Congress grass, Parthenium hysterophorus L. (Asteraceae, tribe Heliantheae), is an erect and much branched annual or ephemeral herb, known as a pest for environmental, medical and agricultural reasons. Survey of fungal pathogens of P. hysterophorus was conducted in Kurukshetra and its adjoining areas, Haryana, India. A leaf spot pathogen Alternaria sp. PMK1 was isolated. Koch's postulates were performed and proved that this isolate was pathogenic to this weed. This is the first report of this pathogen causing leaf spot on parthenium weed. 相似文献

Scale insects are important ecologically and as agricultural pests. The majority of scale insect taxa feed exclusively on plant phloem sap, which is carbon rich but deficient in essential amino acids. This suggests that, as seen in the related aphids and psyllids, scale insect nutrition might also depend upon bacterial symbionts, yet very little is known about scale insect-bacteria symbioses. We report here the first identification and molecular characterization of symbiotic bacteria associated with the New Zealand giant scale Coelostomidia wairoensis, using fluorescence in situ hybridization (FISH), transmission electron microscopy (TEM) and 16S rRNA gene-based analysis. Dissection and FISH confirmed the location of the bacteria in large, paired, multilobate organs in the abdominal region of the insect. TEM indicated that the dominant pleomorphic bacteria were confined to bacteriocytes in the sheath-enclosed bacteriome. Phylogenetic analysis revealed the presence of three distinct bacterial types, the bacteriome-associated B-symbiont (Bacteroidetes), an Erwinia-related symbiont (Gammaproteobacteria) and Wolbachia sp. (Alphaproteobacteria). This study extends the current knowledge of scale insect symbionts and is the first microbiological investigation of the ecologically important coelostomidiid scales. 相似文献

Aims
There are few studies on the interactive effect of salinity and sodicity in soils exposed to drying and wetting cycles. We conducted a study to assess the impact of multiple drying and wetting on microbial respiration, dissolved organic carbon and microbial biomass in saline and saline-sodic soils.Methods
Different levels of salinity (EC1:5 1.0 or 2.5) and sodicity (SAR?<?3 or 20) were induced by adding NaCl and CaCl2 to a non-saline/non-sodic soil. Finely ground wheat straw residue was added at 20?g?kg?1 as substrate to stimulate microbial activity. The constant moist (CM) treatment was kept at optimum moisture content for the length of the experiment. The drying and rewetting (DW) treatments consisted of 1 to 3 DW cycles; each DW cycle consisted of 1?week drying after which they were rewet to optimum moisture and then maintained moist for 1?week.Results
Drying reduced respiration more strongly at EC2.5 than with EC1.0. Rewetting of dry soils produced a flush in respiration which was greatest in the soils without salt addition and smallest at high salinity (EC2.5) suggesting better substrate utilisation by microbes in soils without added salts. After three DW events, cumulative respiration was significantly increased by DW compared to CM, being 24% higher at EC1.0 and 16% higher at EC2.5 indicating that high respiration rates after rewetting may compensate for the low respiration rates during the dry phase. The respiration rate per unit MBC was lower at EC2.5 than at EC1.0. Further, the size of the flush in respiration upon rewetting decreased with each ensuing DW cycle being 50–70% lower in the third DW cycle than the first.Conclusions
Both salinity and sodicity alter the effect of drying and rewetting on soil carbon dynamics compared to non-saline soils. 相似文献64.
When mammalian cells experience radiation insult, DNA replication is stalled to prevent erroneous DNA synthesis. UV-irradiation triggers proteolysis of Mcm10, an essential human replication factor, inhibiting the ongoing replication. Here, we report that Mcm10 associates with E3 ubiquitin ligase comprising DNA damage-binding protein, DDB1, cullin, Cul4 and ring finger protein, Roc1. Depletion of DDB1, Roc1 or Cul4 abrogates the UV-triggered Mcm10 proteolysis, implying that Cul4-Roc1-DDB1 ubiquitin ligase mediates Mcm10 downregulation. The purified Cul4-Roc1-DDB1 complex ubiquitinates Mcm10 in vitro, proving that Mcm10 is its substrate. By screening the known DDB1 interacting proteins, we discovered that VprBP is the substrate recognition subunit that targets Mcm10 for degradation. Hence, these results establish that Cul4-DDB1-VprBP ubiquitin ligase mediates the stress-induced proteolysis of replication factor, Mcm10. 相似文献

During breathing, activation of respiratory muscles is coordinated by integrated input from the brain, brainstem, and spinal cord. When this coordination is disrupted by spinal cord injury (SCI), control of respiratory muscles innervated below the injury level is compromised1,2 leading to respiratory muscle dysfunction and pulmonary complications. These conditions are among the leading causes of death in patients with SCI3. Standard pulmonary function tests that assess respiratory motor function include spirometrical and maximum airway pressure outcomes: Forced Vital Capacity (FVC), Forced Expiratory Volume in one second (FEV1), Maximal Inspiratory Pressure (PImax) and Maximal Expiratory Pressure (PEmax)4,5. These values provide indirect measurements of respiratory muscle performance6. In clinical practice and research, a surface electromyography (sEMG) recorded from respiratory muscles can be used to assess respiratory motor function and help to diagnose neuromuscular pathology. However, variability in the sEMG amplitude inhibits efforts to develop objective and direct measures of respiratory motor function6. Based on a multi-muscle sEMG approach to characterize motor control of limb muscles7, known as the voluntary response index (VRI)8, we developed an analytical tool to characterize respiratory motor control directly from sEMG data recorded from multiple respiratory muscles during the voluntary respiratory tasks. We have termed this the Respiratory Motor Control Assessment (RMCA)9. This vector analysis method quantifies the amount and distribution of activity across muscles and presents it in the form of an index that relates the degree to which sEMG output within a test-subject resembles that from a group of healthy (non-injured) controls. The resulting index value has been shown to have high face validity, sensitivity and specificity9-11. We showed previously9 that the RMCA outcomes significantly correlate with levels of SCI and pulmonary function measures. We are presenting here the method to quantitatively compare post-spinal cord injury respiratory multi-muscle activation patterns to those of healthy individuals. 相似文献

In the years 2011–2014, a series of surveys were conducted to record infestation and search natural fungal pathogens of Trianthema portulacastrum in various states of India. Among ten different agricultural important crops, heavy infestation and percentage of occurrence were observed. Gibbago trianthema, phaeodictyconidial genus, causing leaf spot disease was found on horse purslane at different districts of Haryana, Punjab and Uttar Pradesh from infected leaves and stems. Identification was confirmed on the basis of ITS rDNA sequence analysis by Macrogen Inc., Advancing through Genomics, Korea, and from CABI International Mycological Institute, England (IMI Number 502781, 502782). Sequence has been submitted to National Centre for Biotechnology Information (NCBI) with GenBank accession number KJ825852. By in vitro inoculation on Trianthema leaves, the pathogen showed similar symptoms as occurred in nature and proved Koch’s postulates. Killer cell immunoglobulin-like receptors (KIR) are encoded by highly polymorphic genes that regulate the activation of natural killer (NK) cells and other lymphocyte subsets and likely play key roles in innate and adaptive immunity. Association studies increasingly implicate KIR in disease predisposition and outcome but could be confounded by unknown KIR genetic structure in heterogeneous populations. To examine this, we characterized the diversity of 16 KIR genes in 712 Northern Californians (NC) stratified by self-assigned ethnicities and compared the profiles of KIR polymorphism with other US and global populations using a reference database. Sixty-eight distinct KIR genotypes were characterized: 58 in 457 Caucasians (NCC), 17 in 47 African Americans (NCAA), 21 in 80 Asians (NCA), 20 in 74 Hispanics (NCH), and 18 in 54 "other" ethnicities (NCO). KIR genotype patterns and frequencies in the 4 defined ethnicities were compared with each other and with 34 global populations by phylogenetic analysis. Although there were no population-specific genotypes, the KIR genotype frequency patterns faithfully traced the ancestry of NCC, NCAA, and NCA but not of NCH whose ancestries are known to be more heterogeneous. KIR genotype frequencies can therefore track ethnic ancestries in modern urban populations. Our data emphasize the importance of selecting ethnically matched controls in KIR-based studies to avert spurious associations. 相似文献

Background
Cycads are ancient seed plants (living fossils) with origins in the Paleozoic. Cycads are sometimes considered a 'missing link' as they exhibit characteristics intermediate between vascular non-seed plants and the more derived seed plants. Cycads have also been implicated as the source of 'Guam's dementia', possibly due to the production of S(+)-beta-methyl-alpha, beta-diaminopropionic acid (BMAA), which is an agonist of animal glutamate receptors.Results
A total of 4,200 expressed sequence tags (ESTs) were created from Cycas rumphii and clustered into 2,458 contigs, of which 1,764 had low-stringency BLAST similarity to other plant genes. Among those cycad contigs with similarity to plant genes, 1,718 cycad 'hits' are to angiosperms, 1,310 match genes in gymnosperms and 734 match lower (non-seed) plants. Forty-six contigs were found that matched only genes in lower plants and gymnosperms. Upon obtaining the complete sequence from the clones of 37/46 contigs, 14 still matched only gymnosperms. Among those cycad contigs common to higher plants, ESTs were discovered that correspond to those involved in development and signaling in present-day flowering plants. We purified a cycad EST for a glutamate receptor (GLR)-like gene, as well as ESTs potentially involved in the synthesis of the GLR agonist BMAA.Conclusions
Analysis of cycad ESTs has uncovered conserved and potentially novel genes. Furthermore, the presence of a glutamate receptor agonist, as well as a glutamate receptor-like gene in cycads, supports the hypothesis that such neuroactive plant products are not merely herbivore deterrents but may also serve a role in plant signaling.70.
In vitro treatment of erythrocytes of normal and hypercholesterolemic rats with N-nitrosodiethylamine (NDEA), an important carcinogen frequently present in human environment and food chain resulted in a marginal increase in osmotic fragility of erythrocytes without affecting their antioxygenic potential as evidenced by insignificant effect on lipid peroxidation (LPO). However, (LPO) of all the tissues (heart, lung, liver, kidney and spleen) increased significantly on in vitro treatment with NDEA.
