A simple, rapid, novel and sensitive liquid chromatography-tandem mass spectrometry method was developed and validated for quantification of tacrolimus (I) in human plasma, a narrow therapeutic index, potent macrolide immunosuppressive drug. The analyte and internal standard (tamsulosin (II)) were extracted by liquid-liquid extraction with t-butylmethylether using a Glas-Col Multi-Pulse Vortexer. The chromatographic separation was performed on reverse phase Xterra ODS column with a mobile phase of 99% methanol and 1% 10mM ammonium acetate buffer. The deprotonate of analyte was quantitated in negative ionization by multiple reaction monitoring (MRM) with a mass spectrometer. The mass transitions m/z 802.5-->560.3 and m/z 407.2-->151.9 were used to measure I and II, respectively. The assay exhibited a linear dynamic range of 0.05-25ng/ml for tacrolimus in human plasma. The lower limit of quantitation was 50pg/ml with a relative standard deviation of less than 20%. Acceptable precision and accuracy were obtained for concentrations over the standard curve ranges. Run time of 2min for each sample made it possible to analyze a throughput of more than 400 human plasma samples per day. The validated method has been successfully used to analyze human plasma samples for application in comparative bioavailability studies. The tacrolimus plasma concentration profile could be obtained for pharmacokinetic study. The observed maximum plasma concentration (C(max)) of tacrolimus (5mg oral dose) is 440pg/ml, time to observed maximum plasma concentration (T(max)) is 2.5h and elimination half-life (T(1/2)) is 21h. 相似文献

Using spectroscopic, calorimetric and microscopic methods, we demonstrate that calcium binds to beta-2-microglobulin (β2m) under physiological conditions of pH and ionic strength, in biological buffers, causing a conformational change associated with the binding of up to four calcium atoms per β2m molecule, with a marked transformation of some random coil structure into beta sheet structure, and culminating in the aggregation of the protein at physiological (serum) concentrations of calcium and β2m. We draw attention to the fact that the sequence of β2m contains several potential calcium-binding motifs of the DXD and DXDXD (or DXEXD) varieties. We establish (a) that the microscopic aggregation seen at physiological concentrations of β2m and calcium turns into actual turbidity and visible precipitation at higher concentrations of protein and β2m, (b) that this initial aggregation/precipitation leads to the formation of amorphous aggregates, (c) that the formation of the amorphous aggregates can be partially reversed through the addition of the divalent ion chelating agent, EDTA, and (d) that upon incubation for a few weeks, the amorphous aggregates appear to support the formation of amyloid aggregates that bind to the dye, thioflavin T (ThT), resulting in increase in the dye''s fluorescence. We speculate that β2m exists in the form of microscopic aggregates in vivo and that these don''t progress to form larger amyloid aggregates because protein concentrations remain low under normal conditions of kidney function and β2m degradation. However, when kidney function is compromised and especially when dialysis is performed, β2m concentrations probably transiently rise to yield large aggregates that deposit in bone joints and transform into amyloids during dialysis related amyloidosis. 相似文献

The antiporters, uniporters and symporters are the functional classes of MFS that play major role in ions homeostasis, regulation of pumps and channels, membrane structure, transporters activity in tolerance to abiotic stresses. Major facilitator superfamily (MFS) encodes Na+/H+ antiporter that are considered as being sensors of the molecule transports. A large number of MFS proteins have been identified in several plants, rice, maize, Arabidopsis etc. However, the majority of proteins in sorghum are described as putative, uncharacterized till date. This suggested that identified proteins of MFS in sorghum are far from saturation. Hence, we developed gene ontology (GO) terms semantic similarity based method using GOSemSim measure of R package. As a result, total 2,568 high (100 %) semantic similar orthologous proteins from 7 plant species were obtained. These data were used to predict function of 257 putative uncharacterized proteins from 18 families of MFS in Sorghum. Consequently, the identified proteins belonged to the function of regulation of pumps and channels, membrane structure, transporters activity, ions homeostasis, transporter mechanisms and binding process. These identified functions appear to have a distinct mechanism of salt-stress adaptation in plants. The use of plant growth regulators is well established and they are used in many fields of plant science for enhancing growth. Brassica juncea plants were treated with 2.5, 5.0 and 7.5 μM auxin indole-3-butyric acid (IBA), which promotes rooting. The IBA-treated plants were also sprayed with 100 μM gibberellic acid (GA3) and kinetin (Kin) to increase leaf-foliage. Gold (I) chloride (AuCl) was added to the growth medium of plants to achieve required gold concentration. The solubilizing agent ammonium thiocyanate (1 g kg?1) (commonly used in mining industries to solubilize gold) was added to the nutrient solution after six weeks of growth and, two weeks later, plants were harvested. Plant growth regulators improved shoot and root dry biomass of B. juncea plants. Inductively Coupled Plasma Optical Emission Spectrometry analysis showed the highest Au uptake for plants treated with 5.0 μM IBA. The average recovery of Au with this treatment was significantly greater than the control treatment by 45.8 mg kg?1 (155.7%). The other IBA concentrations (2.5 and 7.5 μM) also showed a significant increase in Au uptake compared to the control plants by 14.7 mg kg?1 (50%) and 42.5 mg kg?1 (144.5%) respectively. A similar trend of Au accumulation was recorded in the roots of B. juncea plants. Natural products have been a great source of pharmaceuticals since ages. Vast screening of natural products from different sources has led to the discovery of plethora of chemotherapeutic drugs and other compounds for the betterment of human life. Several bioactive entities have been generated by the structural modifications of the natural products or by using the natives as key models in synthetic chemistry. Nonetheless, a number of natural compounds with potential bioactivities remain unexploited in the medicinal field due to their stringent chemical properties. Andrographis paniculata Nees., a traditional medicinal herb from family Acanthaceae is known for its multiple pharmacological activities. It’s major bioactive constituent “andrographolide”, possesses promising anticancer potential and is one such unexploited treasure. The architecture of the molecule consists of an α-alkylidene γ-butyrolactone moiety, two olefin bond [Δ8(17) and Δ12(13)], three hydroxyls at C-3, C-19, and C-14 and highly substituted trans decalin. Of the three hydroxyl groups, one is allylic at C-14, and the others are secondary and primary at C-3 and C-19, respectively. By modification of the above structural features a number of andrographolide derivatives have been synthesized. The intricacy of the molecule has always been a constraint in developing a commercialized drug, nevertheless the efforts in this direction via synthetic chemistry are still continuous and prominent. The present review highlights the chemistry and anticancer activity of andrographolide. It discusses the limitations of the molecule as a pharmacological agent. Modifications in the key molecule along different moieties has been discussed which might lead to desirable bioactive molecules. The compiled information will be helpful in further developing specific modifications in andrographolide moiety which will have significant contribution in semi synthesis of anti-cancer agents. 相似文献

Soybean crops showing systemic mottling, mosaic and leaf deformation were observed at high disease incidences (25.1–71.0%) in the kharif season of 2011 and 2012 in the experimental farm of the Indian Agricultural Research Institute (IARI), New Delhi. Symptomatic soybean leaves contained flexuous particles (650 × 12 nm), suggesting an infection by a Carlavirus. The causal virus was characterized as a strain of Cowpea mild mottle virus (CPMMV) on the basis of mechanical inoculation, whitefly transmission, seed transmission and sequencing of the viral genome. This is the first report of natural infection by a distinct strain of CPMMV in soybean in India. 相似文献

Current practice of validating predicted protein structural model is knowledge-based where scoring parameters are derived fromalready known structures to obtain decision on validation out of this structure information. For example, the scoring parameter,Ramachandran Score gives percentage conformity with steric-property higher value of which implies higher acceptability. On theother hand, Force-Field Energy Score gives conformity with energy-wise stability higher value of which implies loweracceptability. Naturally, setting these two scoring parameters as target objectives sometimes yields a set of multiple models for thesame protein for which acceptance based on a particular parameter, say, Ramachandran score, may not satisfy well with theacceptance of the same model based on other parameter, say, energy score. The confusion set of such models can further beresolved by introducing some parameters value of which are easily obtainable through experiment on the same protein. In thispiece of work it was found that the confusion regarding final acceptance of a model out of multiple models of the same protein canbe removed using a parameter Surface Rough Index which can be obtained through semi-empirical method from the ordinarymicroscopic image of heat denatured protein. 相似文献

Effect of feeding coconut and blackgram fiber isolated as neutral detergent fiber (NDF) on the levels of lipids and lipid peroxides was studied in rats given a high fat diet. Concentration of cholesterol, free falty acid and phospholipids showed significant decrease in the serum, liver aorta and intestine of coconut and blackgram fiber groups. Concentration of malondialdehyde (MDA) and conjugated dienes was significantly decreased in liver and intestine of both fiber groups, while hydroperoxides showed significant increase in liver and heart of both the fiber groups. SOD and catalase activity was found to be increased in liver, intestine, heart proximal colon and distal colon of both the fiber groups. Serum ceruloplasmin levels showed a slight increase in animals fed coconut and blackgram fiber groups. Glutathione levels in liver, intestine proximal colon, distal colon and heart also showed a significant decrease in the animals of both the fiber groups. 相似文献
