We purified recombinant bovine -lactoglobulin (r-LG) from the culture supernatant of transformed yeast and investigated whether r-LG maintained the functional ability and antigenicity of native -LG. Immunostaining following gel electrophoresis and reversed-phase high-performance liquid chromatography confirmed that r-LG was purified homogeneously. r-LG showed almost the same retinol-binding ability as native -LG purified from bovine milk. However, affinities of two anti--LG monoclonal antibodies (mAbs) to r-LG were different from those to native -LG, although three other mAbs bound these two proteins equally. Since our panel of five mAbs has been previously shown to be able to detect structural changes occurring in -LG, this variance in antigenicity can be attributed to conformational differences between r-LG and native -LG. Then, we studied which step in the production and purification procedure was responsible for altering the antigenicity of r-LG. Bovine milk native -LG was added to several steps in this procedure and purified in the same manner as r-LG. The results suggested that incubation in the yeast culture had adverse effects on maintaining the antigenicity of this recombinant protein. We conclude from these results that even if no difference between the native and recombinant proteins can be detected by functional analysis, some subtle conformational change which can be distinguished by mAbs may be incorporated into the recombinant protein during its production and ultimately cause a different immune reaction in vivo.Abbreviations -LG, -lactoglobulin; r-LG, recombinant -LG; PBS, phosphate-buffered saline; PBS-Tween, PBS containing 0.05% Tween 20; ELISA, enzyme-linked immunosorbent assay.  相似文献

T cell responses can be antagonized by some single amino acid-substituted analogs of a peptide ligand for T-cell receptors (TCR), and these are called TCR antagonists. In this study, we addressed the question of whether TCR antagonism can be elicited by a whole protein antigen carrying a mutated T-cell determinant region corresponding to a TCR antagonist peptide. To clarify this, we examined the ability of a single amino acid-substituted mutant form of bovine β-lactoglobulin (β-Lg) to inhibit three CD4+ T-cell clones recognizing a peptide corresponding to an immunodominant determinant region 119-133 of β-Lg (p119-133). First, we identified pD129A, an analog of p119-133 with a substitution of Ala for 129Asp, as an antagonist which can inhibit the response of two of the three T-cell clones. Then, using a yeast expression system, we prepared a mutant β-Lg (mutD129A) with the same substitution of Ala for 129Asp as that in pD129A. This mutant protein could inhibit the proliferation of the two T-cell clones in a manner similar to the effect of pD129A. From these results we can demonstrate that TCR antagonism can be elicited by peptides naturally processed from a single-substituted mutant protein as well as by the corresponding peptides added exogenously. We isolated a 38 kDa ssDNA-binding protein from the unicellular cyanobacterium Synechococcus sp. strain PCC 6301 and determined its N-terminal amino acid sequence. A genomic clone encoding the 38 kDa protein was isolated by using a degenerate oligonucleotide probe based on the amino acid sequence. The nucleotide sequence and predicted amino acid sequence revealed that the 38 kDa protein is 306 amino acids long and homologous to the nuclear-encoded 370 amino acid chloroplast ribosomal protein CS1 of spinach (48% identity), therefore identifying it as ribosomal protein (r-protein) S1. Cyanobacterial and chloroplast S1 proteins differ in size from Escherichia coli r-protein S1 (557 amino acids). This provides an additional evidence that cyanobacteria are closely related to chloroplasts. The Synechococcus gene rps1 encoding S1 is located 1.1 kb downstream from psbB, which encodes the photosystem 11 P680 chlorophyll a apoprotein. An open reading frame encoding a potential protein of 168 amino acids is present between psbB and rps1 and its deduced amino acid sequence is similar to that of E. coli hypothetical 17.2 kDa protein. Northern blot analysis showed that rps1 is transcribed as a monocistronic mRNA.  相似文献

Roots of the agravitropic pea (Pisum sativum L.) mutant, ageotropum, responded to a gradient in water potential as small as 0.5 MPa by growing toward the higher water potential. This positive response occurred when a sorbitol-containing agar block was unilaterally applied to the root cap but not when applied to the elongation region. Unilateral application of higher concentrations of sorbitol to the elongation region caused root curvature toward the sorbitol source, presumably because of growth reduction on the water-stressed side. The control blocks of plain agar applied to either the root cap or the elongation region did not cause significant curvature of the roots. These results demonstrate that hydrotropism in roots occurs following perception of a gradient in water potential by the root cap.  相似文献

The quantitative relationship between reaction centers I andII was studied with blue-green algae Anabaena cylindrica, Anabaenavariabilis and Anacystis nidulans grown under different lightconditions. The number of reaction centers I was estimated fromthe P700 content and that of reaction centers II, from the O2yield of repetitive short flashes. Supplementary determinationswere done with three other blue-green algae and one red alga.The maximum number of reaction centers II counted from the O2yield of repetitive short flashes was markedly smaller thanthe total number of P700 in all algae tested when the algaewere grown under weak light; in the extreme case (Anabaena cylindrica),the ratio was only 0.258?0.015. This ratio became larger andclose to unity when the algae were grown under stronger light.Variation in the number of reaction centers in a single cellsuggested that reaction center I was a variable component. Deuteration of asparagine, glutamine, arginine, tryptophan, and tyrosine has been found to cause a small but appreciable spectral change in the 200- to 230-nm region. By the use of a stopped-flow ultraviolet absorption method, the rates of deuteration of these amino acids have been determined. On the basis of these results, and on the basis of our previous study on the hydrogen-deuterium exchange of the peptide groups, a few comments are given as for how to apply the method to protein study.  相似文献

The effects of chronic treatment of the rat with methacholine and atropine on the cardiac muscarinic cholinergic receptors were investigated. [3H]Quinuclidinyl benzilate ([3H]QNB) was used to directly estimate the number and affinity of the receptors in the heart ventricular membrane. Methacholine treatment decreased, in a dose-related and time-dependent manner, the specific binding of [3H]QNB by 34% as compared to the control. Atropine treatment, on the other hand, resulted in a dose-related increase (28 to 66%) in the number of the receptors. The equilibrium dissociation constant (KD) of the receptors for the ligand was the same (about 200 pM) for the control and the methacholine treated groups of rats, whereas a dose-related increase (39 to 105%) in the KD was noted for the atropine treated rats. Similarly, the concentration of acetylcholine causing a 50 percent inhibition (IC50) of the [3H]QNB binding was unaltered for the methacholine treated rats (4 μM), but it was increased 43% for the atropine treated rats.  相似文献

The oral administration of hyaluronans (HAs) (molecular weight, 300k and less than 10k) to photoaged hairless mice increased the moisture content of the stratum corneum and decreased the epidermal thickness, respectively. Furthermore, orally administered HAs suppressed the low-molecular weight of HA content of the skin. This study indicates oral administered HAs may ameliorate the skin condition resulting from photoaging.  相似文献
