The aim of this study was evaluate cortical granule (CG) distribution during in vitro maturation (IVM) and fertilisation of prepubertal goat oocytes compared to CG distribution of ovulated and in vitro fertilised oocytes from adult goats. Oocytes from prepubertal goats were recovered from a slaughterhouse and were matured in M199 with hormones and serum for 27 hr. Ovulated oocytes were collected from gonadotrophin treated Murciana goats. Frozen-thawed spermatozoa were selected by centrifugation in percoll gradient and were capacitated in DMH with 20% steer serum for 1 hr. Ovulated and IVM-oocytes were inseminated in DMH medium with steer serum and calcium lactate for 20 hr. Oocytes and presumptive zygotes were stained with FITC-LCA (Lens culinaris agglutinin labelled with fluorescein isothiocyanate) and observed under a confocal laser scanning microscope. Ultrastructure morphology of oocytes and presumptive zygotes were analysed by transmission electron microscopy (TEM). Prepubertal goat oocytes at germinal vesicle stage show a homogeneous CG distribution in the cytoplasm. IVM-oocytes at Metaphase II (MII) and ovulated oocytes presented CGs located in the cortex with the formation of a monolayer beneath to the plasma membrane. At 20 hr postinsemination (hpi), zygotes from IVM-oocytes showed a complete CG exocytosis whereas zygotes from ovulated oocytes presented aggregates of CGs located at the cortical region. Images by TEM detected that CGs were more electrodense and compacts in oocytes from prepubertal than from adult goats.  相似文献

A number of cell types express inducible nitric-oxide synthase (NOS2) in response to exogenous insults such as bacterial lipopolysaccharide or proinflammatory cytokines. Although it has been known for some time that the N-terminal end of NOS2 suffers a post-translational modification, its exact identification has remained elusive. Using radioactive fatty acids, we show herein that NOS2 becomes thioacylated at Cys-3 with palmitic acid. Site-directed mutagenesis of this single residue results in the absence of the radiolabel incorporation. Acylation of NOS2 is completely indispensable for intracellular sorting and .NO synthesis. In fact, a C3S mutant of NOS2 is completely inactive and accumulates to intracellular membranes that almost totally co-localize with the Golgi marker beta-cop. Likewise, low concentrations of the palmitoylation blocking agents 2-Br-palmitate or 8-Br-palmitate severely affected the .NO synthesis of both NOS2 induced in muscular myotubes and transfected NOS2. However, unlike endothelial NOS, palmitoylation of inducible NOS is not involved in its targeting to caveolae. We have created 16 NOS2-GFP chimeras to inspect the effect of the neighboring residues of Cys-3 on the degree of palmitoylation. In this regard, the hydrophobic residue Pro-4 and the basic residue Lys-6 seem to be indispensable for palmitoylation. In addition, agents that block the endoplasmic reticulum to Golgi transit such as brefeldin A and monensin drastically reduced NOS2 activity leading to its accumulation in perinuclear areas. In summary, palmitoylation of NOS2 at Cys-3 is required for both its activity and proper intracellular localization.  相似文献

Gas-phase biofilters used for the treatment of waste gases were originally packed with compost or other natural filter beds containing indigenous microorganisms. Over the past decade much effort has been made to develop new carrier materials, more performant biocatalysts and new types of bioreactors. Elimination capacities reached nowadays are 5 to 10 times higher than those originally reported with conventional compost biofilters. With the recently developed inert filter beds, inoculation is a prerequisite for successful start-up and operation. Either non-defined mixed cultures or pure bacterial cultures have originally been used. The search for efficient fungal biocatalysts started only a few years ago, mainly for the biofiltration of waste gases containing hydrophobic compounds, such as styrene, alpha-pinene, benzene, or alkylbenzenes. In this review, recently isolated new fungal strains able to degrade alkylbenzenes and other related volatile organic pollutants are described, as well as their major characteristics and their use as biocatalysts in gas-phase biofilters for air pollution control. In biofiltration, the most extensively studied organism belongs to the genus Exophiala, although strains of Scedosporium, Paecilomyces, Cladosporium, Cladophialophora, and white-rot fungi are all potential candidates for use in biofilters. Encouraging results were obtained in most of the cases in which some of those organisms were present in gas-phase biofilters. They allow reaching high elimination capacities and are resistant to low pH values and to reduce moisture content.  相似文献

The effects of five plant growth promoting rhizobacteria on the biological nitrogen fixation (BNF), nodulation, and growth promotion of plants of Lupinus albus cv. Multolupa were investigated. The plants were selected for their capacity to use 1‐aminocyclopropane‐1‐carboxylic acid (ACC) as the sole source of nitrogen. Four strains belonged to the genus Pseudomonas (Luc 1, Luc 2, Luc 3, and Luc 4) and one (Luc 5) belonged to the genus Bacillus. Three patterns of inoculation were examined. In the first pattern, PGPRs were inoculated seven days before being inoculated with B. japonicum. In the second pattern, PGPRs and B. japonicum were co‐inoculated, and in the last pattern, PGPRs were inoculated seven days after being inoculated with B. japonicum. The plants were sampled 30 and 45 days after being inoculated with B. japonicum (T1 and T2). In the first pattern of inoculation, Luc 5 significantly increased the biological nitrogen fixation compared to the control at the first sampling time, as did Luc 1 and Luc 4 at the second sampling time. In the second pattern of inoculation, Luc 5 negatively effected the biological nitrogen fixation at both sampling times. In the third pattern of inoculation, all PGPRs caused a decrease in the nitrogen content of the plants compared to the control. The results obtained according to the patterns of inoculation showed that the mechanisms of action of the effects and routes used by Gram‐negative and Gram‐positive strains were clearly different. Competition between PGPRs and B. japonicum, competition for the niches in the rhizoplane, production of auxins, and induction of systemic resistance (ISR) by the production of siderophores or by lipopolysaccharides present in the outer membrane (LPS) are discussed as probable reasons for the effects observed.  相似文献

Francisella tularensis is a gram-negative intracellular bacterium that has been classified as a Category A biothreat because of its ability to induce deadly pneumonic tularemia when inhaled. In the present study, an experimental model of F. tularensis LVS intranasal infection was used to study the immune cells involved in cytokine secretion in the lungs after infection. Dramatic increases in the numbers of cells secreting IFN-gamma were observed 72 h after intranasal infection of BALB/c and C57BL/6 mice with sublethal (1000 CFU) or lethal (10,000 CFU) doses of F. tularensis LVS and the cells primarily responsible for this IFN-gamma expression were identified as CD11b+ DX5+ NK cells. The findings were further confirmed in C57BL/6 mice showing that cells responsible for IFN-gamma secretion in the lungs were CD11b+ DX5+ NK1.1+. NK cell depletion studies showed a decrease in the percentage of IFN-gamma secreting cells, due not only to a diminished proportion of IFN-gamma secreting NK cells, but also to a reduced percentage of T cells secreting IFN-gamma. The results indicate that IFN-gamma is secreted in response to respiratory infection with F. tularensis LVS, and that NK cells are the early responders responsible for IFN-gamma secretion.  相似文献

Contribution of three regions (phosphate-binding, 50’s and 90’s loops) of Anabaena apoflavodoxin to FMN binding and reduction potential was studied. Thr12 and Glu16 did not influence FMN redox properties, but Thr12 played a role in FMN binding. Replacement of Trp57 with Glu, Lys or Arg moderately shifted Eox/sq and Esq/hq and altered the energetic of the FMN redox states binding profile. Our data indicate that the side chain of position 57 does not modulate Eox/sq by aromatic stacking or solvent exclusion, but rather by influencing the relative strength of the H-bond between the N(5) of the flavin and the Asn58-Ile59 bond. A correlation was observed between the isoalloxazine increase in solvent accessibility and less negative Esq/hq. Moreover, Esq/hq became less negative as positively charged residues were added near to the isoalloxazine. Ile59 and Ile92 were simultaneously mutated to Ala or Glu. These mutations impaired FMN binding, while shifting Esq/hq to less negative values and Eox/sq to more negative. These effects are discussed on the bases of the X-ray structures of some of the Fld mutants, suggesting that in Anabaena Fld the structural control of both electron transfer steps is much more subtle than in other Flds.  相似文献

The predaceous mite Amblyseius herbicolus (Chant) is the second most abundant phytoseiid on coffee plants (Coffea arabica L), after Euseius alatus DeLeon, in Lavras, MG, Brazil, associated to the vector of the coffee ring spot virus, Brevipalpus phoenicis (Geijskes) (Acari: Tenuipalpidae). Its life history was studied taking into account biological aspects, life table, predatory activity and functional and numerical responses in relation to the density of the prey. The adult female has longevity of 38 days when supplied with B. phoenicis. The intrinsic rate of population increase (r m) was 0.150 and the mean generation time (T) 25.3 days. The population doubles every 4.6 days. Thirty mites B. phoenicis /3-cm diameter coffee leaf arenas were separately offered to one specimen of each predator phase. Adult females were more efficient in killing all developmental phases of B. phoenicis, followed by the nymph stages. For the functional and numerical responses studies, from 0.14 to 42.3 immature specimens of the prey /cm(2) of arena were submitted to the predator, the preferred phase for predation. Predation and the oviposition of A. herbicolus increased with increasing prey density, with a positive and highly significant correlation. Regression analysis suggests a functional type II response, with a maximum daily predation near 35 B. phoenicis /cm(2) /one adult female.  相似文献

Lizards often respond to increased predation risk by increasing refuge use, but this strategy may entail a loss of thermoregulatory opportunities, which may lead to a loss of body condition. This may be especially important for pregnant oviparous female lizards, because they need to maintain optimal body temperatures as long as possible to maximize developmental embryos rate until laying. However, little is known about how increased time spent at low temperatures in refuges affects body condition and health state of pregnant female lizards. Furthermore, it is not clear how initial body condition affects refuge use. Female Iberian rock lizards forced to increase time spent at low temperatures showed lower body condition and tended to show lower cell-mediated immune responses than control females. Therefore, the loss of thermoregulatory opportunities seems to be an important cost for pregnant females. Nevertheless, thereafter, when we simulated two repeated predatory attacks, females modified refuge use in relation to their body condition, with females with worse condition decreasing time hidden after attacks. In conclusion, female lizards seemed able to compensate increased predation risk with flexible antipredatory strategies, thus minimizing costs for body condition and health state.  相似文献
