Biochemical markers associated with two Mv chromosomes from Aegilops ventricosa in wheat-Aegilops addition lines

Summary The distribution of three biochemical markers, U-1, CM-4 and Aph_v-a, -b, among wheat-Aegilops addition lines carrying M^v chromosomes from Aegilops ventricosa (genomes D^vM^v) has been investigated. Addition lines which had been previously grouped together on the basis of common non-biochemical characters carried marker U-1, a protein component from the 2M urea extract. The added chromosome, in the appropriate genetic background, seems to confer a high level of resistance to the eyespot disease, caused by the fungus Cercosporella herpotrichoides. The other two markers were concomitantly associated with another similarly formed group of addition lines. Both CM-4, a protein component from the chloroform:methanol extract, and Aph_v-a, -b, alkaline phosphate isozymes, have been previously shown to be associated with homoeologous chromosome group 4, which suggests that the added chromosome in the second group of addition lines is 4M^v.     

Trisomy 13 in a 4-year-old child

Summary The karyotype 47,XY,13+ was observed in a mentally retarded four-year-old child, with numerous abnormalities and the typical dermatoglyphics of a trisomy 13. Banding analysis showed a complete extra chromosome 13.     

Severe mental deficiency,proportionate dwarfism,and delayed sexual maturation

Summary Two 46,XY brothers were found to have a previously undescribed syndrome characterized by severe mental deficiency, proportionate dwarfism, and delayed sexual development. A recessive mode of inheritance, either autosomal or X-linked, is assumed.     

Electron-cytochemical localization of phospho(enol)pyruvate carboxylase (EC 4.1.1.31) in fungal cells

Summary New cytochemical method, based on biochemical experiments, was elaborated for the ultrastructural localization of phospho(enol)pyruvate carboxylase (EC 4.1.1.31). The procedure was used to study the saprophytic submerged mycelium of the ascomycetous fungusClaviceps purpurea Tul. producing clavine alkaloids. The pelleted mycelium was fixed in ice cold 3% glutaraldehyde in 50 mM cacodylate buffer pH 7.2 and washed repeatedly in the same cold buffer. The reaction mixture contained 100 mM Tris-HCl buffer pH 9.0, 10 mM phospho(enol)pyruvate, 30 mM sodium potassium tartrate, 3 mM Pb(NO₃)₂, 60 mM MgCl₂ and 30 mM NaHCO₃. Enzyme activity was localized in vacuoles, particularly inside lipid globules (spherosomes) and less frequently in membranous vesicles. Acetyl-CoA activated PEP-carboxylase both in cell free extracts and in the cytochemical staining. Aspartate inhibited the enzyme in the biochemical assay with coupled malate dehydrogenase system; the cytochemical reaction was not influenced, probably due to the interference of asparagine synthase (EC 6.3.1.1).     

Some effects of the induction of gametogenesis in the populations of the homothallic algaChlamydomonas geitleri Ettl

The induction of gametogenesis in the studied homothallic alga by the transfer to a nitrogen-Less medium leads to an adaptation of the cell population until a dynamic equilibrium is attained. Throughout the adaptation, the homeostatic forces regulating the number of the cells in the population assert themselves on the basis of internal nitrogen circulation. Part of the induced cells which perform no conjugation is probably destroyed by lysing, whereas the cells being just in the pre-Induction phase as well as those having passed the phase without being induced to gametes, continue their cycles by means of the released nitrogenous substances. The conditions which in some cells cause the induction of gametogenesis, produce a partial spontaneous synchronisation in other cells. The course of the cell cycles in parts of the culture may be determined according to the pulses of the increasing number of the zygotes within the whole cell population, if the cell cycles have run at a certain degree of synchrony. The culture conditions which induce gametogenesis cause a quick decrease of the biosyntheses of the RNAs while the biosyntheses of the DNAs are almost maintained at the original level. During the dynamic equilibrium of the population, the average content of the RNAs per cell remains very low, whereas the average content of the DNAs per cell virtually does not change (compared with the original state).     

The effect of quaternary ammonium compounds and amine oxides on spores of bacillus cereus

The effect of 1-dodecylpiperidine 1-oxide and N,N’-bis(dodecyldimethyl)-1,2-ethane diammonium dibromide on the spores ofBacillus cereus. particularly their binding to intact spores and spores with reduced cystine bonds, was investigated. The Langmuir type of binding is involved in both cases. Both compounds decreased the thermoresistance of spores. DPNO decreased the fraction of non-germinating spores, the effect of the drug increasing with increasing concentration. This phenomenon was associated with a faster release of dipicolinic acid to the medium. Only microgermination proceeded in the presence of BDED and dipicolinic acid was released only in substantially lower amounts. Both compounds also influenced respiration.
