar solubilization. Competition experiments carried out with brain lipids provide evidence for a preference of the myelin protein for sulfatide, phosphatidylinositol, and phosphatidylserine, in that order. The resulting proteolipid, although differing in relative composition, showed good solubility in the membrane-mimetic system. In contrast, reconstitution experiments carried out with the lipid-depleted protein resulted in weak lipid binding and poor micellar incorporation. These results suggest that the tightly bound acidic lipids may stabilize a protein conformation required for insertion into the micellar system. 相似文献

- Upon herbivory, plants emit specific herbivore-induced plant volatiles (HIPVs) that can attract natural enemies of the herbivore thus serving as indirect plant resistance. Not only insect herbivores, but microorganisms may also affect HIPV emission before or after plant colonisation, which in turn can affect behaviour of natural enemies of the herbivore. Yet, it remains elusive whether volatiles from microorganisms influence HIPV emission and indirect plant resistance.
- In this study, we investigated whether exposure of Brassica rapa roots to volatiles from soil-borne fungi influence HIPV emission and the recruitment of natural enemies of Pieris brassicae larvae.
- Using a two-compartment pot system, we performed greenhouse and common-garden experiments, and we profiled plant HIPV emission.
- We found that exposure of plant roots to fungal volatiles did not affect the number of P. brassicae larvae recollected from the plants, suggesting a neutral effect of the fungal volatiles on natural predation. Likewise, in a greenhouse, similar numbers of larvae were parasitised by Cotesia glomerata wasps on control plants as on fungal volatile-exposed plants. Additionally, chemical analysis of HIPV profiles revealed no qualitative and quantitative differences between control plants and fungal volatile-exposed plants that were both infested with P. brassicae larvae.
- Together, our data indicate that root exposure to fungal volatiles did not affect indirect plant resistance to an insect herbivore. These findings provide new insight into the influence of indirect plant resistance by fungal volatiles that are discussed together with the effects of fungal volatiles on direct plant resistance.

The paper presents the results of amino acid analyses in xylem sap during leaf regrowth of ryegrass plants defoliated firstly
at the 8th and secondly at the 12th week of culture. The free amino acid composition of leaves, stubble and roots was also
determined and some of the results are reported. Prior to defoliation, xylem sap contained a high proportion of amides, particularly
glutamine. During regrowth after defoliation, the proportion of asparagine in the xylem sap increased until the third day
when the highest ratios of asparagine/glutamine appeared. The results are compared with relative amounts of free amino acids

We describe a protocol for preparing cultures of the PSP (paralytic shellfish poisoning) dinoflagellate Alexandrium minutum, towards subsequent studies of cell adhesion onto artificial substrates. First, phenotypic uniformity of the strain used and reproducibility of the standard growth profile are obtained by optimising key parameters. Batches of A. minutum at mid-exponential proliferation phase are radiolabeled with 33P-containing medium in order to later quantify cell adhesion. A mortality corrective index is applied to the latter, using the vital fluorochrome acridine orange, i.e. dead cells show no nuclear incorporation under epifluorescence microscopy. 相似文献

1. In any group of organisms, one can almost invariably find some species that are ecologically dominant (i.e. disproportionately more abundant and widespread), whereas others are comparatively less prevalent. Understanding of the causes of variation in ecological dominance has been elusive, particularly given that dominant and subordinate species often lack obvious features that could predict their abundance in nature. 2. In this study, physiological, behavioural, morphological, and phylogenetic information is integrated in an effort to understand the mechanisms underlying ecological dominance in ants using the hyperdiverse ant genus Pheidole (Formicidae: Myrmicinae) as a model system. Field estimates of the relative abundance of 10 Pheidole species were compared with potential correlates, which included behavioural (walking velocity), physiological (tolerance to high and low temperatures and desiccation), and morphological traits (body size and degree of dimorphism in the worker caste). A molecular phylogeny of the tested species was also generated to account for potential confounding effects of phylogenetic non‐independence. 3. Dominant Pheidole species were characterised by higher environmental tolerance with respect to temperature and humidity, as well as faster walking speeds. On the other hand, no morphological correlates of ecological dominance were detected. Interestingly, subordinate species showed no evidence of trade‐off in performance, being both more fragile to environmental challenges and slower in their walking speeds. 4. Generalised atrophic benign epidermolysis bullosa (GABEB) is a form of junctional epidermolysis bullosa with a recessive
mode of inheritance. The gene considered likely to be involved in this disease is COL17A1, since in the majority of GABEB
patients the product of that gene, the 180-kD bullous pemphigoid antigen (BP180), is undetectable in skin. We have identified
an intragenic COL17A1 microsatellite marker for which 83% of randomly selected control individuals are heterozygous. We observed
homozygosity for different alleles of this marker in five out of six collagen type XVII-negative GABEB patients of different
European descent. Five of the six COL17A1 alleles of three patients originating from the eastern part of The Netherlands were
identical, as were the haplotypes including flanking markers. The 2342delG mutation was identified in all these five alleles.
This confirms the expectation that due to genetic drift and hidden inbreeding for an autosomal recessive disorder with low
gene frequency, such as collagen type XVII-negative GABEB, most disease alleles from a restricted geographical area will be
“identical by descent”. Our results demonstrate that involvement of a candidate gene can be confirmed by looking for identity
by descent of highly informative intragenic markers.
