Jatropha curcas L. is one potential source of non-edible biofuel-producing energy crop. Its importance also lies in its medicinal properties. The species is primarily propagated through heterozygous seeds, and thus the seed oil content varies from 4 to 40%. Moreover, due to its perennial nature, seed setting requires 2 to 3 years time. The seed viability and rate of germination are low, and quality seed screening is another laborious task; thus, seed propagation alone cannot provide quality planting material for sustainable use. Somatic embryogenesis, a powerful tool of plant biotechnology for faster and quality plant production has been successfully applied to regenerate plants in Jatropha curcas for the first time. Embryogenic calli were obtained from leaf explants on MS basal medium supplemented with only 9.3 μM Kn. Induction of globular somatic embryos from 58% of the cultures was achieved on MS medium with different concentrations of 2.3–4.6 μM Kn and 0.5–4.9 μM IBA; 2.3 μM Kn and 1.0 μM IBA proved to be the most effective combination for somatic embryo induction in Jatropha curcas. Addition of 13.6 μM adenine sulphate stimulated the process of development of somatic embryos. Mature somatic embryos were converted to plantlets on half strength MS basal medium with 90% survival rate in the field condition. The whole process required 12–16 weeks of culture for completion of all steps of plant regeneration. This protocol of somatic embryogenesis in Jatropha curcas may be an ideal system for future transgenic research.  相似文献

We describe an efficient process for regeneration of Taxus wallichiana (Zucc) plants from callus cultures derived from zygotic embryos. Zygotic embryos cultured on half strength Lloyd and McCown’s basal medium supplemented with SH vitamin (1/2 WPMSH), 0.5 mg I?1 6-benzyladenine (BA) and 1.0–2.0 mg I?1 á-Napthaleneacetic acid (NAA) produced compact yellow (CY) callus after 4 weeks of culture. The 8-week-old CY call! (lines CY-A and CY-B) were initially slow growing but proliferated on transfer to WPM basal medium supplemented with 8.0 mg I?1 2,4-D, 0.1–0.9 mg?1 NAA and 0.3–1.0 mg?1 BA after 4 weeks. Four morphologically distinct calli lines were obtained, of which only two call! lines, CY-B-FW and CY-B-FY were embryogenic. The 12-week-old callus line CY-B-FW developed globular somatic embryos on transfer to secondary medium after 8 weeks and matured in maturation medium after 4 weeks. Only 10% of the mature somatic embryos regenerated into complete plantlets after 4 weeks on conversion medium. Although the frequency of conversion was low, complete regenerated plantlets via somatic embryogenesis were obtained after 7–8 months of initiation of culture. Taxane analysis showed that the paclitaxel accumulation was higher in embryogenic callus than in non-embryogenic callus.  相似文献

Neutrophils (first line of cellular defense) are capable of detecting presence of foreign genome in the mother’s womb. Role of neutrophils during full gestation cycle of ruminants and the difference in their number, type, and activity in successful and unsuccessful pregnancies is not known. To evaluate this, blood samples were collected at artificial insemination (0 day) and on days 10, 14, 16, 18, and 21 in non-pregnant (NP) cows. However in pregnant (P) cows, samples were collected as indicated above and every 30 days for the complete gestation. In aborted cows, samples were collected as above till abortion. Higher total leukocyte counts were observed in NP and aborted cows at abortion. Neutrophil: lymphocyte ratio increased significantly (p < 0.05) in NP and aborted cows. Phagocytic activity (PA) and myeloperoxidase concentrations were significantly higher (p < 0.05) on day 18 post insemination in NP cows. PA and myeloperoxidase also increased significantly (p < 0.05) at abortion in aborted cows. Neutrophils exhibited limited decrease in their number and activity in successful pregnancies during implantation. After that their number and activity were constantly maintained throughout the gestation cycle. Any increase in the number and inflammatory activity of neutrophils may lead to non-pregnancy or loss in pregnancy.  相似文献

Host pathogen interaction results in a variety of responses, which include phagocytosis of the pathogen, release of cytokines, secretion of toxins, as well as production of reactive oxygen species (ROS). Recent studies have shown that many pathogens exert control on the processes that regulate apoptosis in the host. The induction of apoptosis upon infection results from a complex interaction of parasite proteins with cellular host proteins. Abrogation of host cell apoptosis is often beneficial for the pathogen and results in a successful host invasion. However, in some cases, it has been shown that induction of apoptosis in the infected cells significantly imparts protection to the host from the pathogen. There is a strong correlation between apoptosis and the host protein translation machinery: the pathogen makes all possible efforts to modify this process so as to inhibit cell suicide and ensure that it can survive and, in some cases, establish latent infection. This review discusses the significance of various pathways/steps during virus-mediated modulation of host cell apoptosis.  相似文献

The polycomb protein Bmi-1 represses the INK4a locus, which encodes the tumor suppressors p16 and p14(ARF). Here we report that Bmi-1 is downregulated when WI-38 human fibroblasts undergo replicative senescence, but not quiescence, and extends replicative life span when overexpressed. Life span extension by Bmi-1 required the pRb, but not p53, tumor suppressor protein. Deletion analysis showed that the RING finger and helix-turn-helix domains of Bmi-1 were required for life span extension and suppression of p16. Furthermore, a RING finger deletion mutant exhibited dominant negative activity, inducing p16 and premature senescence. Interestingly, presenescent cultures of some, but not all, human fibroblasts contained growth-arrested cells expressing high levels of p16 and apparently arrested by a p53- and telomere-independent mechanism. Bmi-1 selectively extended the life span of these cultures. Low O(2) concentrations had no effect on p16 levels or life span extension by Bmi-1 but reduced expression of the p53 target, p21. We propose that some human fibroblast strains are more sensitive to stress-induced senescence and have both p16-dependent and p53/telomere-dependent pathways of senescence. Our data suggest that Bmi-1 extends the replicative life span of human fibroblasts by suppressing the p16-dependent senescence pathway.  相似文献

Conserving large populations with unique adaptations is essential for minimizing extinction risks. Sundarban mangroves (>10,000 km2) are the only mangrove inhabited by tigers. Baseline information about this tiger population is lacking due to its man-eating reputation and logistic difficulties of sampling. Herein, we adapt photographic capture-mark-recapture (CMR) and distance sampling to estimate tiger and prey densities. We placed baited camera stations in a typical mangrove in 2010 and 2012. We used telemetry based tiger home-range radius (5.73 km, SE 0.72 km) to estimate effective trapping area (ETA). An effort of 407 and 1073 trap nights were exerted to photocapture 10 and 22 unique tigers in 2010 and 2012. We accounted for use of bait by modelling behaviour and heterogeneity effects in program MARK and secr package in program R. Using traditional CMR, tiger number was estimated at 11 (SE 2) and density at 4.07 (SE range 3.09–5.17) in 2010 while in 2012, tiger number was 24 (SE 3) and density 4.63 (SE range 3.92–5.40) tigers/100 km2. With likelihood based spatially explicit CMR, tiger densities were estimated at 4.08 (SE 1.51) in 2010 and 5.81 (SE 1.24) tigers/100 km2 in 2012. Using distance sampling along water channels, we estimated chital density at 5.24/km2, SE 1.23 which could potentially support 4.68 tigers/100 km2 [95 % CI (3.92, 5.57)]. Our estimates suggest that Sundarban tiger population is one of the largest in the world and therefore merits high conservation status.  相似文献

The influence of various heterogeneous parameters, stochastic uncertain factors, and pollutant particles from the industrial effluents in the water system is investigated using advection dispersion equation (ADE) and the Bayesian approximation. Here, the decay coefficient is decomposed into the exact part and the deviation part. The coefficient is used to find out the errors and deviation in decay during the flow of pollutants. Two Bayesian models are developed to analyze the posterior distributions and to find out the Bayes factor for the stochastic covariance estimation. The Bayesian calibration focused the characteristics of both on mechanistic and statistical approximation. The efficiency and accuracy of the developed models are checked from the results obtained on the basis of the confidence interval. Markov chain Monte Carlo simulation is used to acquire the convergence point of parameters for the posterior estimation. The stochastic covariance or white noise represents the effect of random factors on the river system. The analysis revealed that the rate of decay is dependent upon the duration and distance traveled by the pollutants. The collaboration of ADE and Bayesian approximation encourage the water-quality management and environmental modeling.  相似文献

The Sundarban of India and Bangladesh (about 6000 km²) are the only mangrove forests inhabited by a sizeable population of tigers. The adjoining area also supports one of the highest human densities and experiences severe human-tiger conflicts. We used GPS-Satellite and VHF radio-collars on 6 (3 males and 3 female) tigers to study their ranging patterns and habitat preference. The average home range (95% Fixed Kernel) for resident females was 56.4 (SE 5.69) and for males it was 110 (SE 49) km². Tigers crossed an average of 5 water channels > 30 meters per day with a mean width of 54 meters, whereas channels larger than 400 meters were rarely crossed. Tigers spent over 58% of their time within Phoenix habitat but compositional analysis showed a habitat preference of the order Avicennia-Sonneratia > Phoenix > Ceriops > Barren > Water. Average daily distance moved was 4.6 km (range 0.1–23). Activity of tigers peaked between 05:00 hours and 10:00 hours showing some overlap with human activity. Territory boundaries were demarcated by large channels which tigers intensively patrolled. Extra caution should be taken while fishing or honey collection during early morning in Avicennia-Sonneratia and Phoenix habitat types along wide channels to reduce human-tiger conflict. Considering home-range core areas as exclusive, tiger density was estimated at 4.6 (SE range 3.6 to 6.7) tigers/100 km2 giving a total population of 76 (SE range 59–110) tigers in the Indian Sundarban. Reluctance of tigers to cross wide water channels combined with increasing commercial boat traffic and sea level rise due to climate change pose a real threat of fragmenting the Sundarban tiger population.  相似文献
