Chi's are genetic elements that stimulate generalized recombination in their locale in phage λ. All Chi's, wherever located on λ's chromosome, act asymmetrically in crosses blocked in DNA replication: (1) They stimulate exchange primarily to their left on the conventional λ map, and (2) the stimulated exchange is frequently nonreciprocal, the recombinant carrying the Chi element being produced less often than the complementary product. 相似文献

Long-chain alcohols are synthesized in the mouse preputial gland tumor (ESR-586) by NADPH:acyl-CoA oxidoreductase. In this study, a series of labeled acids was tested as substrates for the oxidoreductase in a cell-free system from the tumor, and the distribution of label into alcohols, waxes, and other products was determined. The system contained the labeled acid, an acyl-CoA-generating system, an NADPH-generating system, and tumor homogenate. The highest rates of alcohol synthesis were obtained with palmitic (16:0), heptadecanoic (17:0), stearic (18:0), myristic (14:0), elaidic (18:1 trans), and linoleic (18:2) acids, which yielded, respectively, 151, 124, 102, 76, 65, and 35 pmol alcohol/min per mg protein. Decanoic (10:0), lauric (12:0), oleic (18:1 cis), linolenic (18:3), arachidonic (20:4), and behenic (22:0) acids all gave lower activities. Acyl-CoA formation did not appear to be rate limiting with any of the substrates tested except behenic acid. In addition to the fatty alcohol product, a small amount of fatty aldehyde was formed in the system. Incorporation of the labeled fatty acids into wax esters was examined and the distribution of label between the alcohol and acid components of the waxes was determined. Incubation of [1-(14)C]palmitic acid yielded 3.4% free alcohol, 8.3% alcohol esterified in waxes, and 7.7% palmitoyl groups esterified into waxes, whereas, at the other extreme, [1-(14)C]linolenic acid yielded 0.8%, 0.6%, and 38%, respectively, into the homologous components.-Wykle, R. A cytochemical study has been made on the localization of ATPase activity in corn (Zea mays L.) roots. Light microscopy shows washing for 4 hours to increase the general ATPase activity in the peripheral layers of the root cortex; oligomycin and N,N-dicyclohexylcarbodiimide inhibit this activity, oligomycin being more effective. Ultrastructural studies of ATPase location show oligomycin treatment to inhibit both mitochondrial and plasmalemma ATPase, but only in the epidermis and outer cortex. Studies with lipid-soluble dyes indicate that oligomycin might not penetrate very deeply into root tissue in the time span of these experiments. It is suggested that the strong inhibition of ion absorption by oligomycin without a corresponding decline in ATP content is probably due to inhibition of ion absorption in the peripheral cell layers, thus limiting the supply of ion for symplastic transport to the uninhibited tissues. 相似文献

As part of a program to develop drugs which will delay or prevent cancer in humans, the Chemoprevention Branch, National Cancer Institute, National Institutes of Health, is sponsoring 12 Phase I and 22 Phase II and III clinical trials. Three agent classes are significantly advanced in the trials. These are the retinoids, including 13-cis-retinoic acid, retinol, and 4-hydroxyphenylretinamide (nine studies), beta-carotene (seven studies), and calcium compounds (three studies). In addition, six promising new compounds are in Phase I or Phase II trials. These are: piroxicam, ibuprofen, oltipraz (a dithiolthione), difluoromethylornithine, glycyrrhetinic acid, and N-acetylcysteine. Acyltransferases are present in microsomes from alveolar type II cell adenomas (produced by urethan injections) that transfer palmitic acid in the presence of CoA, ATP, and Mg++ to -glycerol-3-P to form phosphatidic acid, to dihydroxyacetone-P to form acyldihydroxyacetone-P, and to 1-acyl--glycero-3-phosphocholine to form 3--phosphatidylcholine. The data clearly demonstrate that the microsomal preparations can catalyze significant incorporation of palmitic acid into the 2-position of the disaturated species of 3-sn-phosphatidylcholine independently of phosphatidic acid formation as evidenced by the fact that -glycerol-3-P and calcium ions (which inhibit choline phosphotransferase) did not influence the incorporation of palmitic acid into the main surfactant lipid. Thus, a deacylation-acylation reaction involving 2-lysophosphatidylcholine appears to be an important pathway for the synthesis of surfactant lipid in alveolar type II cells; the control of acyl specificity at the 2-position is determined by the relative concentrations of the coparticipating substrates, l-palmitoyl--glycero-3-phosphocholine and palmitoyl-CoA. 相似文献

Non-destructive assessment of developing hydraulic connections in the graft union of tomato 总被引:3,自引:1,他引:2
Hydraulic architecture prescribes water flow in plants and is,therefore, fundamental to many areas of plant physiology. Itis usually analysed destructively, or on excised material. Amethod is explored here based on displacement transducers forthe continuous, nondestructive assessment of functional hydraulicconnections within the intact plant. The graft union was chosenas a test system. The technique involves repeated applicationof water at some point in the system, while simultaneously observingpatterns of swelling (increase in water status) at other points.Such patterns will reflect the hydraulic resistance of the interveningpathways. It is demonstrated that the major hydraulic connections withinthe graft union of tomato become functional over a period ofabout 48 h from the fifth day after grafting. This is consistentwith histological observations on the appearance of wound-xylembridges at this time. This approach could be useful for non-destructive monitoringof changes in hydraulic connections in various other intactsystems, for example, during abscission, drought-induced embolism,or attack by vascular-wilt pathogens. Stomatal density, stomatal aperture length, area/leaf, and number of stomata/leaf were measured after the annual C3 agronomic grasses oats (Avena sativa) and wheat (Triticum aestivum), the C, woody legume honey mesquite (Prosopis glandulosa), and the perennial C4 grass little bluestem (Schizachyrium scoparium) were grown across a subambient carbon dioxide concentration ([CO2]) gradient from near 200 to 350 μmol/mol in a growth chamber. The purpose was to determine if the size and density of stomata vary in response to atmospheric [CO2] during growth, across a subambient [CO2] range representative of the doubling that has occurred since the last ice age. Changes in stomatal density and aperture length with increasing [CO2] were small when detected. Stomatal density decreased on adaxial flag leaf surfaces of wheat, and aperture length increased slightly with [CO2], Leaf area and number of stomata/flag leaf increased by similar proportions with [CO2] in two wheat cultivars. No consistent relationship between [CO2] and stomatal density or size was detected in mesquite, oats, or little bluestem. We conclude that individual plants of these species lack the plasticity to significantly alter stomatal density and aperture length in response to increasing atmospheric [CO2] in a single generation (annuals) or growing season (perennials). 相似文献
