Perinatal human immunodeficiency virus type 1 (HIV-1) infections cause a broad spectrum of clinical disease and are variable in both the age of the patient at onset of serious disease and the progression of the clinical course. Heterozygotic perinatally infected twins with a marked difference in their clinical courses were monitored during the first 2 years of life. Twin B, the second-born twin, developed AIDS by 6 months of age and died at 22 months of age, while twin A remained minimally symptomatic through the first 2 years. Sequential blood specimens were obtained from the twins in order to characterize the immunologic properties of the children and the phenotypes and genotypes of the HIV-1 isolates at various times. Twin A developed neutralizing antibodies and a high-level antibody-mediated cellular cytotoxicity (ADCC) response, while twin B had no neutralizing antibody and a much lower ADCC response. The virus isolates obtained from the two children at various time points proliferated equally well in peripheral blood mononuclear cells, were nonsyncytium inducing, and could not infect established T-cell lines. They differed in their ability to infect primary macrophages. In parallel to the biological studies, the HIV-1 tat and part of the env gene sequences of the longitudinal isolates at four time points were determined. Sequences of virus from both twins at different time points were highly conserved; the viruses evolved at a similar rate until the last analyzed time point, at which there was a dramatic increase in sequence diversity for the sicker child, especially in the tat gene. Our results show that the viruses isolated at different times do not have significant changes in growth properties. The absence or low levels of neutralizing antibodies may correlate with disease progression in the twins.  相似文献

根据灰色系统关联分析的基本原理,提出了白背飞虱种群动态的加权关联度预测法。衢县早稻后期白背飞虱发生量与历年6月25~30日平均百丛虫量X_1(t)、同期若虫比例X_2(t)、迟熟品种比例X_3(t)、6月下旬水分积分指数X_4(t)和平均气温X_5(t)等因素的关联序为:X_2(t)>X_1(t)>X_3(t)>X_5(t)>X_4(t)。据此建立的加权关联度预测模型,经12年资料回测和试报验证,结果令人满意。  相似文献

对蔷薇属 (Rosa) 38个野生种果实 (以下简称蔷薇果 )的经济性状进行了分析 ,并测定了 VC、VE 和胡萝卜素等重要维生素的含量。蔷薇果 VC 含量在该属种间差异很大 ,以秦岭蔷薇 (R.tsinglingensis)的含量为最高 (2 576mg/ 1 0 0 g) ,德钦蔷薇 (R.deqenensis)的含量为最低 (49mg/ 1 0 0 g)。胡萝卜素含量种间差异明显 ,以软条七蔷薇 (R.henryi)的含量为最高 (1 9.2 4 mg / 1 0 0 g) ,黄刺玫 (R.xanthina)的含量为最低 (0 .0 6 mg/ 1 0 0 g)。 VE 含量种间差异较小 ,在 1 .34 mg/ 1 0 0 g(黄刺玫 )至 3.86 mg/ 1 0 0 g(硕苞蔷薇R.bracteata)之间。对蔷薇亚属 54种野生种果实重要维生素含量的统计分析表明 ,维生素含量与分组具有一定相关性 ,尤以 VC 含量与分组的相关性最为明显 ,桂味组和小叶组 Vc含量很高 (均值都高于 1 80 0mg/ 1 0 0 g) ;合柱组、月季组、木香组和硕苞组含量很低 (均值都在 30 0 mg/ 1 0 0 g以下 ) ,芹叶组除宽刺蔷薇 (R.platyacantha) VC含量很高外 ,其余种类含量都很低 (均值为 1 90 mg/ 1 0 0 g)。胡萝卜素含量与分组也具有一定相关性 ,桂味组、芹叶组、合柱组和硕苞组的胡萝卜素含量较高 ,均值在 6 mg/ 1 0 0 g以上 ;月季组、小叶组和木香组含量较低 ,均值在 0 .4mg/ 1 0 0 g以下。V  相似文献

A study was undertaken to develop a protoplast regeneration system for pinellia. A yield of 19 29 x 105 protoplasts/g F. W. could be obtained from cell suspension cultures incubated in a digestion enzyme solution with 2% cellulase Onzuka R-10, 10% pectinase (Sigma), 0.01% pectolyase Y23. K8P and modified MS media were used to culture protoplasts in: a) liquid, b) liquid-solid double layer, or c) agarose embedded protoplast culture. The former two were conducive to colony formation from protoplast-derived cells. The frequency of cell division was about 8% after 3 days in culture. Gradually adding fresh medium of lower osmotic pressure into the medium for protoplast culture favored cell division. Calli (1–2 mm in diameter) formed after 30–40 days in culture. The calli transferred onto medium supplemented with KT (0.5 mg 1–1) and NAA (0.2 mg 1)–1) could regenerate plants after 40–50 days. RNA (guanine-7-)-methyltransferase is the enzyme responsible for methylating the 5' cap structure of eukaryotic mRNA. The Saccharomyces cerevisiae enzyme is a 436-amino-acid protein encoded by the essential ABD1 gene. In this study, deletion and point mutations in ABD1 were tested for the ability to support growth of an abd1 null strain. Elimination of 109 amino acids from the N terminus had no effect on cell viability, whereas a more extensive N-terminal deletion of 155 residues was lethal, as was a C-terminal deletion of 55 amino acids. Alanine substitution mutations were introduced at eight conserved residues within a 206-amino-acid region of similarity between ABD1 and the methyltransferase domain of the vaccinia virus capping enzyme. ABD1 alleles H253A (encoding a substitution of alanine for histidine at position 253), T282A, E287A, E361A, and Y362A were viable, whereas G174A, D178A, and Y254A were either lethal or severely defective for growth. Alanine-substituted and amino-truncated ABD1 proteins were expressed in bacteria, purified, and tested for cap methyltransferase activity in vitro. Mutations that were viable in yeast cells had either no effect or only a moderate effect on the specific methyltransferase activity of the mutated ABD1 protein, whereas mutations that were deleterious in vivo yielded proteins that were catalytically defective in vitro. These findings substantiate for the first time the long-held presumption that cap methylation is an essential function in eukaryotic cells.  相似文献

Using the two-hybrid method, we isolated a Saccharomyces cerevisiae cDNA encoding a protein homologous to Schizosaccharomyces pombe protein Dis3sp, using as bait, human GTPase Ran. The DIS3 gene is essential for viability and complements S.pombe mutant dis3-54 which is defective in mitosis. Although Dis3sc has no homology to RanBP1, it bound directly to Ran and the S.cerevisiae Ran homologue Cnr1, but not to the S.cerevisiae RCC1 homologue Srm1. Upon binding to Ran with a 1:1 molar ratio, Dis3sc enhanced a nucleotide-releasing activity of RCC1 on Ran. In the presence of Dis3sc, the K(m) of RCC1 on Ran decreased by half, while the kcat was unchanged. In vivo, Dis3sp was present as oligomers of M(r) 670-200 kDa as previously reported, and the 200 kDa oligomer of Dis3sp was found to include Spi1 and Pim1, the S.pombe homologues of Ran and RCC1, respectively. Although the biological function of the heterotrimeric oligomer consisting of Dis3, Spi1 and Pim1 is unknown, our results indicate that Dis3 is a component of the RCC1-Ran pathway.  相似文献

This study documents several alternative approaches for the optimization of the ion-exchange and affinity chromatographic purification of proteins. In these approaches, the chromatographic process has been treated as a four-stage (adsorption, washing, elution, and regeneration) operation. Central to these investigations has been the elaboration of practical iterative procedures based on the use of theoretical models describing each of these stages. Predictions derived from these models have then been evaluated in terms of experimental data obtained using batch adsorption measurements in finite bath configurations and frontal breakthrough measurements with packed beds of different dimensions, containing nonporous and porous adsorbents of different selectivities and capacities for proteins. Commencing with the kinetic and distribution parameters derived from batch equilibrium measurements, the effect of the initial concentration of the target protein, the solid-liquid volume ratio, the superficial velocity and the column dimensions on the pressure drop, production rate, concentration profile, column utilization, and yield have been determined with packed beds. The potential of these iterative approaches to simplify the determination of key mass transfer and interaction parameters required for scale-up and economic optimization of chromatographic purifications of proteins has been examined using ion exchange, immobilized metal ion affinity, and triazine dye pseudo-affinity adsorbents of different selectivity and adsorption capacities. Cnidium officinale Makino is important medicinally and economically, but its origin is uncertain. The phylogenetic relationship ofC. officinale is provided from the analyses based on the ribulose-1,5-bisphosphate carboxylase/oxgenase gene (rbcL) sequences of 41 species which represent the 34 genera of Aplaceae, the four genera of Araliaceae, and one genus each of Pittosporaceae, Cornaceae, and Caprifoliaceae. The strict consensus tree obtained supports a close relationship ofC. officinale to the Chinese members ofLigusticum, especially toL. chuanxiong. Additionally, the tree shows (1) polyphyly of the genusLigusticum and (2) monophyly of the subfamily Apioideae. Within Apioideae, we recognized some groups in our phylogenetic tree. The grouping is discordant in several respects with the traditional tribal divisions based mainly on fruit morphology.  相似文献

本文首次报道了高粱(SorghumvulgareL.)雄性不育系及其保持系和恢复系以及不育系×恢复系杂交F1的C-显带结果,并进行了核型和带型的比较分析。保持系与不育系的核型和带型基本一致,相同的细胞核处于不同的细胞质中,其核型与带型没有明显的变化。三系第2染色体的臂长和臂比差异明显,第2、6和8染色体的带型也不同。父、母本的不同核型和带型特点在F1中为并显性。讨论了染色体多态及其应用的问题。  相似文献

作者采用碱皂化的方法来达到精炼蚕蛹油的目的。实验表明,碱的浓度以10% ̄13%比较合适,超碱量必须视酸值大小,通过小样试验来确定。精制油的过氧化值与皂化后中性油的洗涤温度、洗涤次数及搅拌速度有密切关系,必须严格控制。  相似文献
