Using monolocus PCR analysis with the pairs of primers designed for the Du215 locus of Darevskia unisexualis, allelic polymorphism at the orthologous locus in the populations of the related parthenospecies D. armeniaca was investigated. It was demonstrated that Du215 (arm) locus was polymorphic and in the populations of parthenospecies D. armeniaca (n = 127) represented by at least three allelic variants, differing from each other by the size and composition of microsatellite cluster, and by single nucleotide substitutions in flanking DNA. Unlike the Du215 locus, Du215 (arm) was shown contain not only GATA, but also (GACA) repeats, which were absent in D. unisexualis. Thus, in this study, the data on the molecular nature of allelic polymorphism at one of the microsatellite loci of the parthenospecies D. armeniaca were reported.  相似文献

Possible antigenic features common to nerve cell membrane structures and cytoplasmic tetrodotoxin-sensitive proteins were investigated by means of an immobilized immunoenzyme test. It was found that antiserum obtained by immunizing rabbits with a purified preparation of cytoplasmic tetrodotoxin-sensitive proteins can react with antigenic determinants present on the membrane fraction of bovine brain cells, on rat synaptosomes and on cells of a clonal line of mouse neuroblastoma. It was shown by means of an inhibition assay test that antibodies of the same specificity contribute to the observed response. Findings would indicate the presence of antigenic determinants common to nerve cell membrane structures and cytoplasmic tetrodotoxin-sensitive proteins. This is consistent with the hypothesis that cytoplasmic tetrodotoxin-sensitive proteins have certain features in common with membrane sodium channels.A. A. It was shown that the protein from the cytoplasm of bovine brain cells, which possesses functional similarity to the protein of membrane potential-dependent sodium channels, is the substrate of cAMP-dependent phosphorylation. Polyamines appreciably inhibit the phosphorylation of cytosolic protein. Spermine proves to be the most active here; in a concentration of 5 mM the inhibition reaches 40%. Unisexual all-female lizards of the genus Darevskia that are well adapted to various habitats are known to reproduce normally by true parthenogenesis. Although they consist of unisexual lineages and lack effective genetic recombination, they are characterized by some level of genetic polymorphism. To reveal the mutational contribution to overall genetic variability, the most straightforward and conclusive way is the direct detection of mutation events in pedigree genotyping. Earlier we selected from genomic library of D. unisexualis two polymorphic microsatellite containing loci Du281 and Du215. In this study, these two loci were analyzed to detect possible de novo mutations in 168 parthenogenetic offspring of 49 D. unisexualis mothers and in 147 offspring of 50 D. armeniaca mothers. No mutant alleles were detected in D. armeniaca offspring at both loci, and in D. unisexualis offspring at the Du215 locus. There were a total of seven mutational events in the germ lines of four of the 49 D. unisexualis mothers at the Du281 locus, yielding the mutation rate of 0.1428 events per germ line tissue. Sequencing of the mutant alleles has shown that most mutations occur via deletion or insertion of single microsatellite repeat being identical in all offspring of the family. This indicates that such mutations emerge at the early stages of embryogenesis. In this study we characterized single highly unstable (GATA)(n) containing locus in parthenogenetic lizard species D. unisexualis. Besides, we characterized various types of mutant alleles of this locus found in the D. unisexualis offspring of the first generation. Our data has shown that microsatellite mutations at highly unstable loci can make a significant contribution to population variability of parthenogenetic lizards.  相似文献

A simple and effective method for typing of CAG repeats in the IT-15 gene has been suggested. This method was applied for examination of the CAG allele distribution in Huntington's disease (HD) patients in five different populations from the Commonwealth of Independent States. A total of 21 normal alleles with the sizes ranging from 9 to 32 triplet repeat units were revealed. Moreover, alleles with the size ranging from 16 to 20 repeats predominated constituting from 54.4 to 74.6% of all alleles in different populations. The number of repeats in one allele in HD patients exceeded 38 units (43 triplets on average). In two families an increase in the CAG repeat units number in the mutant allele upon its paternal transmission was recorded.  相似文献

In the genome of unisexual (parthenogenetic) lizard Darevskia armeniaca, highly variable locus Du 161 (arm) was discovered. Analysis of allelic polymorphism was carried out using locus-specific PCR of the lizard DNA specimens from 13 isolated Armenian populations (N = 138). In the sample examined, a total of 12 Du 161(arm) alleles were identified, and their differences at the level of primary DNA structure were determined. Sequence analysis of the Du 161 (arm) alleles showed that their microsatellite clusters contained repeats of one type (GATA repeats). Allelic Du 161 (arm) variants differed in the number of GATA monomers in microsatellite, point mutations of transition and transversion types, located at fixed distances from microsatellite cluster, and by single nucleotide insertions, as well as by longer insertions located within and outside of the microsatellite cluster. Moreover, point mutations formed different combinations (haplotypes), typical of certain alleles. These combinations can be used for the analysis of the origin and inheritance of these alleles in D. armeniaca, as well as for investigation of their interspecific variation in the representatives of the genus Darevskia.  相似文献

Microbial volatile organic compounds (MVOCs) were collected in water-damaged buildings to evaluate their use as possible indicators of indoor fungal growth. Fungal species isolated from contaminated buildings were screened for MVOC production on malt extract agar by means of headspace solid-phase microextraction followed by gas chromatography-mass spectrometry (GC-MS) analysis. Some sesquiterpenes, specifically derived from fungal growth, were detected in the sampled environments and the corresponding fungal producers were identified. Statistical analysis of the detected MVOC profiles allowed the identification of species-specific MVOCs or MVOC patterns for Aspergillus versicolor group, Aspergillus ustus, and Eurotium amstelodami. In addition, Chaetomium spp. and Epicoccum spp. were clearly differentiated by their volatile production from a group of 76 fungal strains belonging to different genera. These results are useful in the chemotaxonomic discrimination of fungal species, in aid to the classical morphological and molecular identification techniques.  相似文献

DNA methylation is an epigenetic mechanism that regulates gene expression and can be modified by one-carbon nutrients. The objective of this study was to investigate the impact of folic acid (FA) fortification of the US food supply on leukocyte global DNA methylation and the relationship between DNA methylation, red blood cell (RBC) folate, and other one-carbon biomarkers among postmenopausal women enrolled in the Women's Health Initiative Observational Study. We selected 408 women from the highest and lowest tertiles of RBC folate distribution matching on age and timing of the baseline blood draw, which spanned the pre- (1994–1995), peri- (1996–1997), or post-fortification (1998) periods. Global DNA methylation was assessed by liquid chromatography-tandem mass spectrometry and expressed as a percentage of total cytosine. We observed an interaction (P = 0.02) between fortification period and RBC folate in relation to DNA methylation. Women with higher (vs. lower) RBC folate had higher mean DNA methylation (5.12 vs. 4.99%; P = 0.05) in the pre-fortification period, but lower (4.95 vs. 5.16%; P = 0.03) DNA methylation in the post-fortification period. We also observed significant correlations between one-carbon biomarkers and DNA methylation in the pre-fortification period, but not in the peri- or post-fortification period. The correlation between plasma homocysteine and DNA methylation was reversed from an inverse relationship during the pre-fortification period to a positive relationship during the post-fortification period. Our data suggest that (1) during FA fortification, higher RBC folate status is associated with a reduction in leukocyte global DNA methylation among postmenopausal women and; (2) the relationship between one-carbon biomarkers and global DNA methylation is dependent on folate availability.  相似文献

Using light and electron microscopy, the morphology of a new species of pelobionts Pelomyxa flava was studied. The coverings of P. flava are represented by plasma membrane bearing the thick layer of weakly structured glycocalyx on its outer surface. Numerous flagella are often located on the tops of short conical pseudopodia. Kinetosomes of flagella reach a length of 0.9 microm and are hollow with a pronounced central filament. Rootlet system is represented by three groups of microtubules: the radial, basal and microtubules of lateral root. The transition zone is short and does not exceed the level of cell surface; the axoneme is characterized by an unstable set of microtubules. Trophic stages of P. flava life cycle are presented by binuclear cells; plasmotomy is performed at the tetranuclear stage. Nuclei have a granular structure. Fibrillar nuclear bodies were revealed in karyoplasm. The nuclei shell has a complex organization. On its surface, the outer membrane has a layer of electron-dense material which contacts with short microtubules, located in a row at the surface of the nuclear envelope. The bubbles and cisterns of endoplasmic reticulum, which are the derivatives of the nuclear envelope, are located outward from the microtubules. The presence of structural and digestive vacuoles and grains of glycogen was noticed in P. flava endoplasm. Three types of prokaryotic cytobionts were revealed. Large multi-membranous organelles reaching 5 pm in diameter were described for the first time. We discuss morphology and biology features of P. flava in comparison with the previously studied Pelomyxa species.  相似文献
