The effect of Artemisia monosperma wild plant hexane extract on some biological and physiological aspects of Spodoptera littoralis, as well as some biochemical and histopathological changes after treatment with this extract have been studied. The results indicated that treatment with larvae for two days revealed satisfactory effects in larval mortality (50%) and in larval weight and prolonged the effect in the pupal stage. Also the treatment reduced the food consumption and the amount of faeces of the tested larvae which led to a significant reduction in the final body weight with 76.4%. Treatment also extended the time of food passage through the midgut to 3.2 h compared to 1.38 h in control, and the larvae had a smaller midgut. The enzymatic activity of the digestive enzymes amylase and invertase revealed a significant inhibition in the treated larvae. Also transaminase enzyme activity (AST and ALT) reduced after treatment. The results of phosphatase enzyme activity (ACP and ALP) showed a potent inhibitory effect on both enzymes which more was pronounced in ALP than ACP activity, the percent inhibition being 73.8% and 30.6%, respectively. The histopathological effects of A. monosperma treatment on the midgut of Spodoptera littoralis revealed that the epithelium membrane was exfoliated and completely destroyed. Meanwhile, epithelia cells were strongly vacuolated and considerably elongated. Boundary lines between the cells disappeared.  相似文献

Background

Hepatocellular carcinoma (HCC) associated to infection with hepatitis C virus (HCV) has become the fastest-rising cause of cancer-related deaths. Genetic variations may play an important role in the development of HCC in HCV patients. Ghrelin exerts anti-inflammatory, antifibrotic and hepatoprotective effects on chronically injured hepatic tissues. Ghrelin gene shows several single nucleotide polymorphisms (SNPs) including − 604G/A, Arg51Gln, and Leu72Met. Hemochromatosis gene (HFE) mutations namely C282Y and H63D may cause hepatic iron overload, thus increasing the risk of HCC in HCV patients.

Aim

To investigate the association of progression of HCC with ghrelin and HFE gene polymorphisms in HCV Egyptian patients.

Methods

Seventy-nine chronic HCV patients (thirty-nine developed HCC and forty did not), and forty healthy control subjects were included in the study. The polymorphisms were evaluated by PCR/RFLP analysis, and related protein levels were measured by either ELISA or colorimetric assays.

Results

The three tested SNPs on ghrelin gene were detected in the studied groups, only one SNP (Arg51Gln) showed significantly higher GA, AA genotypes and A allele frequencies in hepatitis C patients who developed HCC than in hepatitis C patients without HCC and controls. Of the two mutations studied on HFE gene only H63D heterozygous allele was detected, and its frequency did not statistically differ among studied groups.

Conclusion

Our results suggest that A allele at position 346 of the ghrelin gene is associated with susceptibility to HCC in hepatitis C patients.  相似文献

Spodoptera littoralis caterpillars were transferred from an artificial diet to potato leaves at the start of the third or fifth instar and exposed to the infective juveniles of the nematode Steinernema feltiae since the beginning of the sixth instar until the start of pupation. Leaves were taken from the control potatoes or from genetically modified potatoes expressing either Cry3Aa toxin of Bacillus thuringiensis (Bt) or Galanthus nivalis agglutinin (GNA) which are mainly non-specific to S. littoralis larvae. The nematodes killed all the caterpillars within seven days compared with the starved larvae in the same period of exposure. The average time to death and the number of nematodes successfully invaded the larvae were affected by the period of feeding on potato leaves. In the non-starved caterpillars, which received potato leaves throughout the whole period of exposure to the nematodes, the type of potato leaves had no effect on the number of nematodes inside cadavers (p = 0.352 and F = 1.070) and also on the effect on the length of survival after exposure to the nematodes (p = 0.7892 and F = 0.596). No hazardous effect on the development and survival of entomopathogenic nematode S. feltiae which successfully invaded larvae fed on modified potato (Bt or GNA) was reported.  相似文献

Leishmaniasis is a group of infectious and noncontagious severe parasitic diseases, caused by protozoans of the Leishmania genus. Natural products characterize a rich source of prospective chemical entities for the development of new effective drugs for neglected diseases. Scientific evaluation of medicinal plants has made it possible to use some metabolites from flavonoids and polyphenols compounds for the treatment of parasitic diseases. Therefore, we aimed in the present study to evaluate the protective effect of silver nanoparticles (Ag-NPs) biosynthesized using Fig and Olive extracts (NFO) against Cutaneous leishmaniasis in female Balb/c mice. A total of 70 mice were used and divided into seven groups. Treatment was initiated when local lesions were apparent, we found that Fig and Olive extracts were found to be a good source for the synthesis of (Ag-NPs), their formation was confirmed by color change and stability in solution. Nanoparticles biosynthesized using Fig and Olive extracts induced a reduction in the average size of cutaneous leishmaniasis lesions compared with the untreated mice. Moreover, nanoparticles treatment decreased oxidative stress (LPO, NO), down-regulation gene expression levels (TNF-α, IL-1β, and BAX), and this antileishmanial activity of nanoparticles was associated with enhanced antioxidant enzyme activities. In addition, histopathological evaluation proved the antileishmanial activity of nanoparticles compared with the positive control.Therefore, we aimed in the present study to evaluate the protective effect of silver nanoparticles biosynthesized using Fig and Olive extracts against cutaneous lesions induced by Leishmania major infection through their anti-inflammatory, antioxidant activities, and faster clinical efficacy than standard pentavalent antimonial treatment.  相似文献

The role of muscle activation in both pathological and spastic populations is of interest for understanding central nervous system function. Muscle activation patterns may provide insight into pathological changes compared to healthy controls. To gain a better understanding of surgical interventions, gait muscle activation patterns are studied before and after surgery. Previous studies using surface electromyography have indicated that muscle activation onset, time to peak, and peak amplitude may be helpful in assessing the neuromuscular control strategy that underlies pathological populations. Geometric artifact may influence electromyographic variables as recorded by different electrode types and electrode placement. The purpose of this investigation was to compare surface and fine-wire activation patterns during gait to elucidate the influence electrode type has on electromyographic variables. Lower leg surface and fine-wire electromyographic activity was recorded simultaneously during gait to assess if electrode type (fine-wire vs. surface) affects muscle onset, time to peak, peak amplitude, and activation patterns. No significant differences were recorded between surface and fine-wire electrodes for muscle onset or time to peak activation. Activation patterns revealed similarity between electrodes. Some significant differences were detected in peak amplitude. Non-invasive surface electrodes provide an adequate representation of timing variables for primary ankle muscles during gait.  相似文献

ABSTRACT: BACKGROUND: The anaerobic spirochaete Brachyspira pilosicoli causes enteric disease in avian, porcine and human hosts, amongst others. To date, the only available genome sequence of B. pilosicoli is that of strain 95/1000, a porcine isolate. In the first intra-species genome comparison within the Brachyspira genus, we report the whole genome sequence of B. pilosicoli B2904, an avian isolate, the incomplete genome sequence of B. pilosicoli WesB, a human isolate, and the comparisons with B. pilosicoli 95/1000. We also draw on incomplete genome sequences from three other Brachyspira species. Finally we report the first application of the high-throughput Biolog phenotype screening tool on the B. pilosicoli strains for detailed comparisons between genotype and phenotype. RESULTS: Feature and sequence genome comparisons revealed a high degree of similarity between the three B. pilosicoli strains, although the genomes of B2904 and WesB were larger than that of 95/1000 (~2,765, 2.890 and 2.596 Mb, respectively). Genome rearrangements were observed which correlated largely with the positions of mobile genetic elements. Through comparison of the B2904 and WesB genomes with the 95/1000 genome, features that we propose are non-essential due to their absence from 95/1000 include a peptidase, glycine reductase complex components and transposases. Novel bacteriophages were detected in the newly-sequenced genomes, which appeared to have involvement in intra- and inter-species horizontal gene transfer. Phenotypic differences predicted from genome analysis, such as the lack of genes for glucuronate catabolism in 95/1000, were confirmed by phenotyping. CONCLUSIONS: The availability of multiple B. pilosicoli genome sequences has allowed us to demonstrate the substantial genomic variation that exists between these strains, and provides an insight into genetic events that are shaping the species. In addition, phenotype screening allowed determination of how genotypic differences translated to phenotype. Further application of such comparisons will improve understanding of the metabolic capabilities of Brachyspira species.  相似文献

MOTIVATION: Recognition of poly(A) signals in mRNA is relatively straightforward due to the presence of easily recognizable polyadenylic acid tail. However, the task of identifying poly(A) motifs in the primary genomic DNA sequence that correspond to poly(A) signals in mRNA is a far more challenging problem. Recognition of poly(A) signals is important for better gene annotation and understanding of the gene regulation mechanisms. In this work, we present one such poly(A) motif prediction method based on properties of human genomic DNA sequence surrounding a poly(A) motif. These properties include thermodynamic, physico-chemical and statistical characteristics. For predictions, we developed Artificial Neural Network and Random Forest models. These models are trained to recognize 12 most common poly(A) motifs in human DNA. Our predictors are available as a free web-based tool accessible at http://cbrc.kaust.edu.sa/dps. Compared with other reported predictors, our models achieve higher sensitivity and specificity and furthermore provide a consistent level of accuracy for 12 poly(A) motif variants. The entomopathogenic nematode Steinernema feltiae strain Ustinov Russia was used on potato foliage to control larvae of the Colorado potato beetle, Leptinotarsa decemlineata (Say.) (Coleoptera: Chrysomelidae). Nematodes were applied in formulations of agar at 4%, 2%, 1% and 0.5% concentrations and compared to a control application of nematodes in water for nematode survival. Agar formulation significantly improved efficacy by increasing nematode survival through reduction in desiccation when compared to water-based formulation. More than 70% of infective juvenile nematodes (IJs) died after being incubated in the highest concentration of agar for 12 h, while only 8% mortality was recorded at the 1% concentration. Suspension of nematodes in 1% agar gel was shown to be efficacious in both laboratory and greenhouse tests for extension of the nematodes’ survival. Agar formulation droplets dried slower than control droplets by 127.8 min. Leptinotarsa decemlineata mortality significantly increased when insects were exposed to infective juvenile nematodes for four hours after application. In conclusion, the agar formulation enhanced nematode survival by providing a suitable environment thereby delaying dryingand increasing the possibility for nematodes to invade their host on the foliage.  相似文献

A sensitive, specific and selective method has been developed for the simultaneous determination of bisoprolol and hydrochlorothiazide in human plasma. The method employed a state of the art LC–MS/MS operated in the positive and negative ionization switching modes. A simple sample preparation step involving protein precipitation with acetonitrile has been optimized; the analytes and the internal standard moxifloxacin were separated on a Purosphere® STAR C8 column (125 mm × 4 mm, 5 μm). The mobile phase was an ammonium acetate solution (1 mM) with formic acid (0.2%): methanol and acetonitrile (65:17.5:17.5, v/v/v (%)), the flow rate was set at 0.65 mL/min. Bisoprolol and hydrochlorothiazide were ionized using ESI source prior to detection by Multiple Reaction Monitoring (MRM) mode while monitoring at the following transitions: positive m/z 326  116 for bisoprolol, negative m/z 296  269 and m/z 296  205 for hydrochlorothiazide. Linearity was demonstrated over the concentration range 0.10–30.0 (ng/mL) for bisoprolol and 1.00–80.00 ng/mL for hydrochlorothiazide. The limits of detection were 0.100 (ng/mL) for bisoprolol and 1.00 (ng/mL) for hydrochlorothiazide. The validated method was successfully applied to a pharmacokinetic study of 5 mg bisoprolol fumarate with 12.5 mg hydrochlorothiazide tablet in healthy volunteers.  相似文献

This study was undertaken to evaluate selenium (Se) and glutathione peroxidase (GPX) status in patients with newly diagnosed acute myeloid leukemia (AML) before and after induction therapy. Twenty-five patients with newly diagnosed AML and 15 healthy age- and sex-matched control subjects were included in this study. Serum Se level by the graphite furnace atomic absorption spectrometric technique and GPX activity by an adaptation of Beutler method was performed for the patients before and after receiving the induction therapy. Serum Se level was significantly lower in patients with AML versus control subjects (63.1?±?8.8 versus 77?±?8.8 µg/L before therapy with a P value <0.01 and 69?±?6.8 versus 77?±?8.8 µg/L after therapy with a P value <0.01).GPX activity was significantly lower in patients with AML versus control subjects (1.6?±?0.4 versus 3.4?±?0.7 µ/g protein pretreatment with a P value <0.01and 1.9?±?0.6 versus 3.4?±?0.7 µ/g protein post induction treatment with P value <0.01).Se level and GPX activity significantly increased in AML patients after treatment. Patients who accomplished complete remission after induction harbored significantly higher Se levels than resistant patients before and after treatment. There was no significant correlation between serum Se level and GPX activity. Decreased Se level and reduced GPX activity in AML patients support the association of carcinogenesis and subnormal Se states.  相似文献
