Abstract. The Northern Iberian Peninsula is dominated by various types of vegetation from deciduous oak and ash to evergreen oak woodlands. A recent vegetation map of Spain portrays vegetation series which are characterized in terms of their phytogeographic region or bioclimatic (altitudinal) belt. The aim of this paper is to determine whether the areas comprised by both phytogeographic regions (Eurosiberian and Mediterranean) in the study area, as established from the phytogeographic characterization of the vegetation, can be discriminated by climatic variables using multivariate methods, and to compare these with other conventional approaches. In addition, bioclimatic (altitudinal) belts and the main vegetation types were tested for discrimination by climatic variables. Conventional climatic criteria as well as discriminant and principal component analysis were applied to climatic data from 205 meteorological stations for which vegetation information had been taken from the vegetation map. Conventional criteria are good predictors of the phytogeographic division (Mediterranean and Eurosiberian regions) in the study area. Results were improved by multiple discriminant analysis based on climatic data of the dry period of the year (June to September). Both regions in the study area can be predicted with over 95 % accuracy. Using the same multivariate procedure and temperature data the bioclimatic (altitudinal) belts of the study area can be predicted with over 90 % accuracy. The main vegetation groups of the study area can also be predicted with over 80 % accuracy. Ordination analysis supported the results of the discriminant analysis. Empirical models have been generated to predict the phytogeographic- and belt character of any station in the area. The significance of the various periods of the year for discriminating regions and belts is evaluated. The responsiveness to climatic events during the year may be region specific. This study confirms the strong relationship between climate and vegetation in the Northern Iberian Peninsula, particularly regarding the Eurosiberian-Mediterranean boundary.  相似文献

Trypanosoma brucei, a protist responsible for human African trypanosomiasis (sleeping sickness), is transmitted by the tsetse fly where the procyclic forms of the parasite develop in the proline-rich (1–2 mM) and glucose-depleted digestive tract. Proline is essential for the midgut colonization of the parasite in the insect vector, however other carbon sources could be available and used to feed its central metabolism. Here we show that procyclic trypanosomes can consume and metabolize metabolic intermediates, including those excreted from glucose catabolism (succinate, alanine and pyruvate), with the exception of acetate, which is the ultimate end-product excreted by the parasite. Among the tested metabolites, tricarboxylic acid (TCA) cycle intermediates (succinate, malate and α-ketoglutarate) stimulated growth of the parasite in the presence of 2 mM proline. The pathways used for their metabolism were mapped by proton-NMR metabolic profiling and phenotypic analyses of thirteen RNAi and/or null mutants affecting central carbon metabolism. We showed that (i) malate is converted to succinate by both the reducing and oxidative branches of the TCA cycle, which demonstrates that procyclic trypanosomes can use the full TCA cycle, (ii) the enormous rate of α-ketoglutarate consumption (15-times higher than glucose) is possible thanks to the balanced production and consumption of NADH at the substrate level and (iii) α-ketoglutarate is toxic for trypanosomes if not appropriately metabolized as observed for an α-ketoglutarate dehydrogenase null mutant. In addition, epimastigotes produced from procyclics upon overexpression of RBP6 showed a growth defect in the presence of 2 mM proline, which is rescued by α-ketoglutarate, suggesting that physiological amounts of proline are not sufficient per se for the development of trypanosomes in the fly. In conclusion, these data show that trypanosomes can metabolize multiple metabolites, in addition to proline, which allows them to confront challenging environments in the fly.  相似文献

Baculoviruses are arthropod-specific viruses currently used as biological insecticides in several countries of the world. One important factor limiting the efficacy of these bioinsecticides is related to the inactivation of the virus by solar ultraviolet (UV) radiation. This has motivated studies focused on the use of optical brightener compounds that can protect the virus from UV inactivation. The effects of the optical brightener Tinopal C1101 (ethenedyl benzenesulfonic derivative) on the persistance of a nucleopolyhedrovirus (SfMNPV) was quantified in third instar Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae), the principal pest of maize in the Americas. For this, laboratory studies were performed to determine the relationship between virus concentrations and radiation-caused inactivation of SfMNPV alone or in mixtures with 1.25 or 0.1% (vol/vol) Tinopal C1101 using a UV system [radiation was provided by a 15 W UV tube (emission maxima at 312 nm)]. SfMNPV alone was more sensitive to UV than in mixtures with 1.25 or 0.1% Tinopal C1101. For instance, at concentrations of 2.22 x 10(8) and 2.75 x 10(6) occlusion bodies (OBs)/ml, SfMNPV alone-caused mortality was reduced from 88 and 44% (without UV radiation) to 0%, in both cases, after 30 and 15 min exposure to the UV tube, respectively. In contrast, the incorporation of 1.25% Tinopal C1101 into the SfMNPV inoculum at 2.22 x 10(8) and 2.75 x 10(6) OBs/ml caused a mortality of 86.6 and 96.6% after 240 min, respectively. UV-caused inactivation of SfMNPV was directly related to the optical brightener concentration. We conclude that optical brighteners are likely to represent valuable components of nucleopolyhedrovirus formulations.  相似文献

TLR3 recognizes dsRNA and activates antiviral immune responses through the production of inflammatory cytokines and type I IFNs. Genetic association studies have provided evidence concerning the role of a polymorphism in TLR3 (rs3775291, Leu412Phe) in viral infection susceptibility. We genotyped rs3775291 in a population of Spanish HIV-1-exposed seronegative (HESN) individuals who remain HIV seronegative despite repeated exposure through i.v. injection drug use (IDU-HESN individuals) as witnessed by their hepatitis C virus seropositivity. The frequency of individuals carrying at least one 412Phe allele was significantly higher in IDU-HESN individuals compared with that of a matched control sample (odds ratio for a dominant model = 1.87; 95% confidence interval, 1.06-3.34; p = 0.023). To replicate this finding, we analyzed a cohort of Italian, sexually HESN individuals. Similar results were obtained: the frequency of individuals carrying at least one 412Phe allele was significantly higher compared with that of a matched control sample (odds ratio, 1.79; 95% confidence interval, 1.05-3.08; p = 0.029). In vitro infection assays showed that in PBMCs carrying the 412Phe allele, HIV-1(Ba-L) replication was significantly reduced (p = 0.025) compared with that of Leu/Leu homozygous samples and was associated with a higher expression of factors suggestive of a state of immune activation (IL-6, CCL3, CD69). Similarly, stimulation of PBMCs with a TLR3 agonist indicated that the presence of the 412Phe allele results in a significantly increased expression of CD69 and higher production of proinflammatory cytokines including IL-6 and CCL3. The data of this study indicate that a common TLR3 allele confers immunologically mediated protection from HIV-1 and suggest the potential use of TLR3 triggering in HIV-1 immunotherapy.  相似文献

The mangrove oyster Saccostrea palmula coexists with the pleasure oyster Crassostrea corteziensis in coastal lagoons of northwest Mexico. Recent discovery of Perkinsus marinus infecting the pleasure oyster in the region prompted evaluation of S. palmula as an alternative P. marinus host. An analysis to determine the possible presence of P. marinus in natural and cultured populations of S. palmula at four coastal lagoons in Sinaloa, Mexico was carried out during October-November 2010. Tissues from apparently healthy S. palmula were evaluated using Ray's fluid thioglycollate method (RFTM), which revealed a Perkinsus sp. to be present in all four locations at 6.7-20.0% prevalence. Histopathological analysis of these specimens showed tissue alterations and parasite forms consistent with moderate P. marinus infection, which was confirmed by ribosomal non-transcribed spacer (NTS)-based PCR assays on DNA samples from oysters positive by RFTM and histology. DNA sequencing of amplified NTS fragments (307 bp) produced a sequence 98-100% similar to GenBank-deposited sequences of the NTS from P. marinus. Fluorescent in situ hybridization for Perkinsus spp. and P. marinus corroborated the PCR results, showing clear hybridization of P. marinus in host tissues. This is the first record of P. marinus infecting a species from genus Saccostrea and the first record of the parasite from coastal lagoons in Sinaloa, Mexico.  相似文献

Evolutionary ecological theory predicts that among insect herbivores ‘mothers know best’ when selecting a plant to deposit their eggs. Host‐plant selection is usually studied for the adult stage exclusively, although mothers have not always been reported to know best. Here, we investigate the host‐plant selection behaviour of caterpillars, which are considered to be completely dependent on their mothers’ choices. We use a system that offers a biologically relevant framework to compare the degree of participation of adults and juveniles in host‐plant selection. Our results show that neonate Pieris brassicae caterpillars can actively discriminate between conspecific Brassica oleracea plants with or without aphid (Brevicoryne brassicae) infestation. The caterpillars prefer aphid‐infested plants on which their performance is significantly better, while their mothers, the female butterflies, did not discriminate. We compared caterpillar preferences of individuals released individually or in groups, because P. brassicae is a gregarious species. We found that the strength of the preference for aphid‐infested plants was not affected by the degree of grouping. Caterpillar choices were made before contact with the plants, indicating that plant odours were used for orientation. However, the composition of the volatile blends emitted by plants with and without aphids did not show strong differences. Similarly, like with aphid‐infested plants, plants treated with salicylic acid (SA) were also preferred by neonates over untreated control, indicating that the infestation by aphids may have rendered the plants more attractive to the neonates via changes related to interference with JA‐signaling. The main parasitoid of the caterpillars did not discriminate between plants with hosts in the presence or absence of aphids, showing that top–down forces do not influence the relative suitability of the different food sources for the caterpillars. These data are discussed in the context of mothers and offspring having both important, but different roles in the process of host‐plant selection. Butterflies may select the plant species patch, while their offspring adjust and/or update the choices of their mothers at the local scale, within the micro‐habitat selected by the adult.  相似文献

Entamoeba histolytica virulence has been attributed to several amoebic molecules such as adhesins, amoebapores and cysteine proteinases, but supporting evidence is either partial or indirect. In this work we compared several in vitro and in vivo features of both virulent E. histolytica (vEh) and non-virulent E. histolytica (nvEh) axenic HM-1 IMSS strains, such as complement resistance, proteinase activity, haemolytic, phagocytic and cytotoxic capacities, survival in mice caecum, and susceptibility to O2. The only difference observed was a higher in vitro susceptibility of nvEh to O2. The molecular mechanism of that difference was analyzed in both groups of amoebae after high O2 exposure. vEh O2 resistance correlated with: (i) higher O2 reduction ( and H2O2 production); (ii) increased H2O2 resistance and thiol peroxidase activity, and (iii) reversible pyruvate: ferredoxin oxidoreductase (PFOR) inhibition. Despite the high level of carbonylated proteins in nvEh after O2 exposure, membrane oxidation by reactive oxygen species was not observed. These results suggest that the virulent phenotype of E. histolytica is related to the greater ability to reduce O2 and H2O2 as well as PFOR reactivation, whereas nvEh undergoes irreversible PFOR inhibition resulting in metabolic failure and amoebic death.  相似文献

The aim of this study was to determine the in vitro susceptibility of amphotericin B, fluconazole and itraconazole, to several Candida spp recovered from blood cultures on hospitalized patients at the University Hospital of Maracaibo, Venezuela. The determination of the antifungal susceptibility was carried out according to the microdilution method in broth developed by The European Committee for Antimicrobial Susceptibility Testing (EUCAST). The profile of susceptibility of the 74 isolates showed that all the studied species were susceptible to amphotericin B, and 97.2% and 89.2% to fluconazole and itraconazole, respectively.  相似文献

The relationship between heterozygosity at genetic markers (six allozyme and eight microsatellite loci), and fluctuating asymmetry (FA), length and weight was investigated in two samples of Atlantic salmon (Salmo salar L.) with different timings of first active feeding (early (EA) and late (LA) salmon). This trait had previously been related to fitness. EA fish show smaller values of FA, are longer, heavier and are more heterozygous at allozyme loci than are conspecific LA fish. Also within both samples, heterozygosity at allozyme loci was inversely related to FA and was positively related to weight and length. However, no significant differences in microsatellite diversity (heterozygosity and mean d2 measurements) were observed between samples (EA vs LA). Furthermore, no association was observed between the variability at microsatellite loci and FA, weight or length within each sample. These results suggest that allozyme loci, in themselves, influence fitness components, rather than associations arising from associative overdominance.  相似文献
