The surface ionogenicity of five enteropathogenicEscherichia coli serogroups (O111:H2, O111:H12, O125:H9, O119:H6, and O26:H11) was investigated by electrokinetical approaches. All of the studied surfaces are negatively charged with their mean values of zeta potential (ZP) varying from −9.0 (O26:H11) to −11.9 mV (O111:H2). The populational behavior of the all bacteria are similar since very high ZP values varying from −26 to −30 mV were obtained in experiments carried out with the slip plane calculated at 6.83 nm from the cell surface. All the surfaces are extremely acidic, because the isoelectrophoretic points are localized at pH values below 3.0. Treatment of the microorganisms with neuraminidase did not alter their surface anionogenicity, while treatment with trypsin or phospholipase C reduced their negative charge.  相似文献

BackgroundThe variability in the association of host innate immune response to Hepatitis C virus (HCV) infection requires ruling out the possible role of host KIR and HLA genotypes in HCV-related disorders: therefore, we therefore explored the relationships between KIR/HLA genotypes and chronic HCV infection (CHC) as they relate to the risk of HCV-related hepatocarcinoma (HCC) or lymphoproliferative disease progression.ConclusionsOur data highlight a role of the innate-system in developing HCV-related disorders and specifically KIR2DS3 and KIR2D genes demonstrated an ability to direct HCV disease progression, and mainly towards lymphoproliferative disorders. Moreover the determination of KIR3D/HLA combination of genes direct the HCV progression towards a lymphoma rather than an hepatic disease. In this contest IFN-α therapy, a standard therapy for HCV-infection and lymphoproliferative diseases, known to be able to transiently enhance the cytotoxicity of NK-cells support the role of NK cells to counterstain HCV-related and lymphoproliferative diseases.  相似文献

Quantitative changes of mouse oocyte protein synthesis during oogenesis have been studied by determining the rate of leucine incorporation into total cell protein. The leucine intracellular poor was artificially expanded by exposing the isolated oocytes to different concentrations of radiolabelled leucine, and leucine incorporation was followed as a function of the time. Protein synthesis by mouse oocytes increases linearly with the increase of the cell volume, and such increase is constant throughout the entire period of oocyte growth.  相似文献

Bacillus thuringiensis is a Gram positive bacterium that is used for the production of biopesticides. The toxic action of different strains and serovars ofB. thuringiensis can be extremely selective towards specific pests, or, in contrast, it can affect a wide variety of non-target organisms such as insects, vertebrates or humans. A reliable characterization of the cultivated strains is of primary importance for the biopesticide industry, in order to assess the contamination of the final product with strains with different pesticide actions or that might be dangerous for human health. The aim of this study was to develop useful methods for the typing of differentB. thuringiensis strains using two PCR-based methods, RAPD and Rep-PCR with BOXA1R and ERIC2 primers. The molecular fingerprints obtained using ERIC2-PCR showed a reliable ability to discriminateBacillus thuringiensis strains.  相似文献

Follicle cell regulation of mammalian oocyte growth   总被引:2,自引:0,他引:2  
To investigate mechanisms of follicle cell control on mammalian oocyte growth, preantral mouse oocytes free from surrounding follicle cells were individually cocultured with monolayers of different somatic cells competent to form gap junctions, and the rate of in vitro oocyte growth was directly correlated with the level of metabolic coupling on the same cells. The results indicate that 1) at a similar extent of metabolic coupling, mouse oocytes grew on follicle cells but not on 3T3 and Sertoli cell monolayers, and 2) the growth rate of oocytes cultured on follicle cells was dependent on the extent of metabolic coupling. It was concluded that gap-junction-mediated nutrition of ovarian mouse oocytes exerted by somatic cells is necessary but not sufficient to maintain oocyte growth. A specific regulatory role of follicle cells on mammalian oocyte growth is proposed.  相似文献

In order to verify whether the HER-2/neu gene is involved in the initial phases of neoplastic disease or in its progression, we evaluated the amplification and overexpression of this gene in the primary tumor and in synchronous metastatic axillary lymph nodes of 26 women with operable breast cancer. HER-2/neu was amplified in 35% and overexpressed in 33% of the primary sites; similar percentages were found in lymph nodes. The clear correlation between the two disease sites regarding gene, mRNA and protein levels, supports the hypothesis that this gene is involved in the initial and invasive phases of neoplasia. Its actual role with respect to other biological tumor characteristics during the metastatic process should be investigated further.  相似文献

Functional magnetic resonance spectroscopy (fMRS) allows the non-invasive measurement of metabolite concentrations in the human brain, including changes induced by variations in neurotransmission activity. However, the limited spatial and temporal resolution of fMRS does not allow specific measurements of metabolites in different cell types. Thus, the analysis of fMRS data in the context of compartmentalized metabolism requires the formulation and application of mathematical models. In the present study we utilized the mathematical model introduced by Simpson et al . (2007) to gain insights into compartmentalized metabolism in vivo from the fMRS data obtained in humans at ultra high magnetic field by Mangia et al . (2007a) . This model simulates brain glucose and lactate levels in a theoretical cortical slice. Using experimentally determined concentrations and catalytic activities for the respective transporter proteins, we calculate inflow and export of glucose and lactate in endothelium, astrocytes, and neurons. We then vary neuronal and astrocytic glucose and lactate utilization capacities until close correspondence is observed between in vivo and simulated glucose and lactate levels. The results of the simulations indicate that, when literature values of glucose transport capacity are utilized, the fMRS data are consistent with export of lactate by neurons and import of lactate by astrocytes, a mechanism that can be referred to as a neuron-to-astrocyte lactate shuttle. A shuttle of lactate from astrocytes to neurons could be simulated, but this required the astrocytic glucose transport capacity to be increased by 12-fold, and required that neurons not respond to activation with increased glycolysis, two conditions that are not supported by current literature.  相似文献

Modifications of leucine transport system of mouse oocytes have been studied throughout Graafian follicle development and oocyte maturation. In contrast to sheep oocytes (Moor and Smith, 1979), in the mouse kinetic constants and efflux rate of leucine transport system did not vary in diestrus, proestrus, and metaphase II (met II) oocytes. However, kinetics of leucine equilibration in proestrus and met II oocytes was significantly slower than that found in diestrus cells, and this may reflect a decreased availability of internal amino acids for exchange.  相似文献
