The "moubatin-clade" of soft tick lipocalins, although monophyletic, shows clear signs of paralogy as indicated by the various functions associated with this protein family. This includes anti-platelet (moubatin), anti-complement (OMCI) and toxic (TSGP2) activities in the vertebrate host. In order to understand the evolution of function and how it relates to the various paralogs in this clade, we characterized a number of different proteins in regard to undefined function and mechanism. By utilizing gain-of-function for TSGP2 and loss-of-function for TSGP3, we show that inhibition of collagen-induced platelet aggregation by moubatin and TSGP3 is due to scavenging of thromboxane A(2). Moubatin, TSGP2 and TSGP3 are also able to bind leukotriene B(4) with high affinity. TSGP2 and TSGP3, but not moubatin, binds complement C5, with kinetics that indicates that conformation change occurs during interaction. A conserved loop and histidine residue in the sequences of OMCI, TSGP2 and TSGP3 are implicated in the interaction with complement C5. The data presented suggest that the ancestral function evolved in this clade was aimed at inhibition of vasoconstriction, platelet aggregation and neutrophil aggregation, primarily by scavenging of thromboxane A(2) and leukotriene B(4). C5 complement targeting activity evolved within this clade, probably within the Old World Ornithodorinae. The moubatin-clade itself most probably derived from the related histamine and serotonin-binding lipocalin sub-family that is conserved within the Argasidae.  相似文献

The genetic composition of a population reflects several aspects of the organism and its environment. The Icelandic Arctic fox population exceeds 8000 individuals and is comprised of both coastal and inland foxes. Several factors may affect within-population movement and subsequent genetic population structure. A narrow isthmus and sheep-proof fences may prevent movement between the north-western and central part and glacial rivers may reduce movement between the eastern and central part of Iceland. Moreover, population density and habitat characteristics can influence movement behaviour further. Here, we investigate the genetic structure in the Icelandic Arctic fox population ( n  = 108) using 10 microsatellite loci. Despite large glacial rivers, we found low divergence between the central and eastern part, suggesting extensive movement between these areas. However, both model- and frequency-based analyses suggest that the north-western part is genetically differentiated from the rest of Iceland (FST = 0.04, DS = 0.094), corresponding to 100–200 generations of complete isolation. This suggests that the fences cannot be the sole cause of divergence. Rather, the isthmus causes limited movement between the regions, implying that protection in the Hornstrandir Nature Reserve has a minimal impact on Arctic fox population size in the rest of Iceland.  © 2009 The Linnean Society of London, Biological Journal of the Linnean Society , 2009, 97 , 18–26.  相似文献

Preparations that contain well-spread metaphase chromosomes are critical for plant cytogenetic analyses including chromosome counts, banding procedures, in situ hybridization, karyotyping and construction of ideograms. Chromosome spreading is difficult for plants with large and numerous chromosomes. We report here a technique for obtaining cytoplasm-free, well-spread metaphases from two Amaryllidaceae species: Sprekelia formosissima (2n = 120) and Hymenocallis howardii (2n = 96). The technique has three main steps: 1) pretreatment to cause chromosome condensation, 2) dripping onto tilted slides coated with a thin layer of pure acetic acid and 3) application of steam and acetic acid to produce cytoplasmic hydrolysis, which spreads the chromosomes.  相似文献

Nuttalliella namaqua has been described as a “living fossil” and the closest extant species to the ancestral tick lineage. It was previously proposed that the Nuttalliella lineage parasitized reptile-like mammals in the Permian and had to switch hosts several times due to mass or host lineage extinctions. Extant hosts include girdled lizards and murid rodents. The current study extends knowledge on the extant host range of N. namaqua using gut meal analysis of field collected specimens. Nymphs and females can parasitize a variety of reptiles that includes skinks, geckos and girdled lizards. Blood-meal from a hyrax was also detected in a specimen suggesting that N. namaqua could parasitize a broader range of mammals than the previously suggested murid rodents. Rather than being host specific, N. namaqua is proposed to be a generalist and the ability to switch and parasitize multiple hosts allowed it to survive multiple mass and host lineage extinctions.  相似文献

Castellum Fectio was one of the largest fortifications along the Limes, the northern border of the Roman Empire. The castellum, situated 5 km southeast of Utrecht, the Netherlands, was occupied from around the start of our Era to ca. a.d. 260. It was situated along a river bend of the Rhine that was cut off from the main stream during the occupation of the Roman fort. A 6 m long sediment sequence was recovered from the infill of the residual channel and pieces of Roman wall plaster, glume bases of Triticum spelta and radiocarbon dates indicate that the sediments were deposited during the period of Roman occupation. The combined palaeoecological analyses—palynological, macrobotanical, entomological and geochemical—allow a detailed reconstruction of changing environmental conditions as a consequence of the Roman occupation. The pollen record reveals a dramatic decrease in arboreal pollen, suggesting that the Romans were involved in large-scale deforestation, transforming semi-open parkland to a landscape of meadows and agricultural fields. Non-pollen palynomorphs, botanical macrofossils and insect remains support this conclusion. The recorded mycoflora shows a shift from assemblages characterised by the tree pathogen Kretzschmaria deusta to assemblages dominated by spores of fungi associated with herbaceous plants, concurrent with the decrease in arboreal pollen. The presence of masticated bran fragments of cereals, clover remains, eggs of intestinal parasites and entomological and geochemical data in the upper part of the sequence indicates that these sediments largely consist of faeces that were dumped into the former channel. Surprisingly, seeds of salt tolerant species are encountered in the sediments of this inland site, which was situated outside the influence of the sea. Horses may have brought these seeds to Fectio in their intestinal tracts after grazing in coastal meadows.  相似文献
